BACKGROUND: Autosomal dominant polycystic kidney disease (ADPKD) is one of the most common inherited renal disorders with genetic heterogeneity. Mutations of two known genes are responsible for this disease: PKD1 at 16p13.3 and PKD2 at 4q21-23. A majority of cases (85%) are caused by mutations in PKD1. Because direct mutation screening remains complex, we describe here the application of an efficient approach to studies based on highly informative dinucleotide and tetranucleotide repeats flanking genes PKD1 and PKD2. METHODS: For this study a series of microsatellites closely linked to locus PKD1 (D16S291, D16S663, D16S665, D16S283, D16S407, D16S475) and to locus PKD2 (D4S1563, D4S2929, D4S414, D4S1534, D4S423) were selected. Short (81-242 bp) DNA fragments containing the tandem repeats were amplified by polymerase chain reaction (PCR). The number of repeat units of microsatelite markers was determined by fluorescent capillary electrophoresis. RESULTS: DNA microsatellite analysis was performed in 25 Polish ADPKD families and established the type of disease (21 families PKD1-type, 1 family PKD2-type). CONCLUSIONS: While a disease-causing mutation in the PKD1 and PKD2 genes cannot be identified, DNA microsatellite analysis provided an early diagnosis and may be considered in ADPKD families.  相似文献

Electrical activity (action and variation potentials) and ethylene emission were measured in thermally stimulated Vicia faba minor seedlings. It was determined that variation potential with or without super-imposed action potentials was generated and propagated basipetally in response to scorching of the upper leaf. In stimulated plants the level of ethylene production measured in lower, non stimulated leaf was significantly higher than that in the control plants and the difference correlated with the amplitude of the electrical response. Neither variation nor action potential was recorded when ethylene was injected to the chamber covering the experimental leaf. The level of ethylene emission showed clear circadian rhythm when measured at photoperiod 16:8 (LD) or at constant light (LL). It is concluded that the sequence of ion fluxes registered as an electrical response of a plant to the thermal stimulus is a signal evoking an enhancement of ethylene emission.  相似文献

Slowly activating vacuolar channels (SV), were examined in embryogenic and non-embryogenic cultures of winter wheat using a patch-clamp technique. Four different types of cultures were examined: embryogenic and non-embryogenic calli from embryos, embryogenic and non-embryogenic calli from inflorescences. In a cell-attached mode single SV channel events were recorded. Unitary conductance of single SV channels was between 37 pS and 48 pS and did not significantly depend on the kind of the culture, although it was a tendency that SV channels of embryogenic calli possessed lower unitary conductance than those of non-embryogenic. 2,4-D caused significant lowering of unitary conductance from 48±6 pS in the control culture of embryogenic embryos to 28±6 pS in vacuoles treated. The SV channel density was estimated as 0.34 μm−2.  相似文献

Chronic pancreatitis and pancreatic adenocarcinoma represent two pathologic phenomena with marked production of connective tissue stroma containing numerous small blood vessels. The aim of this study was to characterise quantitatively the vascular supply of pancreatic adenocarcinoma and fragments of the periductal tissue collected from patients with chronic pancreatitis. The study material included 18 cases of pancreatitis and 22 cases of pancreatic ductal adenocarcinoma. Microvessels were marked using monoclonal anti-CD34 antibodies. The number of blood vessels in the fibrous stroma was significantly higher in the chronic pancreatitis samples compared to the pancreatic carcinoma group (mean vessel count 298 and 194 vessel/mm2; median 251 and 187 vessel/mm2 respectively; p<0.01). Distributions of the vascular diameter in both studied groups were very similar. The obtained results suggest that the development of vascular network accompanying chronic pancreatitis is more effective in some parts of pancreas compared to angiogenic intensity in pancreatic adenocarcinoma.  相似文献

Conversely, oxytocin antagonists blocking uterine OT receptor can suppress uterus contraction. We found that the Sw-5 gene confers resistance to one of the Polish isolates of tomato spotted wilt virus (TSWV). A series of tomato breeding accessions was analysed along with standards of resistance and susceptibility to TSWV. The presence of the Sw-5 gene was determined using the available PCR marker. Subsequently plants from these accessions were grown in the presence of the TSWV isolate from Poland. Some of them developed severe symptoms of the TSWV disease. Expression of the virus proteins was also assayed in tissues of the investigated plants. We found general agreement between either lack or presence of the disease symptoms, virus proteins and resistance gene. Some observed discrepancies of these data are also discussed. Our results indicate that marker-assisted selection can be used for breeding of the TSWV-resistant tomato in Poland.  相似文献

Sarcolipin (SLN), a regulator of the sarco(endo)plasmic reticulum Ca(2+)-ATPase of fast-twitch skeletal muscle (SERCA1a), is also expressed in cardiac and slow-twitch skeletal muscles where phospholamban (PLN) and SERCA2a are expressed. Co-expression in HEK-293 cells of SLN tagged N-terminally with a FLAG epitope (NF-SLN), PLN, and SERCAs followed by measurement of the Ca(2+) dependence of Ca(2+) transport activity in isolated microsomal fractions showed that NF-SLN can reduce the apparent Ca(2+) affinity of both SERCA1a (DeltaK(Ca) = -0.22 +/- 0.01 pCa units) and SERCA2a (DeltaK(Ca) = -0.37 +/- 0.04 pCa units). When SERCA1a or SERCA2a were co-expressed with both NF-SLN and PLN, inhibition was synergistic, reducing DeltaK(Ca) by about -1.0 pCa units. Co-immunoprecipitation showed that NF-SLN increased the binding of PLN to SERCA, whereas PLN did not increase the binding of NF-SLN to SERCA. Elevated Ca(2+) dissociates both PLN and NF-SLN from their complexes with both SERCA1a and SERCA2a, but NF-SLN induced resistance to Ca(2+) dissociation of the PLN.SERCA complex. Co-immunoprecipitation of PLN and NF-SLN without SERCA showed that NF-SLN binds directly to PLN and that NF-SLN inhibits the formation of PLN pentamers. Thus the ability of NF-SLN to elevate the content of PLN monomers can account, at least in part, for the superinhibitory effects of NF-SLN in the presence of PLN.  相似文献

At low light intensities (less than 50 µmol m–2s–1) illumination evokes transient depolarization of membranepotential in mesophyll cells of the leaf-trap of Dionaea muscipulaEllis. Darkening causes hyperpolarization approximately symmetricto the response to illumination. The amplitude as well as therate of potential changes depend on light intensity. After exceedinga definite threshold (usually between 50 and 80 µmol m–2s–1)the depolarization plays the role of a generator potential andan all-or-none action potential (AP) is released. Switchinglight off in a depolarization phase of an AP does not changeits shape and the amplitude. When the light intensity is increasedto 80–150 µmol m–2 s–1 a single lightstimulus triggers two successive APs. The time interval betweenthe two APs decreases with increasing stimulus strength andreaches the minimum between 300 and 400 µmol m–2s–1. At higher light intensities the interval increasesagain, and finally only a single AP is triggered. It was shownthat the effect was evoked by light but not by temperature changeaccompanying illumination. An inhibitor of the photosyn-theticelectron transport chain, DCMU, blocked the generator potentialsmediating between light absorption and APs. An attempt was made to estimate the role of small mammals and birds in the cycling of nutrients in the littoral zone of lakes. We estimated the approximate amounts of food, organic matter and elements ingested and egested during the breeding season by waterfowl. Small mammal densities were estimated, and their movements studied between different patches of a mosaic landscape, including the lake's shore region. Studies on passerine birds were aimed at determining the territories of some species in alder forest. Special attention was paid to the range of penetration of the reed belts by forest species.  相似文献
