Breast regression protein (BRP39) is a glycoprotein, which is expressed during mammary gland involution in mouse. The physiological function of BRP39 is not known. High levels of expression of BRP39 have also been associated with breast cancer development. In the present investigation a cDNA encoding rBRP39 (recombinant BRP39) was cloned by PCR techniques. It consists of 1,143 nucleotides and encodes an open reading frame of 381 amino acid residues including a signal sequence of 21 amino acids. Recombinant BRP39 was produced in E. coli in a soluble form at low temperature (15 °C). Expression and purification of rBRP39 was confirmed by western blot analysis. Purified rBRP39 showed high chitin-binding activity but no chitinase activity. The lack of chitinase activity may be attributed to the mutation of critical active site residue Glu120 to Leu120 and Asp118 to Ala118 in BRP39. However, a mutant in which the residue was reverted back to Glu, by site directed mutagenesis, displayed no chitinase activity. Purified recombinant BRP39 was crystallized and the crystals diffracted X-rays to 2.8 Å resolution. The crystals belonged to the space group C2 with unit cell parameters a = 130.4 Å, b = 81.3 Å, c = 229.2 Å, β = 105.9°. The structure refinement is in progress. 相似文献

Calmodulin-like activity has been reported for the first time in mycobacterial species, namely Mycobacterium tuberculosis BCG and M. smegmatis ATCC 14468. The activity was mainly located in the soluble fraction of the mycobacterial cells, Radioimmunoassay revealed maximum levels of calmodulin in young growing cells (early logarithmic phase of growth). Calmodulin-dependent phosphodiesterase activation assay revealed low activity (22%) of partially purified calmodulin either due to insufficient amount of calmodulin to activate phosphodiesterase or due to the presence of some factors interfering with the assay. Calmodulin antagonists, viz. trifluoperazine and phenothiazine, significantly inhibited the 32Pi incorporation into mycobacterial phospholipids. Similar inhibition was observed when EGTA (which removes calcium) was added to the medium. Significant inhibition of 32Pi incorporation in the presence of calmodulin antagonists suggested the involvement of calmodulin in mycobacterial phospholipid metabolism. 相似文献

A trial was undertaken with juvenile Nile tilapia fed graded levels of dietary protein (0–35% DP) over 7 weeks (28°C). Measurements of nitrogen and energy utilization were made using data on ADC, comparative carcass analyses, nitrogen excretion and oxygen uptake. Data indicate that the daily protein intake for maximum N gain was c. 12 g/kg/d and the maintenance protein requirements were about of 2g/kg/d. The optimal DP/DE ratio was found to be 18mg/kJ. Differences were observed in the data on endogenous nitrogen and energy utilization depending on the dietary treatments as well as on the criteria used. 相似文献
