Stimulation of polypeptide synthesis by spermidine at the level of initiation in rabbit reticulocyte and wheat germ cell-free systems

It is shown that the stimulation of eukaryotic polypeptide synthesis by spermidine is due to the stimulation at the level of initiation by following reasons. The incorporation of formylmethionine into polypeptides was stimulated by spermidine at the same degree to the incorporation of leucine into polypeptides. Fluorography of the polypeptides formed showed that the number of chains of individual protein synthesized was larger when spermidine was added. The formation of the complex of Met-tRNA_f, globin mRNA and 40-S ribosomal subunits was stimulated by spermidine.     

Correlation between 30S ribosomal proteins of Bacillus stearothermophilus and Escherichia coli

Summary The 30S ribosomal proteins from Bacillus stearothermophilus strains 799 and 10 were purified and correlated with those from E. coli by comparing their two-dimensional electrophoretic mobility, immunological cross-reaction, molecular weight, amino acid composition and partial amino acid sequence. The mode of action of a morphactin,fluoren-9-carboxylic acid

Fluoren-9-carboxylic acid acts not only as an auxin but also as an gibberellin-antagonist. In the standard pea straight test (S₅ section) for auxin it stimulated elongation, the optimum concentration being 10 mg/l. On the other hand, it inhibited elongation at 0.1 mg/l. This inhibitory effect was more marked when younger tissue (S₁ section) which also responds to gibberellin was used. Interaction of FCA and IAA in the S₅ section has shown that at higher concentration of IAA there seemed to be a suppraoptimal effect, indicating that FCA acted as an auxin. However, in the S₁ section, the stimulating effect of GA₃ was markedly inhibited by 0.1 mg/l FCA; 10 mg/l FCA was either additive or less than additive to GA₃. In the cucumber hypocotyl test FCA itself was inactive up to 100 μg/plant, but it inhibited the GA₃-induced elongation. This inhibition was overcome by increasing the dosage of GA₃. In the same material, the IAA-induced elongation was not affected by FCA. These results indicate that whether FCA acts as an auxin or a gibberellin-antagonist depends on whether the tissue is sensitive to gibberellin and/or auxin.     

Relationship of IAA-oxidase Activity to Gibberellinand IAA-induced Elongation of Light-grown Cucumber Seedlings

The growth and IAA-oxidase activity of light-grown cucumber seedlings (cv. Aonagajibae) were investigated in response to GA₃ and IAA. Both GA₃ and IAA induced significant elongation of the hypocotyl. The fresh and dry weights of the hypocotyl increased due to GA₃ or IAA treatment, whereas no significant change was observed in the cotyledons of GA₃-treated seedlings as compared with the controls. The fresh and dry weights of IAA-treated cotyledons were both lower than those of controls. Treatment with GA₃ or IAA resulted in retardation of IAA-oxidase activity in the hypocotyl and cotyledons. The degree of retardation was less in the cotyledons than in the hypocotyl. An inverse relationship was recognized between GA₃- or IAA-induced elongation and IAA-oxidase activity in the hypocotyl. The auxin-mediated mechanism for gibberellin action was discussed.     

Reproductive behavior,eggs and larvae of a lutjanid fish,Lutjanus stellatus,observed in an aquarium

Reproductive behavior and early life history of the white spotted snapper,Lutjanus siellatus Akazaki, are described from aquarium observations. Spawning took place between a female and 2–12 males in the early hours of each evening from mid-May to mid-June, 1984. Six easily distinguishable behavior patterns were involved in the spawning sequence: a) Schooling; b) Searching; c) Nuzzling; d) Rushing toward the surface; e) Spawning; and f) Post spawning. The species was considered to be a group spawner.     

Simplified procedure for purification of Bordetella bronchiseptica dermonecrotic toxin

Abstract Bordetella bronchiseptica dermonecrotic toxin was purified by a simplified method. The method consisted of SP Toyopearl 650M chromatography and high performance liquid chromatography on a TSK gel G3000SW column. 47.5% of the activity of the crude cell extract was recovered. The purified toxin behaved as a homogeneous protein in sodium dodecyl sulfate polyacrylamide gel electrophoresis, high performance liquid chromatography, and agar gel double diffusion tests.     

Purification and Further Characterization of Pyrenoid Proteins and Ribulose-1,5-bisphosphate Carboxylase-Oxygenase from the Green Alga Bryopsis maxima

Pyrenoid proteins and ribulose-1,5-bisphosphate carboxylase-oxygenase(RuBisCO) in the green alga Bryopsis maxima were purified tohigh degrees and their peptide compositions were studied bySDS-polyacrylamide gel electrophoresis. RuBisCO had a largesubunit of 50 kDa and a small one of 16 kDa. The apparent molecularweight of the purified RuBisCO was estimated as 460 kDa by gelfiltration. Pyrenoid proteins had two major polypeptides: 52kDa and 17 kDa. The peptide map of the 52 kDa pyrenoid polypeptidecoincided well with that of the large subunit of RuBisCO, stronglysuggesting that the major component of the pyrenoid of thisalga was RuBisCO. We attempted to survey the distribution ofRuBisCO in the chloroplasts. The results suggested that muchof the RuBisCO of Bryopsis maxima was localized in the pyrenoid.The pyrenoid also contained more than 10 minor polypeptidesnot found in the RuBisCO fraction. The minor polypeptides comprisedabout 15% of the total pyrenoid protein and differed from thepolypeptides of the thylakoid membranes and from those foundin the starch grains surrounding the pyrenoid. Gibberellin-Induced Changes in Starch Content in Isolated Chloroplasts of Light-Grown Cucumber Hypocotyls

Gibberellin A₃ (GA₃) was found to reduce the starch contentin chloroplasts isolated from light-grown cucumber (cv. Aonagajibai)hypocotyls. Chloroplasts incubated with 50 mM sucrose showedan increase in starch content. The GA₃-induced reduction occurredboth in the presence and absence of exogenous sucrose.
