In order to compute the muscle fiber conduction velocity (MFCV) and to clarify how action potentials are conducted, the normalized peak-averaging technique (NPAT) was newly employed together with computer softwares. Twelve pairs of surface electromyograms were selected from biceps brachii muscles during contraction at a level of 50% of the maximum voluntary isometric contraction in seven healthy volunteers. The techniques to compute MFCV from the time delay of the peaks (P-NPAT) and from the cross correlation (CC-NPAT) of averaged pulses were compared to the cross-correlation technique (CCT). The spread rate of averaged pulses was computed to estimate the spread of MFCVs in different motor units. Tri-phasic averaged pulses were obtained clearly by averaging more than 500 detected pulses. The P-NPAT and CC-NPAT highly correlated with the CCT in the computed MFCVs. The MFCVs obtained by P-NPAT were generally larger than those obtained by CCT, and the spread rates had in the definite values. These results suggest that the MFCV could be computed and the spread of MFCVs would be estimated from averaged pulses. The MFCV of a patient with myotonic dystrophy was also studied, and it was suggested that the NPAT would be clinically useful.  相似文献

Background  

The complete genomes of three animals have been sequenced by global research efforts: a nematode worm (Caenorhabditis elegans), an insect (Drosophila melanogaster), and a vertebrate (Homo sapiens). Remarkably, their relationships have yet to be clarified. The confusion concerns the enigmatic position of nematodes. Traditionally, nematodes have occupied a basal position, in part because they lack a true body cavity. However, the leading hypothesis now joins nematodes with arthropods in a molting clade, Ecdysozoa, based on data from several genes.  相似文献

The Sex-lethal (SXL) protein belongs to the family of RNA-binding proteins and is involved in the regulation of pre-mRNA splicing. SXL has undergone an obvious change of function during the evolution of the insect clade. The gene has acquired a pivotal role in the sex-determining pathway of Drosophila, although it does not act as a sex determiner in non-drosophilids. We collected SXL sequences of insect species ranging from the pea aphid (Acyrtho siphom pisum) to Drosophila melanogaster by searching published articles, sequencing cDNAs, and exploiting homology searches in public EST and whole-genome databases. The SXL protein has moderately conserved N- and C-terminal regions and a well-conserved central region including 2 RNA recognition motifs. Our phylogenetic analysis shows that a single orthologue of the Drosophila Sex-lethal (Sxl) gene is present in the genomes of the malaria mosquito Anopheles gambiae, the honeybee Apis mellifera, the silkworm Bombyx mori, and the red flour beetle Tribolium castaneum. The D. melanogaster, D. erecta, and D. pseudoobscura genomes, however, contain 2 paralogous genes, Sxl and CG3056, which are orthologous to the Anopheles, Apis, Bombyx, and Tribolium Sxl. Hence, a duplication in the fly clade generated Sxl and CG3056. Our hypothesis maintains that one of the genes, Sxl, adopted the new function of sex determiner in Drosophila, whereas the other, CG3056, continued to serve some or all of the yet-unknown ancestral functions.  相似文献

We defined gas exchange phenology as the seasonality of the gas exchange characteristics of a forest canopy, and investigated how the gas exchange phenology could be directly detected from an eddy covariance (EC) dataset and its influence on the canopy fluxes within an evergreen Japanese cypress forest. For the detection of gas exchange phenology, we derived three bulk parameters of the extended big-leaf model (Kosugi et al. 2005) inversely from EC flux data over a 7-year period: surface conductance (g c), maximum rate of carboxylation of the “big leaf” (V CMAX), and intercellular CO2 concentration of the “big leaf” (C I). The relationship between g c and the vapor pressure deficit declined in winter and spring. The relationship between the daily ecosystem respiration and air temperature was greater in the spring than in the other seasons. The temperature dependence curve of V CMAX decreased substantially in the winter and was different from that of an evergreen broadleaved forest. A decrease in C I was occasionally coupled with the decrease in canopy gross primary production during April and August, indicating that stomatal closure was responsible for a decline in canopy photosynthesis. Gas exchange phenology should be quantified when understanding the determining factors of the seasonality of canopy fluxes at evergreen coniferous forests.  相似文献

In order to investigate the neural connection of planarian, it is imperative to produce an antibody that specifically stains axons. To identify axon-specific genes, we constructed a cDNA library from a single eye by using a single cell PCR method, in which visual neurons are major components, and sequenced one thousand independent clones. We succeeded in the identification of a planarian homologue of synaptotagmin, Djsyt, whose specific expression in neurons was confirmed by in situ hybridization. The antibody against DjSYT specifically stained axons although its mRNA is distributed in the cell bodies. By using anti-DjSYT, we succeeded in the visualization of neural connections in planarians by whole mount staining. The anti-DjSYT antibody will become a powerful tool to analyze the molecular mechanisms underlying neural network formation in planarian.  相似文献

To estimate muscle fiber conduction velocity (MFCV), the normalized peak-averaging technique was developed along with complementary software and reported in a previous study. Electromyograms were recorded from the vicinity of end-plate zones (EMG data 1) and a site farther from the end-plates and distal tendons (EMG data 2) of biceps brachii muscles using surface array electrodes during 50% maximal voluntary isometric contractions in ten healthy volunteers. The MFCV values obtained with normalized peak-averaging technique were compared to those obtained with the cross-correlation technique and examined by computer simulation of the MFCV distribution. The MFCV values obtained with normalized peak-averaging technique highly correlated with those obtained with cross-correlation technique in both EMG data 1 and EMG data 2. It was also confirmed that MFCV values obtained from EMG data 1 were distributed much more widely than those obtained from EMG data 2. These results show the clinical usefulness of normalized peak-averaging technique for the detailed assessment of MFCV.  相似文献
