The potential of an aqueous two-phase system composed of a polycation, poly(ethyleneimine) (PEI), and an uncharged polymer, (hydroxyethyl) cellulose (HEC), for extractive lactic acid fermentation was tested. Batch fermentation with 20 g/L glucose in two-phase medium using Lactococcus lactis without external pH control resulted in 3-4 times higher amount of lactate and biomass produced as compared to that in a conventional one-phase medium. Lactic acid was preferentially partitioned to the PEI-rich bottom phase. However, the cells which favored the HEC-rich top phase in a fresh two-phase medium were partitioned to a significant extent to the bottom phase after fermentation. Addition of phosphate buffer or pH adjustment to 6.5 after fermentation caused fewer cells to move to the bottom phase. With external pH control, fermentation in normal and two-phase medium showed no marked differences in glucose consumption and lactic acid yield, except that about 1.3 times higher cell density was obtained in the two-phase broth, especially at initial glucose concentrations of 50-100 g/L. Use of higher concentration of phosphate during batch fermentation in the two-phase medium with 50 g/L sugar provided a 15% higher yield of lactic acid, but the growth rate of cells was nearly half of the normal, thus affecting the productivity. Continuous fermentation with twice the normal phosphate concentration resulted in higher cell density, product yield, and productivity in two-phase medium than in monophasic medium. The study addressed to understand whether or not lipoproteins at low concentrations could modulate Receptor-C dependent platelet signalling revealed that LDL, like exogneous cholesterol, had the capacity to initiate PLD-dependent platelet signalling in a dose dependent fashion and this effect was inhibited in presence of HDL; cAMP; DTT; Zn++ and butanol whereas cGMP had no effect upon this PLD-dependent signalling. Further Receptor C from platelet in the purified-form displayed LDL-or cholesterol-dependent autophosphorylation at the tyrosine residues and this Receptor-C tyrosine kinase (Receptor-Ck) activity contributed to the observed LDL-or cholesterol-dependent PLD activity in human platelets. Based upon these results coupled with earlier results, an attempt was made to define the lipoprotein-dependent platelet signalling pathway.  相似文献

The current best serum marker for pancreatic cancer, CA 19-9, detects a carbohydrate antigen on multiple protein carriers. Better knowledge of the protein carriers of the CA 19-9 antigen in various disease states may lead to improved diagnostic tests. To identify proteins that carry the CA 19-9 antigen, we immunoprecipitated the CA 19-9 antigen from pooled sera and identified the associated proteins using MS. Among the high-confidence identifications, we confirmed the presence of the CA 19-9 antigen on Apolipoprotein B-100 by antibody arrays and Western blot and on kininogen, ARVCF, and Apolipoprotein E by antibody arrays. We characterized the frequency and levels of the CA 19-9 antigen on the four proteins across various patient groups (pancreatic cancer, pancreatitis, and healthy controls) using antibody arrays. Nearly, 10-25% of the subjects showed elevations of the antigen on each protein, but the elevations were not associated with disease state or total CA 19-9 levels. These results contribute to our knowledge of the carrier proteins of an important functional glycan and the rate at which the glycan is displayed. This work also demonstrates a strategy for using the complementary methods of MS and antibody microarrays to identify protein carriers of glycans and assess the diagnostic value of measuring glycans on individual proteins.  相似文献

In this study, the relationship between the concentration of extra-heavy crude petroleum in a clayey material and the toxicity, field capacity, temperature, and growth of a tropical forage grass (Brachiara humidicola) was determined empirically. For this type of petroleum the acute toxicity (Microtox®) was slight (CE50 = 63200 - 76400 mg/kg) even at high hydrocarbon concentrations (29279 mg/kg). Nonetheless, serious impacts were encountered in terms of an increase in soil temperature (+ 1.3 °C), reduction in field capacity (-10.7%) and reduction in aerial biomass (-97%). The relationship between hydrocarbon concentration and biomass resulted in a typical dose-response curve (r = 0.99), where a concentration of 2626 mg/kg of hydrocarbons corresponds to a maintenance of 90% biomass. Furthermore, during the duration of this study (one year) the biodegradation was proportional to the pasture biomass production (r = 0.997) indicating a synergistic relationship between the petroleum biodegrading microorganisms in the rhizosphere and the pasture.  相似文献

In the present study the whole sample of 772 desert gerbils was collected from the semi-arid areas in and around Jaipur (Rajasthan, India). The male exhibits active spermatogenesis and well maintained accessory reproductive organs at any time of the year. Reproductive organs of the male gerbil are briefly described. The vaginal and urinary openings are independent in the gerbil. There are four pairs of mammae. Follicular development takes place cyclically. Maximum atresia of follicles is noticed during metoestrus. A well developed Graafian follicle measures 590 μ across its widest diameter. The average diameter of the ovum is 65 μ. At the time of ovulation formation of stigma occurs which gives way to the ovum. The corpus luteum is formed from the remaining granulosa cells of the ovulated follicle. Fully formed corpus luteum of the cycle (during dioestrus) measures 740 μ. Maximum development of the corpus luteum is seen during late pregnancy (22–25 days) when it measures 1250 μ. During pregnancy large Graafian follicles are absent. After parturition the ovary shows corpora lutea of lactation. Histological changes in the uterus and vagina during the oestrous cycle of the gerbil are described. There is less proestrus distension of the uterus as compared with the rat. During oestrus the uterine endometrium hypertrophies and there are some degenerative changes in the epithelial lining. The changes in the vaginal epithelium are clear cut and are correlated with the ovarian activity. During proestrus proliferation of the epithelium occurs and at ‘heat period’ or oestrus the epithelium shows formation of the stratum corneum and the shedding of cornified cells. In the dioestrus the vaginal epithelium is heavily infiltrated by leucocytes. There are three peaks in the incidence of pregnancy rate, i.e., February to April, July, and September to November. Pregnancy rate is lowest during summer (May and June) and winter (December and January). Average monthly litter size shows almost similar changes. The average length of gestation period is 30 days. There is post-partum heat in this animal. Formation of a vaginal plug is noticed following copulation. Litter size varies from 2–7. The average body weight of the new born young is 3.5 and 3.4 gm respectively for the male and the female. Weaning of the young is observed after about 20 days. Descent of the testes into the scrotal sacs occurs around day 28. Vagina opens after about 75 days.  相似文献

Overexpression of the mature form of hyaluronan-binding protein 1 (HABP1/gC1qR/p32), a ubiquitous multifunctional protein involved in cellular signaling, in normal murine fibroblast cells leads to enhanced generation of reactive oxygen species (ROS), mitochondrial dysfunction, and ultimately apoptosis with the release of cytochrome c. In the present study, human liver cancer cell line HepG2, having high intracellular antioxidant levels was chosen for stable overexpression of HABP1. The stable transformant of HepG2, overexpressing HABP1 does not lead to ROS generation, cellular stress, and apoptosis, rather it induced enhanced cell growth and proliferation over longer periods. Phenotypic changes in the stable transformant were associated with the increased "HA pool," formation of the "HA cable" structure, up-regulation of HA synthase-2, and CD44, a receptor for HA. Enhanced cell survival was further supported by activation of MAP kinase and AKT-mediated cell survival pathways, which leads to an increase in CYCLIN D1 promoter activity. Compared with its parent counterpart HepG2, the stable transformant showed enhanced tumorigenicity as evident by its sustained growth in low serum conditions, formation of the HA cable structure, increased anchorage-independent growth, and cell-cell adhesion. This study suggests that overexpression of HABP1 in HepG2 cells leads to enhanced cell survival and tumorigenicity by activating HA-mediated cell survival pathways.  相似文献

Mammalian cells deploy autophagy to defend their cytosol against bacterial invaders. Anti‐bacterial autophagy relies on the core autophagy machinery, cargo receptors, and “eat‐me” signals such as galectin‐8 and ubiquitin that label bacteria as autophagy cargo. Anti‐bacterial autophagy also requires the kinase TBK1, whose role in autophagy has remained enigmatic. Here we show that recruitment of WIPI2, itself essential for anti‐bacterial autophagy, is dependent on the localization of catalytically active TBK1 to the vicinity of cytosolic bacteria. Experimental manipulation of TBK1 recruitment revealed that engagement of TBK1 with any of a variety of Salmonella‐associated “eat‐me” signals, including host‐derived glycans and K48‐ and K63‐linked ubiquitin chains, suffices to restrict bacterial proliferation. Promiscuity in recruiting TBK1 via independent signals may buffer TBK1 functionality from potential bacterial antagonism and thus be of evolutionary advantage to the host.  相似文献

Summary In the goldfish, Carassius auratus, morphological and functional aspects of the pituitary gland were studied at the ultrastructural level and six cell types could be distinguished in the pars distalis. Acidophilic cells of the rostral pars distalis were identified as prolactin cells, the chromophobic cells of the rostral pars distalis as ACTH cells, the non-globular basophilic cells of the rostral and the proximal pars distalis as TSH cells, the globular basophils of the proximal pars distalis as gonadotropic cells and the acidophils of the proximal pars distalis as somatotrophs.Besides some of the well established criteria of morphological and functional identification of different cell types, two new approaches have been used in the present study. One was to express the electron density of secretory granules objectively by means of a photometric method. It was found that both types of acidophilic cells which produce the proteohormones prolactin and somatotropin respectively, had granules with the highest electron densities. The basophilic cells producing the glycoproteins gonadotropin and TSH respectively, possessed granules of intermediate electron density whereas the chromophobic cells storing the peptide hormone ACTH had granules of lowest densities. The second new approach was the administration of the synthetic mammalian releasing hormones LH-RP and TRF, which helped in identifying gonadotropic and thyrotropic cells respectively. Pyoverdine is the primary siderophore of the gram-negative bacterium Pseudomonas aeruginosa. The pyoverdine region was recently identified as the most divergent locus alignable between strains in the P. aeruginosa genome. Here we report the nucleotide sequence and analysis of more than 50 kb in the pyoverdine region from nine strains of P. aeruginosa. There are three divergent sequence types in the pyoverdine region, which correspond to the three structural types of pyoverdine. The pyoverdine outer membrane receptor fpvA may be driving diversity at the locus: it is the most divergent alignable gene in the region, is the only gene that showed substantial intratype variation that did not appear to be generated by recombination, and shows evidence of positive selection. The hypothetical membrane protein PA2403 also shows evidence of positive selection; residues on one side of the membrane after protein folding are under positive selection. R', previously identified as a type IV strain, is clearly derived from a type III strain via a 3.4-kb deletion which removes one amino acid from the pyoverdine side chain peptide. This deletion represents a natural modification of the product of a nonribosomal peptide synthetase enzyme, whose consequences are predictive from the DNA sequence. There is also linkage disequilibrium between the pyoverdine region and pvdY, a pyoverdine gene separated by 30 kb from the pyoverdine region. The pyoverdine region shows evidence of horizontal transfer; we propose that some alleles in the region were introduced from other soil bacteria and have been subsequently maintained by diversifying selection.  相似文献

We report here a novel carrier of quantum dots (QDs) for intracellular labeling. Monodisperse hybrid nanoparticles (38 nm in diameter) of QDs were prepared by simple mixing with nanogels of cholesterol-bearing pullulan (CHP) modified with amino groups (CHPNH2). The CHPNH2-QD nanoparticles were effectively internalized into the various human cells examined. The efficiency of cellular uptake was much higher than that of a conventional carrier, cationic liposome. These hybrid nanoparticles could be a promising fluorescent probe for bioimaging.  相似文献
