Summary A human macrophagelike cell line which grows in serum-free medium was established from a histiocytic lymphoma cell line, U-937. U-937 cells failed to differentiate into macrophagelike cells in serum-free medium plus 12-O-tetradecanoyl phorbol 13-acetate (TPA). Fibronectin and albumin in serum were necessary for differentiation of U-937 ceds into macrophagelike cells in enriched RDF medium supplemented with insulin, transferrin, ethanolamine, selenite, egg yolk lipoprotein (eRDF-ITESL medium). The established cell line exhibited several characteristic properties of macrophage such as nitroblue tetrazolium reduction, phagocytic and α-naphthylbutyrate-esterase activities, and tumor necrosis factor and interleukin 1 production. At present the cells have been continuously maintained in eRDF-ITESL medium through over 150 passages.  相似文献

We propose and compare multiple approaches to automatically process data measured through surface‐enhanced Raman scattering (SERS), in the context of intracellular molecule probing. It relies on locally detecting the most relevant spectra to retrieve all data independently through indexing, thus avoiding any pre‐filtering which occurs with standard processing methods. We first assess our approach on simulated data of the spectrum of Rhodamine 6G, and then validate high‐performing methods on experimental measurements of this compound. The optimized method is then utilized to extract and classify the complex SERS response behavior of gold nanoparticles taken in live cells. (© 2013 WILEY‐VCH Verlag GmbH & Co. Though birch and alder are the common pioneer tree species which dominate in northeast Asia, little is known about the effects of the predicted increase in atmospheric CO2 concentrations ([CO2]) upon their photosynthesis in field conditions. To investigate this, we grew 2-year-old saplings of three Betulaceae species (Betula platyphylla var. japonica Hara, Betula maximowicziana Regel, and Alnus hirsuta Turcz) for 2 years in a free air CO2 enrichment system in northern Japan. Since the effect of high [CO2] is known to depend on soil conditions, we evaluated the responses in two soils which are widely distributed in northern Japan: infertile and immature volcanic ash (VA) soil, and fertile brown forest (BF) soil. For B. platyphylla, photosynthetic down-regulation occurred in both soils, but for B. maximowicziana, down-regulation occurred only in VA soil. The explanation is reduced nitrogen and Rubisco content in the leaf. For A. hirsuta, down-regulation occurred only in BF soil because of the accumulation of starch in foliage, which restricts CO2 diffusion inside the chloroplast. The higher photosynthetic rate of A. hirsuta in infertile VA soil could be due to the sink for photosynthates in the N2-fixing symbiont. These three species are all able to down-regulate at high [CO2]. However, it is possible that A. hirsuta would dominate in VA soil and B. maximowicziana in BF soil in the early stages of forest succession in a CO2-enhanced world.  相似文献

The ponerine ant Brachyponera chinensis was introduced to the USA, where it has become invasive. Although various ecological data have been collected for B. chinensis populations in the USA, most aspects concerning the biology and ecology of native populations in Japan, a presumed origin, remain unknown. Here we investigated the social structure and nestmate discrimination in native populations of B. chinensis and a closely related species, B. nakasujii. Both species showed functional polygyny over seasons. Only in B. nakasujii was there a seasonal change in the numbers of queens and workers per nest. In arena tests, workers of neither species showed aggressive behaviors to conspecific non‐nestmates from the same population, and the mean aggression score did not increase with the distance between nests. However, some differences in non‐aggressive responses were detected between nestmate and non‐nestmate pairs in both species. In an experiment to introduce a single worker into a nest, B. chinensis accepted non‐nestmates with a high probability just like nestmates, whereas in B. nakasujii non‐nestmates were less accepted than nestmates. These findings suggest that native populations of B. chinensis already possess some of the key characteristics shared by many invasive exotic ants in introduced ranges, such as stable polygyny, weak internest aggression and acceptance of non‐nestmates. These tendencies are remarkable in comparison to the closely related B. nakasujii.  相似文献

Vertical transmission is the primary route of the endosymbiont Wolbachia for its own spread among invertebrate hosts, but horizontal transmission between different hosts is believed to have occurred multiple times. However, it is not well known how Wolbachia commonly spread among closely related hosts. We focused on the closely related species of the minute pirate bugs belonging to the genus Orius, which are important biological control agents in agricultural crops because they are the most useful natural enemy of various tiny pests, such as thrips. Here, we examined five Orius species (Orius sauteri, Orius nagaii, Orius minutus, Orius strigicollis, and Orius tantillus) from eight geographic localities in Japan for Wolbachia infection. Two distinct strains, wOus1 and wOus2, were detected based on Wolbachia surface protein (wsp) gene sequencing. Furthermore, multilocus sequence typing revealed that each of the strains comprised two variants that differed in a single nucleotide. The overall distribution patterns of the two Wolbachia strains were found to differ among host species: prevalent double infection with wOus1 and wOus2 in O. strigicollis; fixation of single infection with wOus2 in O. nagaii; occurrence of single infection with wOus1 in O. sauteri; prevalence of single infection with wOus1 in O. minutus with an exception in a single population; and lack of Wolbachia infection in O. tantillus. Such differences in the distribution patterns of Wolbachia may reflect the evolutionary history of Wolbachia infection among Orius species and/or ecological and physiological differences among the Orius species that determine the invasiveness and maintenance of the two Wolbachia strains.  相似文献

We have devised a mapping method for rapid assembly and ordering of bacterial artificial chromosome (BAC) clones on a radiation hybrid (RH) panel, using sequence-tagged sites (STSs) and PCR. The protocol consists of two rounds of two-dimensional screening from a limited number of BACs to correspond each to an STS. In the first round, STSs are assembled in the RH bins and ordered according to PCR signals derived from 384-well microtiter plates (MTPs) in which BAC clones have been arrayed. In the second round, individual BAC clones are isolated from the MTPs to build a contig. We applied this method to a 35-Mb region spanning human chromosome 1p35-p36 and assembled 1366 BACs in 11 contigs, the longest being about 20 Mb. The working draft sequences of the human genome have been integrated into the contigs to validate the accuracy.  相似文献

The crystal structures of Li[Fe(trtda)]·3H2O and Na[Fe(eddda)]·5H2O (trtda = trimethylenediaminetetraacetate and eddda = ethylenediamine-N,N′-diacetate-N,N′-di-3-propionate) have been determined by single crystal X-ray diffraction techniques. The former crystal was monoclinic with the space group P21/n,a = 17.775(3),b = 10.261(1),c = 8.883(2)Å, β = 95.86(4)° and Z = 4. The latter was also monoclinic with the space group P21/n,a = 6.894(2),b = 20.710(6),c = 13.966(3)Å, β = 101.44(2)° and Z = 4. Both complex anions were found to adopt an octahedral six-coordinated structure with all of six ligand atoms of trdta4− or eddda4− coordinated to the Fe(III) ion, unlike the corresponding edta4− complex which is usually seven-coordinate with the seventh coordination site occupied by H2O. Of the three geometrical isomers possible for the eddda complex, the trans(O5) isomer was actually found in the latter crystal. Factors determining the structural types of metal–edta complexes are discussed in detail.  相似文献

In vitro incubation of three Kai proteins, KaiA, KaiB, and KaiC, with ATP induces a KaiC phosphorylation cycle that is a potential circadian clock pacemaker in cyanobacterium Synechococcus elongatus PCC 7942. The Kai proteins assemble into large heteromultimeric complexes (periodosome) to effect a robust oscillation of KaiC phosphorylation. Here, we report real-time measurements of the assembly/disassembly dynamics of the Kai periodosome by using small-angle X-ray scattering and determination of the low-resolution shapes of the KaiA:KaiC and KaiB:KaiC complexes. Most previously identified period-affecting mutations could be mapped to the association interfaces of our complex models. Our results suggest that the assembly/disassembly processes are crucial for phase entrainment in the early synchronizing stage but are passively driven by the phosphorylation status of KaiC in the late oscillatory stage. The Kai periodosome is assembled in such a way that KaiA and KaiB are recruited to a C-terminal region of KaiC in a phosphorylation-dependent manner.  相似文献

Here we report on an advanced survey system that combines multibeam echo sounding with underwater photography, which was used to collect accurate data on the distribution and abundance of submerged aquatic vegetation (SAV) in Lake Towada. The use of this system enabled us to visualize the cover, height, and biomass of the SAV over the lake bottom, as well as to distinguish between different components of the SAV such as vascular and algal plants. The spatial distributions of these major components of the SAV varied as a function of the depth gradient. The vascular component was mostly represented by Potamogeton species, which accounted for around one-third of the standing mass of SAV, whilst more than half of the SAV was algal (charophytes). This abundance of charophytes may well be responsible for the high water quality and transparency of Lake Towada.  相似文献
