Pannexins (Panxs) are a multifaceted family of ion and metabolite channels that play key roles in a number of physiological and pathophysiological settings. These single membrane large-pore channels exhibit a variety of tissue, cell type, and subcellular distributions. The lifecycles of Panxs are complex, yet must be understood to accurately target these proteins for future therapeutic use. Here we review the basics of Panx function and localization, and then analyze the recent advances in knowledge regarding Panx trafficking. We examine several intrinsic features of Panxs including specific post-translational modifications, the divergent C-termini, and oligomerization, all of which contribute to Panx anterograde transport pathways. Further, we examine the potential influence of extrinsic factors, such as protein-protein interactions, on Panx trafficking. Finally, we highlight what is currently known with respect to Panx internalization and retrograde transport, and present new data illustrating Panx1 internalization following an activating stimulus.  相似文献

Enrichment culture experiments employing soil and water samples obtained from petroleum-contaminated environments succeeded in the isolation of a pure culture possessing the ability to utilize quinoline as a sole nitrogen source but did not utilize quinoline as a carbon source. This culture was identified as Pseudomonas ayucida based on a partial 16S rRNA gene sequence, and the strain was given the designation IGTN9m. Examination of metabolites using thin-layer chromatography and gas chromatography-mass spectrometry suggests that P. ayucida IGTN9m converts quinoline to 2-quinolinone and subsequently to 8-hydroxycoumarin. Resting cells of P. ayucida IGTN9m were shown to be capable of selectively removing about 68% of quinoline from shale oil in a 16-h treatment time. These results suggest that P. ayucida IGTN9m may be useful in petroleum biorefining for the selective removal of organically bound nitrogen from petroleum.  相似文献

How environmental changes are affecting bird population dynamics is one of the most challenging conservation issues. Dietary studies of top avian predators could offer scope to monitor anthropogenic drivers of ecosystem changes. We investigated the diet of breeding Eleonora''s falcon in an area of Northeastern Algeria in the years 2010–2012. Feathers and insect remains originating from prey plucking behavior were analyzed, providing insights into the seasonally changing diet of this raptor, as well as the trans‐Mediterranean avian migration. A total of 77 species of birds (16 Sylviidae, 11 Turdidae, and 4 Emberizidae), 3 species of insects, and 1 lizard were identified among prey remains, reflecting a diverse diet. Diet composition and prey abundance varied seasonally, faithfully correlating with the passage of migrant birds as recorded from bird ring recoveries. Our findings suggest that dietary studies of predators might be deployed to investigate changes in bird migration. We discuss our results in the context of trans‐Mediterranean migration, with early‐season prey mainly comprising trans‐Saharan migrants (Apus apus and Merops apiaster) and late‐season prey being dominated by Mediterranean winter migrants (Erithacus rubecula, Turdus philomelos, Sylvia atricapilla, and Sturnus vulgaris). Notably, we observed a significant reduction in species richness of passerine remains in 2012, potentially highlighting a decline in the diversity of avian migrants.  相似文献

The Massim, a cultural region that includes the southeastern tip of mainland Papua New Guinea (PNG) and nearby PNG offshore islands, is renowned for a trading network called Kula, in which different valuable items circulate in different directions among some of the islands. Although the Massim has been a focus of anthropological investigation since the pioneering work of Malinowski in 1922, the genetic background of its inhabitants remains relatively unexplored. To characterize the Massim genomically, we generated genome-wide SNP data from 192 individuals from 15 groups spanning the entire region. Analyzing these together with comparative data, we found that all Massim individuals have variable Papuan-related (indigenous) and Austronesian-related (arriving ∼3,000 years ago) ancestries. Individuals from Rossel Island in southern Massim, speaking an isolate Papuan language, have the highest amount of a distinct Papuan ancestry. We also investigated the recent contact via sharing of identical by descent (IBD) genomic segments and found that Austronesian-related IBD tracts are widely distributed geographically, but Papuan-related tracts are shared exclusively between the PNG mainland and Massim, and between the Bismarck and Solomon Archipelagoes. Moreover, the Kula-practicing groups of the Massim show higher IBD sharing among themselves than do groups that do not participate in Kula. This higher sharing predates the formation of Kula, suggesting that extensive contact between these groups since the Austronesian settlement may have facilitated the formation of Kula. Our study provides the first comprehensive genome-wide assessment of Massim inhabitants and new insights into the fascinating Kula system.  相似文献
