We had previously shown that selective thromboxane synthetase inhibition with furegrelate increases urinary excretion of 6-ketoPGF1 alpha, the hydrolysis product of prostacyclin after stimulation of renal prostaglandin synthesis with furosemide. The present study assessed the functional significance of this "redirection" of prostaglandin formation using a more physiologic stimulus, angiotensin II. Sprague-Dawley rats (n = 27) were fitted with a transabdominal bladder cannula. Five days later they were given angiotensin II (10 mg.kg-1.min-1) by intravenous infusion. After 30 min, an infusion of furegrelate, 2 mg/kg, then 2 mg.kg-1.h-1, (n = 9); indomethacin, 2 mg/kg, then 2 mg.kg-1.h-1 (n = 9); or vehicle, 250 microL, then 0.018 mL/min (n = 9) was begun for 60 min. Clearance of [14C]para-aminohippuric acid was taken as a measure of renal plasma flow. Angiotensin II raised the mean arterial pressure in all groups. Administration of furegrelate or indomethacin did not change mean arterial pressure or heart rate. Angiotensin II reduced [14C]p-aminohippuric acid clearance by about 32% (1.42 +/- 0.18 to 0.97 +/- 0.07 mL.min-1.100 g-1, p less than 0.05). Furegrelate attenuated this renal vasoconstriction (0.97 +/- 0.07 to 1.38 +/- 0.17 mL.min-1.100 g-1, p less than 0.05), while indomethacin increased it by a further 32% (1.78 +/- 0.12 to 1.20 +/- 0.12 mL.min-1.100 g-1, p less than 0.05). Vehicle alone had no effect. Furegrelate reduced serum thromboxane B2 by 90% (6.52 +/- 0.030 to 0.7 +/- 0.21 ng/100 microL, p less than 0.05), while indomethacin reduced it by 73% (5.9 +/- 0.99 to 1.4 +/- 0.20 ng/100 microL, p less than 0.05). We conclude that furegrelate attenuates the renal vasoconstriction of angiotensin II, presumably by enhancing the formation of vasodilator prostaglandins. 相似文献

We established a long-term field study in an old growth coniferous forest at the H.J. Andrews Experimental Forest, OR, USA,
to address how detrital quality and quantity control soil organic matter accumulation and stabilization. The Detritus Input
and Removal Treatments (DIRT) plots consist of treatments that double leaf litter, double woody debris inputs, exclude litter
inputs, or remove root inputs via trenching. We measured changes in soil solution chemistry with depth, and conducted long-term
incubations of bulk soils from different treatments in order to elucidate effects of detrital inputs on the relative amounts
and lability of different soil C pools. In the field, the addition of woody debris increased dissolved organic carbon (DOC)
concentrations in O-horizon leachate and at 30 cm, but not at 100 cm, compared to control plots, suggesting increased rates
of DOC retention with added woody debris. DOC concentrations decreased through the soil profile in all plots to a greater
degree than did dissolved organic nitrogen (DON), most likely due to preferential sorption of high C:N hydrophobic dissolved
organic matter (DOM) in upper horizons; percent hydrophobic DOM decreased significantly with depth, and hydrophilic DOM had
a much lower and less variable C:N ratio. Although laboratory extracts of different litter types showed differences in DOM
chemistry, percent hydrophobic DOM did not differ among soil solutions from different detrital treatments in the field, suggesting
that microbial processing of DOM leachate in the field consumed easily degradable components, thus equalizing leachate chemistry
among treatments. Total dissolved N leaching from plots with intact roots was very low (0.17 g m−2 year−1), slightly less than measured deposition to this very unpolluted forest (~s 0.2 g m−2 year−1). Total dissolved N losses showed significant increases in the two treatments without roots whereas concentrations of DOC
decreased. In these plots, N losses were less than half of estimated plant uptake, suggesting that other mechanisms, such
as increased microbial immobilization of N, accounted for retention of N in deep soils. In long-term laboratory incubations,
soils from plots that had both above- and below-ground litter inputs excluded for 5 years showed a trend towards lower DOC
loss rates, but not lower respiration rates. Soils from plots with added wood had similar respiration and DOC loss rates as
control soils, suggesting that the additional DOC sorption observed in the field in these soils was stabilized in the soil

Although many climate predictions suggest that the frequency and intensity of large storm events might increase in the coming decades, few studies document the full impact of such events along their path. Here, we synthesize information on the impact of Hurricane Irene (formed August 21 2011) and Tropical Storm Lee (formed August 30, 2011) on erosion and sediment transport, lake metabolism, riparian hydrology and biogeochemistry, and stream water quality, from North Carolina to Maine. In almost all cases, these storms generated unprecedented changes in water quality (concentrations, loads), from tenfold increases in DOC and 100-fold increases in POC in Maryland, to 100-fold increases in TSS concentrations in Pennsylvania. Overbank flooding and up to 200-year streamflow events were recorded in New York and Vermont. In many cases, particulate loads (e.g. POC, PP, TSS) occurring during Irene and Lee represented more than 30% of the annual load. The dominance of particulate exports over solutes during Irene and Lee is consistent with the mobilization of normally immobile sediment pools, and massive erosion as reported at many locations across the Northeastern US. Several studies reported long lasting (> 1 year) effects of Irene and Lee on cyanobacterial blooms, erosion, or stream suspended sediment concentrations. However, this review also highlighted the lack of a consistent strategy in terms of methods, and measured water quality parameters. Kifunensine, produced by the actinomycete Kitasatosporia kifunense 9482, is an alkaloid that corresponds to a cyclic oxamide derivative of 1-amino mannojirimycin. This compound was reported to be a weak inhibitor of jack bean alpha-mannosidase (IC50 of 1.2 x 10(-4) M) (Kayakiri, H., Takese, S., Shibata, T., Okamoto, M., Terano, H., Hashimoto, M., Tada, T., and Koda, S. (1989) J. Org. Chem. 54, 4015-4016). We also found that kifunensine was a poor inhibitor of jack bean and mung bean aryl-alpha-mannosidases, but it was a very potent inhibitor of the plant glycoprotein processing enzyme, mannosidase I (IC50 of 2-5 x 10(-8) M), when [3H]mannose-labeled Man9GlcNAc was used as substrate. However, kifunensine was inactive toward the plant mannosidase II. Studies with rat liver microsomes also indicated that kifunensine inhibited the Golgi mannosidase I, but probably does not inhibit the endoplasmic reticulum mannosidase. Kifunensine was tested in cell culture by examining its ability to inhibit processing of the influenza viral glycoproteins in Madin-Darby canine kidney cells. Thus, when kifunensine was placed in the incubation medium at concentrations of 1 microgram/ml or higher, it caused a complete shift in the structure of the N-linked oligosaccharides from complex chains to Man9(GlcNAc)2 structures, in keeping with its inhibition of mannosidase I. On the other hand, even at 50 micrograms/ml, deoxymannojirimyucin did not prevent the formation of all complex chains. Thus kifunensine appears to be one of the most effective glycoprotein processing inhibitors observed thus far, and knowledge of its structure may lead to the development of potent inhibitors for other processing enzymes. 相似文献

Microfilariae, infective larvae, and adult worms of Brugia malayi were incubated with a panel of seven lectins in order to study the expression of surface carbohydrates. Infective larvae and adult worms did not bind any of the lectins utilized. Microfilariae, on the other hand, bound wheat germ agglutinin. The binding of this lectin was saturable and specific, and attributed to the presence of N-acetyl-D-glucosamine. In addition, microfilariae derived in vitro bound concanavalin A, indicating the presence of glucose and/or mannose on this stage of the parasite. The fact that similar concanavalin A binding was not seen on microfilariae recovered directly from the infected host implies that there is masking or loss of parasite surface antigens as microfilariae mature in vivo. 相似文献

Cardiovascular disease is the largest cause of morbidity and mortality among patients with chronic kidney disease (CKD) and end-stage kidney disease, with nearly half of all deaths attributed to cardiovascular disease. Hydroxychloroquine (HCQ), an anti-inflammatory drug, has been shown to have multiple pleiotropic actions relevant to atherosclerosis. We conducted a proof-of-efficacy study to evaluate the effects of hydroxychloroquine in an animal model of atherosclerosis in ApoE knockout mice with and without chronic kidney disease. Forty male, 6-week-old mice were divided into four groups in a 2 x 2 design: sham placebo group; sham treatment group; CKD placebo group; and CKD treatment group. CKD was induced by a two-step surgical procedure. All mice received a high-fat diet through the study duration and were sacrificed after 16 weeks of therapy. Mice were monitored with ante-mortem ultrasonic echography (AUE) for atherosclerosis and vascular stiffness and with post-mortem histology studies for atherosclerosis. Therapy with HCQ significantly reduced the severity of atherosclerosis in CKD mice and sham treated mice. HCQ reduced the area of aortic atherosclerosis on en face examination by approximately 60% in HCQ treated groups compared to the non-treated groups. Additionally, therapy with HCQ resulted in significant reduction in vascular endothelial dysfunction with improvement in vascular elasticity and flow patterns and better-preserved vascular wall thickness across multiple vascular beds. More importantly, we found that presence of CKD had no mitigating effect on HCQ’s anti-atherosclerotic and vasculoprotective effects. These beneficial effects were not due to any significant effect of HCQ on inflammation, renal function, or lipid profile at the end of 16 weeks of therapy. This study, which demonstrates structural and functional protection against atherosclerosis by HCQ, provides a rationale to evaluate its use in CKD patients. Further studies are needed to define the exact mechanisms through which HCQ confers these benefits. 相似文献

We report the first draft genome sequences of the strains of plague-causing bacteria, Yersinia pestis, from India. These include two strains from the Surat epidemic (1994), one strain from the Shimla outbreak (2002) and one strain from the plague surveillance activity in the Deccan plateau region (1998). Genome size for all four strains is ~4.49 million bp with 139–147 contigs. Average sequencing depth for all four genomes was 21x. 相似文献
