Physiological mechanisms of triggering and occurrence of a short-distancesystemic (sub-systemic) oxidative burst (OXB) caused by inductionof local OXB induced by the elicitor were investigated usingpotato tuber tissues. The sub-systemic OXB was detected as luminol-mediatedchemiluminescence (CL) on the non-treated side of tissue slicesfollowing a transient appearance of local OXB on the other sidedirectly treated with an elicitor. The sub-systemic OXB wasnot induced when the elicitor was applied in the presence ofa radical scavenger, H2O2-catabolising enzyme, or inhibitorsof activation of local OXB and NADPH oxidase (diphenyleneiodonium:DPI), Ca2+ che-lator and Ca2+ channel blockers). Treatment withH2O2 solution rapidly caused the sub-systemic OXB, which wasinhibited by the presence of the above inhibitors either duringthe treatment with H2O2 or detection of CL. These results suggestedthat the elicitor-stimulated sub-systemic OXB may be dependenton Of generating NADPH oxidase activated by an unknown systemicsignal stimulated by H2O2 generated via local activation ofthe NADPH oxidase.

The high incidence (40.6%) of elevated serum pancreatic phospholipase A2 (PLA2) was demonstrated in patients with various malignancies. Serum PLA2 was significantly increased in cancer patients compared with healthy sex- and age-matched blood donors (358.4 +/- 168.0 vs. 241.7 +/- 69.0 ng/dl; p less than 0.01). No correlation was observed between serum PLA2 and carcinoembryonic antigen (CEA) in these patients. Although patients with advanced and distantly metastatic cancer of the liver, gallbladder and pancreas showed higher PLA2 levels in serum than those with early cancer, patients with other cancers showed no correlation between serum PLA2 and clinical stage. A combined assay of PLA2 and CEA increased the sensitivity of detection of cancers to 60.8%. 相似文献

Calcitonin gene-related peptide (CGRP) in porcine ventricular muscles. positive inotropic effects in the isolated, electrically driven false tendon of the porcine heart. Specific CGRP-binding sites were present in solubilized membrane fractions; the dissociation constant (Kd) and the maximum binding (Bmax) were 50.4 pM and 180 fmol/mg protein, respectively. SDS-PAGE analysis of CGRP-binding sites revealed the molecular mass of 70 K and 120 K. Few CGRP-like immunoreactive nerves were present in the ventricular muscle layer. These results indicate that CGRP activates specific receptor sites on the ventricular muscles and causes positive inotropic responses. CGRP receptors in ventricles are likely to be activated by circulating CGRP. 相似文献

The carotenoids of five species of heliobacteria (Heliobacillus mobilis, Heliophilum fasciatum, Heliobacterium chlorum, Heliobacterium modesticaldum, and Heliobacterium gestii) were examined by spectroscopic methods, and the C30 carotene 4,4′-diaponeurosporene was found to be the dominant pigment; heliobacteria were previously thought to contain the
C40 carotenoid neurosporene. In addition, trace amounts of the C30 diapocarotenes diapolycopene, diapo-ζ-carotene, diapophytofluene, and diapophytoene were also found. Up to now, diapocarotenes
have been found in only three species of chemoorganotrophic bacteria, but not in phototropic organisms. Furthermore, the esterifying
alcohol of bacteriochlorophyll g from all known species of heliobacteria was determined to be farnesol (C15) instead of the usual phytol (C20). Heliobacteria may be unable to produce geranylgeranyol (C20).

The high-molecular-weight glycopeptides characteristic of early embryonic cells were isolated from F9 embryonal carcinoma cells grown in vitro and also from the cells grown in vivo as subcutaneous tumors. The two preparations had similar carbohydrate compositions. The major components were galactose and N-acetylglucosamine (molar ratio 1:0.86) in the glycan isolated from the cultured cells. In addition, small amounts of fucose, N-acetylgalactosamine and mannose were present. The glycan from the in vitro grown cells was found to have a molecular weight of more than 10,000 by gel filtration after mild alkaline treatment or hydrazinolysis. The structural characteristics of the core portion of the glycan were studied by using the radioactively labeled glycopeptide from the in vitro grown cells. Methylation analysis provided the following informations. 1) The glycan was highly branched at galactosyl residues. 2) Large numbers of galactosyl residues were also present at non-reducing termini. 3) Monosubstitution of galactose occurred at C-3. 4) Glucosamine residues were mainly monosubstituted. That the disaccharide GlcNAc-Gal was the major structural unit of the glycan was suggested by the isolation of the deacetylated disaccharide after alkaline thiophenol cleavage followed by acid hydrolysis. Furthermore, methylation analysis of the glycan from the in vivo grown tumors indicated that monosubstitution of glucosamine occurred at C-4 and that disubstitution of galactose occurred at least mainly at C-3 and C-6. We propose that the basic structural unit of the core portion is 4GlcNAc 1 leads to 3Gal, and that the galactosyl residue serves as a branching point at C-6. Thus, the structural unit of the core portion of the large glycan appears to be the same as that of lactosaminoglycans found in adult cells. 相似文献

Under anaerobic atmosphere where the gas phase was simply replacedby N2, photo-inhibition of PS I of isolated spinach chloroplastswas insignificant. However, when dithionite was included inthe irradiation mixture, severe photoinhibition of the NADP+and the MV photo-reduction occurred. Neither P700 determinedby continuous illumination-induced difference spectroscopy,Fe-S centers determined by EPR under cryogenic temperatures,nor vitamin K-l determined by HPLC analysis were significantlydecreased under these photoinhibition conditions. Although photobleachingof antenna chlorophylls occurred to more or less extent, NADP+photoreduction activities were markedly depressed even undersaturating actinic light. The maximal amplitude of the flashinduced absorbance changes of P700 in ms range decreased almostin parallel with the loss of NADP+ photoreduction activity.These results indicate that although the Fe-S centers of thephotoinhibited chloroplasts were reducible by continuous illumination,to almost the same extents as that of the control chloroplasts,the efficiency of reduction by each flash was much lower thanthat of the control chloroplasts. An efficient separation of corticosteroids in plasma of rats was obtained by reversed-phase high-performance liquid chromatography (HPLC). Plasma corticosteroid assays with HPLC separation were used to determine the circadian rhythm of 18-hydroxycorticosterone (18-OHB) and its possible relationship to aldosterone or corticosterone in conscious rats under standard conditions (regular diet; 12-hour light and 12-hour dark cycle). Significant circadian rhythms of plasma corticosterone, 18-OHB and aldosterone were observed with peak values at 20.00 h and nadir values at 08.00 h. The mean ratio of plasma 18-OHB to aldosterone during 24 h was 2.4. The circadian rhythm of 18-OHB was also correlated with that of plasma aldosterone or corticosterone. 相似文献

We have designed a novel aggregate of DNA block copolymer (DBC) that is sensitive to hypoxic X-irradiation. The DBC consists of tetrahydropyrane-protected 2-hydroxyethyl methacrylate as a hydrophobic unit and oligodeoxynucleotides as a hydrophilic unit, which are linked to a radiation-sensitive disulfide bond. The DBC self-assembled efficiently to form aggregates that encapsulated small molecules such as nile red and pyrene. Hypoxic X-irradiation could then induce reductive degradation of the DBC aggregates via an exchange reaction of the disulfide bond to release guest molecules. 相似文献
