Vitamin D and bone   总被引:5,自引:0,他引:5  
It is now well established that supraphysiological doses of 1alpha,25-dihydroxyvitamin D(3) [1alpha,25(OH)(2)D(3)] stimulate bone resorption. Recent studies have established that osteoblasts/stromal cells express receptor activator of NF-kappaB ligand (RANKL) in response to several bone-resorbing factors including 1alpha,25(OH)(2)D(3) to support osteoclast differentiation from their precursors. Osteoclast precursors which express receptor activator of NF-kappaB (RANK) recognize RANKL through cell-to-cell interaction with osteoblasts/stromal cells, and differentiate into osteoclasts in the presence of macrophage-colony stimulating factor (M-CSF). Osteoprotegerin (OPG) acts as a decoy receptor for RANKL. We also found that daily oral administration of 1alpha,25(OH)(2)D(3) for 14 days to normocalcemic thyroparathyroidectomized (TPTX) rats constantly infused with parathyroid hormone (PTH) inhibited the PTH-induced expression of RANKL and cathepsin K mRNA in bone. The inhibitory effect of 1alpha,25(OH)(2)D(3) on the PTH-induced expression of RANKL mRNA occurred only with physiological doses of the vitamin. Supraphysiological doses of 1alpha,25(OH)(2)D(3) increased serum Ca and expression of RANKL in vivo in the presence of PTH. These results suggest that the bone-resorbing activity of vitamin D does not occur at physiological dose levels in vivo. A certain range of physiological doses of 1alpha,25(OH)(2)D(3) rather suppress the PTH-induced bone resorption in vivo, supporting the concept that 1alpha,25(OH)(2)D(3) or its derivatives are useful for the treatment of various metabolic bone diseases such as osteoporosis and secondary hyperparathyroidism.  相似文献

Transgenic insects have been artificially produced to study functions of interesting developmental genes, using insect transposons such as piggyBac. In the case of the cricket, however, transgenic animals have not yet been successfully artificially produced. In the present study, we examined whether the piggyBac transposon functions as a tool for gene delivery in embryos of Gryllus bimaculatus. We used either a piggyBac helper plasmid or a helper RNA synthesized in vitro as a transposase source. An excision assay revealed that the helper RNA was more effective in early Gryllus eggs to transpose a marker gene of eGFP than the helper plasmid containing the piggyBac transposase gene driven by the G. bimaculatus actin3/4 promoter. Further, only when the helper RNA was used, somatic transformation of the embryo with the eGFP gene was observed. These results suggest that the piggyBac system with the helper RNA may be effective for making transgenic crickets.  相似文献

The liverwort Marchantia polymorpha contains high proportions of arachidonic and eicosapentaenoic acids. In general, these C20 polyunsaturated fatty acids (PUFA) are synthesized from linoleic and alpha -linolenic acids, respectively, by a series of reactions catalyzed by Delta(6)-desaturase, an ELO-like enzyme involved in Delta(6) elongation and Delta(5)-desaturase. Here we report the isolation and characterization of the cDNAs, MpDES6, MpELO1 and MpDES5, coding for the respective enzymes from M. polymorpha. Co-expression of the MpDES6, MpELO1 and MpDES5 cDNAs resulted in the accumulation of arachidonic and eicosapentaenoic acids in the methylotrophic yeast Pichia pastoris. Interestingly, Delta(6) desaturation by the expression of the MpDES6 cDNA appears to occur both in glycerolipids and the acyl-CoA pool, although other lower-plant Delta(6)-desaturases are known to have a strong preference for glycerolipids.  相似文献

The hepatitis C virus (HCV) replicon system is a potent tool for understanding the mechanisms of HCV replication and proliferation, and for the development of treatments for patients with HCV. Recently, we established an HCV subgenomic replicon (50-1) using HCV genome RNA obtained from the cultured human T cell line MT-2C infected with HCV (isolate 1B-1) in vitro. In order to further obtain other HCV replicons without difficulty, we generated a replicon RNA library derived from human non-neoplastic hepatocytes infected with HCV (isolate 1B-2) in vitro. Upon transfection of the generated RNA library to "cured cells," from which the 50-1 subgenomic replicon was eliminated by prolonged treatment with interferon-alpha, we successfully established a new HCV subgenomic replicon, 1B-2R1. We characterized 1B-2R1 replicon in terms of efficiency of replication, HCV sequence, and sensitivity to interferons. The results revealed that the replication level of the 1B-2R1 replicon was comparable to that of the 50-1 replicon. We also found that the 1B-2R1 replicon possessed an HCV sequence distinct from those of other replicons established to date, and that the 1B-2R1 replicon was sensitive to interferon-alpha, interferon-beta, and interferon-gamma. Taken together, present results indicate that the replicon RNA library generated using an in vitro HCV infection system is useful for the establishment of an HCV subgenomic replicon.  相似文献

The human induced input of reactive N into the globalbiosphere has increased to approximately 150 Tg N eachyear and is expected to continue to increase for theforeseeable future. The need to feed (125 Tg N) andto provide energy (25 Tg N) for the growing worldpopulation drives this trend. This increase inreactive N comes at, in some instances, significantcosts to society through increased emissions of NOx,NH3, N2O and NO3 and deposition of NOy and NHx.In the atmosphere, increases in tropospheric ozone andacid deposition (NOy and NHx) have led toacidification of aquatic and soil systems and toreductions in forest and crop system production. Changes in aquatic systems as a result of nitrateleaching have led to decreased drinking water quality,eutrophication, hypoxia and decreases in aquatic plantdiversity, for example. On the other hand, increaseddeposition of biologically available N may haveincreased forest biomass production and may havecontributed to increased storage of atmospheric CO2 inplant and soils. Most importantly, syntheticproduction of fertilizer N has contributed greatly tothe remarkable increase in food production that hastaken place during the past 50 years.The development of policy to control unwanted reactiveN release is difficult because much of the reactive Nrelease is related to food and energy production andreactive N species can be transported great distancesin the atmosphere and in aquatic systems. There aremany possibilities for limiting reactive N emissionsfrom fuel combustion, and in fact, great strides havebeen made during the past decades. Reducing theintroduction of new reactive N and in curtailing themovement of this N in food production is even moredifficult. The particular problem comes from the factthat most of the N that is introduced into the globalfood production system is not converted into usableproduct, but rather reenters the biosphere as asurplus. Global policy on N in agriculture isdifficult because many countries need to increase foodproduction to raise nutritional levels or to keep upwith population growth, which may require increaseduse of N fertilizers. Although N cycling occurs atregional and global scales, policies are implementedand enforced at the national or provincial/statelevels. Multinational efforts to control N loss tothe environment are surely needed, but these effortswill require commitments from individual countries andthe policy-makers within those countries.  相似文献

Six carabidicolous species of Laboulbeniales are reported as new for the Japanese mycoflora. They areDixomyces stomonaxi, Laboulbenia picardii, L. tenera, L. slackensis, L. aristata andL. kwangjuensis. Two forms ofD. stomonaxi are distinguished, one form of which resemblesD. nigromarginatus. A remarkable ornamentation consisting of a coillike pattern occurs on the receptacle ofL. picardii. InL. tenera andL. slackensis, the outer appendage has somewhat constricted, blackened septa near the base.Laboulbenia aristata andL. kwangjuensis have spirally arranged outer wall cells. To attempt to develop physicochemical and physiological modelling for methane transport from the rhizosphere to the atmosphere through rice plants, methane flux, methane concentration in the soil water, and the biomass of rice were measured in lysimeter rice paddies (2.5 × 4 m, depth 2.0 m) once per week throughout the entire growing season in 1992 at Tsukuba, Japan. The addition of exogenous organic matter (rice straw) or soil amendments with the presence or absence of vegetation were also examined for their influence on methane emissions. The total methane emission over the growing season varied from 3.2 g CH4 m-2 y-1 without the addition of rice straw to 49.7 g CH4 m-2 y-1 with rice straw and microbiological amendment. In the unvegetated plot with the addition of rice straw, there was much ebullition of gas bubbles, particularly in the summer. The annual methane emission due to the ebullition of gas bubbles,from the unvegetated plot with the addition of rice straw was estimated to be almost the same as that from the vegetated site with the addition of rice straw. In the early growth stage, the methane flux can be analyzed by the diffusion model (Flux=Methane concentration × Conductance of rice body) using parameters for methane concentration in the soil water as a difference in concentration between the atmosphere and the rhizosphere, and for the biomass of rice as a conductance of rice body. On the other hand, although the diffusion model was inapplicable to a large extent from the middle to late growth stage, methane flux could be estimated by air temperature and concentration in the soil water. Thus, methane transport from the rhizosphere to the atmosphere through rice plants consisted of two phases: one was an explainable small part by diffusion in rice body; the other was a large part strongly, governed by air temperature. We established a sensitive RRA system for human FSH, employing PMS-treated immature rat ovary. The dissociation constant of the binding of the receptor preparation to NIAMDD human FSH-2 was 1.15 x 10(-10) M. The standard curve was obtained with 0.2-25.6 ng/tube of NIAMDD hFSH-2. Purified hLH, hTSH, and HCG had no significant effect on the binding. When the anterior pituitary homogenates obtained from humans were assayed by this system, the intra-assay and inter-assay coefficients of variation were 11.9% and 13.4% respectively, and the assay values correlated well with those obtained by RIA. This assay is applicable for the measurement of FSH in serum, when the non-specific inhibitor effects of serum are compensated for by the addition of merthiolate and when FSH-free serum is used instead of the buffer for the standard curve. The intra-assay and inter-assay coefficients of variation were 9.31% and 19.7% respectively. The assay values correlated with those obtained by RIA under the same physiological state. The ratio of the assay values RRA/RIA, was dependent upon the physiological state, e.g. 6.29 in men, 3.84 and 4.18 in women at follicular and luteal phase respectively and 2.40 in menopausal women. During the menstrual cycle, our results showed that the value of RRA/RIA derived from serum did not change significantly.  相似文献
