The insolubility of nitrile substrates in aqueous reaction mixture decreases the enzymatic reaction rate. We studied the interaction of fourteen water miscible organic solvents with immobilized nitrile hydrolyzing biocatalyst. Correlation of nitrilase function with physico-chemical properties of the solvents has allowed us to predict the enzyme behavior in such non-conventional media. Addition of organic solvent up to a critical concentration leads to an enhancement in reaction rate, however, any further increase beyond the critical concentration in the latter leads to the decrease in catalytic efficiency of the enzyme, probably due to protein denaturation. The solvent dielectric constant (epsilon) showed a linear correlation with the critical concentration of the solvent used and the extent of nitrile hydrolysis. Unlike alcohols, the reaction rate in case of aprotic solvents could be linearly correlated to solvent log P. Further, kinetic analysis confirmed that the affinity of the enzyme for its substrate (K (m)) was highly dependent upon the aprotic solvent used. Finally, the prospect of solvent engineering also permitted the control of enzyme enantioselectivity by regulating enantiomer traffic at the active site. 相似文献

3‐Hydroxypropionaldehyde (3HPA) is an important specialty chemical which can be produced from glycerol using resting cells of Lactobacillus reuteri. This biocatalytic route, however, suffers from substrate‐ and product‐mediated loss of enzyme activity within 2 h of biotransformation. In order to overcome the inhibitory effects of 3HPA, complex formation with sodium bisulfite was investigated, optimized and applied for in situ capture of the aldehyde during biotransformation of glycerol in a fed‐batch process. As a result, the activity of the cells was maintained for at least 18 h. The 3HPA produced per gram cell dry weight was increased 5.7 times compared to the batch production process, and 2.2 times compared to fed‐batch process without in situ complex formation. Reports on the incidence of single clockwise occipital whorls as opposed to double and counterclockwise whorls reveal no consistent geographical pattern. Earlier workers arrived at different conclusions. While Bernstein ('25b) and Schwartzburg ('27) suggested a dominant inheritance of counter-clockwise whorls, others like Winchester ('58) and Montagu ('63) believed the clockwise condition to be dominant. In the light of these and some other studies, Scheinfeld's ('56) remarks that chance factor in prenatal development may influence dominance, seem appropriate. The results of present study indicate that clockwise and anti-clockwise conditions show inconsistent variation which may be the result of change element rather than any specific mode of distribution. Two groups from India: 164 Pandits of Kashmir, pre-dominantly Caucasian and 95 Tibetan refugees, predominantly Mongoloid are contrasted. The incidence of a single clockwise whorl among Pandits is 67%, whereas among Tibetans it is 78%. A review of other populations studied shows greater variance within than between the so-called major racial stocks. 相似文献

Saccharomyces cerevisiae cells exposed to 43 degrees C (normal being 30 degrees C) exhibit the synthesis of heat shock proteins (hsps). Time course studies indicated that the major hsps (97 kDa, 85 kDa and 70 kDa family) are induced within 10 min. of heat shock and attain maximum amount with two hours of treatment. The viability of cells decreased by 99% when directly frozen into liquid nitrogen. However, a prior heat shock (2 hours) increased the cell survival by 20-30 fold. Such an effect of prior heat shock treatment could be supported by light and electron microscopical studies. Differential scanning calorimetric analysis of whole cells revealed that heat shock treatment decreases the denaturation (delta H) of total cellular proteins. A direct correlation between the degree of hsp inducibility and protection against freezing and thawing injury was observed. Cycloheximide treatment curtailed the synthesis of hsps as well as protection against subsequent freezing. This suggests that prior heat shock treatment protects the cells from freezing injury and, furthermore, that hsps can act as biological cryoprotectants. 相似文献

Summary The cells of Lactobacillus casei were adsorbed to Poraver, foam glass particles pretreated with polyethylenimine (PEI). Exposure of cells for a relatively short period to Poraver beads coated with a high concentration of PEI resulted in maximal adsorption with good retention of metabolic activity. The immobilized cells were tested in packed-bed and stirred-tank reactors for lactic acid production. Stirred-tank operations were more effective in terms of productivity but the support was sensitive to attrition. The beads exhibited good mechanical stability to withstand pressure in the packed-bed reactor.

Two hundred and eleven dogs (including strictly house and stray dogs) and 170 cattle in and around the city of Madras, India
were screened for the presence of dermatophytosis. 106 strains of dermatophytes (89 strains from dogs and 17 strains from
bovines) were isolated. 57/106 strains were Trichophyton mentagrophytes var. mentagrophytes and 42/106 strains were of the
Microsporum gypseum complex. 5 strains of T. rubrum and 2 strains of T. simii were also obtained in culture. A predominance
of M. gypseum complex isolates was recorded in stray dogs and cattle and T. mentagrophytes var. mentagrophytes and T. rubrum
in strictly house dogs. The family history of the owners of the most of the dogs had clear records of dermatophytosis. Further,
the owners of the 11 dogs that yielded T. mentagrophytes var. mentagrophytes had either tinea corporis or tinea pedis. The
etiological agent of all the 11 human cases was T. mentagrophytes var. interdigitale. Similarly the owners of 4 of the 5 dogs
that yielded T. rubrum were known T. rubrum patients. All these patients responded to oral griseofulvin or ketaconozole, but
the recurrence of lesions was noted with the cessation of treatment. None of the patients had onychomycosis and the family
history of all the patients revealed no reports of T. rubrum infections. The pet dogs were presumed to be the source of re-infection.
Reversed transmission of dermatophytes from humans to animals may be the reason for the selective predominance of these organisms
in strictly house dogs. They also may act as sources of reinfection. Most of the animals had small, occult, scattered lesions.
These lesions may either go unnoticed or are ignored by the owners of the animals. The taxonomic status of T. mentagrophytes
var. mentagrophytes and T. mentagrophytes var. interdigitale was aligned to their teleomorph Arthroderma vanbreuseghemii.
Our study suggests that the periodic screening and medication of all live-stock are essential for the prevention and management
of the public health problem caused by dermatophytes.
