It has been found that plastoquinone-9 and α-tocopherol quinone form charge-transfer complexes with their corresponding reduced forms in anhydrous dipalmitoyl phosphatidylcholine and octadecane throughout the whole temperature range examined, i.e. from -30 to 60°C. The charge-transfer spectra of both prenylquinones in lecithin showed the presence of only one band with λmax in the range 520–600 nm, depending on temperature. There was a gradual long-wavelength shift of λmax of the band observed with decrease in temperature. The intensity of the band was quite constant. Only at the lowest temperatures was a sudden intensity decrease, caused by precipitation of the hydroquinone form, observed. On the other hand, the charge-transfer spectra of both prenylquinones in octadecane showed the presence of two bands with strong temperature dependence. On freezing, the intensity of these two bands increased with a simultaneous long-wavelength shift of both maxima. The results are discussed in terms of different molecular structures of the complex in both systems. 相似文献

The aim of the current study was to characterize Polish Black-and-White cattle by morphological study of the Y chromosome. A total of 14 Y-linked microsatellites from UMN and INRA group were genotyped and assessed for polymorphism in a total 22 bulls. Cytogenetic studies in Polish Black-and-White bulls showed the existence of two morphological forms of Y chromosome. Among the 22 karyotypic analyzed bulls, 12 had submetacentric and 10 metacentric Y chromosome. The centromeric index of Y chromosome measured as percentage length of the p arm to total length ratio in the first case was 28 ± 3.97% and in the second 47 ± 7.28%, whereas the relative size of these chromosomes remained within the same range. Morphology and G- and C-banding patterns of both forms of Y chromosome were typical for other cattle breeds originating from Bos taurus. Out of a total of 14 microsatellite loci examined, 13 showed specific alleles for two forms of Y chromosome. In a pool of 62 alleles, 43 (69.3%) were common in the two groups of cattle, 19 (30.7%) can be considered as specific for the group; among them 8 were typical for metacentric group of Y chromosome and 11 for submetacentric. 相似文献

The effect of three different anion channel inhibitors, namely(5-nitro-2-3-phenylpropyl-amino)benzoic acid (NPPB), Zn2+ andanthracene-9-carboxylic acid (A-9-C) on the action potentialin the liverwort Conocephalum conicum were tested. All threecaused an increase of the excitability threshold and a decreaseof action potential amplitudes. This confirms the involvementof anion channels in the action potentials in Conocephalum.In plants treated with 1 or 2 mM A-9-C but not with NPPB (50or 100 µM) and Zn2+ (100 or 500 µM), a light-inducedtransient depolarization occurred. In contrast to action potentials,the amplitude of this voltage transient depended on the lightintensity and on the duration of preceding dark period. Alsoin contrast to action potentials, which are blocked by TEA,when applied together with A-9-C, TEA even increased the amplitudesof the light-induced voltage transients to up to 170 mV. Thedepolarization was obviously limited by the voltage-dependentopening of K+ channels in the absence of TEA. The amplitudeof the light-induced voltage transients (in the presence ofTEA) increased in elevated CaCl2 concentrations pointing toa Ca2+ permeability giving rise to the depolarization. However,none of the Ca2+ channel blockers tested, La3+, Gd3+, nifedipine,verapamil or diltiazem, had an effect. The light-induced voltagetransients in A-9-C treated plants are quite different fromlight- and electrically triggered action potentials but sharesome similarities with light-induced generator potentials. Thimerosal (THI, ethyl-mercury thiosalicylate) is added to vaccines as a preservative; as a consequence, infants may have been exposed to bolus doses of Hg that collectively added up to nominally 200 µg Hg during the first 6 months of life. While several studies report an association between THI-containing vaccines and neurological disorders, other studies do not support the causal relation between THI and autism. With the purpose to understand the molecular mechanisms of the toxic effect of THI it was assayed on human red cells and in bilayers built-up of dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylethanolamine (DMPE), classes of phospholipids found in the outer and inner monolayers of the human erythrocyte membrane, respectively. The capacity of THI to interact with DMPC and DMPE was determined by X-ray diffraction and differential scanning calorimetry, whereas intact human erythrocytes were observed by optical, defocusing and scanning electron microscopy. The experimental findings of this study demonstrated that THI interacted in a concentration-dependent manner with DMPC and DMPE bilayers, and in vitro interacted with erythrocytes inducing morphological changes. In this paper the classification ofthe gravitational effects in a passive transmembranetransport is presented. Among these effects there arethe flux and force gravitational effects (fluxgraviosmotic effect, osmotic pressure graviosmoticeffect, flux gravidiffusive effect, osmotic pressuregravidiffusive effect, voltage gravielectric effectand current gravielectric effect). The volume fluxgraviosmotic and solute flux gravidiffusive effectsmodel equations for a single-membrane system areelaborated. These models for binary and ternarynon-electrolyte solutions have been verified using anexperimental data volume and solute fluxes forosmotic-diffusion cell with horizontally mountedmembrane. In the experimental set-up, water was placedon one side of the membrane. The opposite side of themembrane was exposed to binary or ternary solutions ofdensities greater than that of water (aqueous glucoseor glucose-0.2 mole/l aqueous ethanol) and binary andternary solutions of densities larger than that ofwater (aqueous ethanol or ethanol-0.05 mole/l aqueousglucose). In the present study, the solubility and enzymatic de-epoxidation of diadinoxanthin (Ddx) was investigated in three different artificial membrane systems: (1) Unilamellar liposomes composed of different concentrations of the bilayer forming lipid phosphatidylcholine (PC) and the inverted hexagonal phase (HII phase) forming lipid monogalactosyldiacylglycerol (MGDG), (2) liposomes composed of PC and the HII phase forming lipid phosphatidylethanolamine (PE), and (3) an artificial membrane system composed of digalactosyldiacylglycerol (DGDG) and MGDG, which resembles the lipid composition of the natural thylakoid membrane. Our results show that Ddx de-epoxidation strongly depends on the concentration of the inverted hexagonal phase forming lipids MGDG or PE in the liposomes composed of PC or DGDG, thus indicating that the presence of inverted hexagonal structures is essential for Ddx de-epoxidation. The difference observed for the solubilization of Ddx in HII phase forming lipids compared with bilayer forming lipids indicates that Ddx is not equally distributed in the liposomes composed of different concentrations of bilayer versus non-bilayer lipids. In artificial membranes with a high percentage of bilayer lipids, a large part of Ddx is located in the membrane bilayer. In membranes composed of equal proportions of bilayer and HII phase forming lipids, the majority of the Ddx molecules is located in the inverted hexagonal structures. The significance of the pigment distribution and the three-dimensional structure of the HII phase for the de-epoxidation reaction is discussed, and a possible scenario for the lipid dependence of Ddx (and violaxanthin) de-epoxidation in the native thylakoid membrane is proposed. 相似文献

Donepezil is used to treat symptomatically the Alzheimer's disease (AD). This drug is a specific inhibitor of the enzyme acetylcholinesterase (AChE), whose main physiological function is to hydrolyze the neurotransmitter acetylcholine. The main objective of this work was to study the effect of donepezil on human erythrocytes as AChE is present in its membrane. For this purpose, human erythrocytes and molecular model of its membrane built-up of dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylethanolamine (DMPE) were used. The latter correspond to classes of phospholipids present in the outer and inner monolayers of the human erythrocyte membrane, respectively. Our experimental evidences obtained from X-ray diffraction and differential scanning calorimetry (DSC) analysis indicated that donepezil was capable of interacting with both phospholipids. Fluorescence spectroscopy results showed a moderate increase in the fluidity of the hydrophobic tails of DMPC and isolated unsealed human erythrocyte membranes (IUM). On the other hand, results by scanning electron microscopy (SEM) and optical defocusing microscopy (DM) showed that the drug changed the normal biconcave shape of the erythrocytes inducing the formation of stomatocytes (cup-shaped cells).
