Proteins of the cytosolic pathway for iron‐sulphur (FeS) cluster assembly are conserved, except that plants lack a gene for CFD1 (Cytosolic FeS cluster Deficient 1). This poses the question of how NBP35 (Nucleotide‐Binding Protein 35 kDa), the heteromeric partner of CFD1 in metazoa, functions on its own in plants. Firstly, we created viable mutant alleles of NBP35 in Arabidopsis to overcome embryo lethality of previously reported knockout mutations. RNAi knockdown lines with less than 30% NBP35 protein surprisingly showed no developmental or biochemical differences to wild‐type. Substitution of Cys14 to Ala, which destabilized the N‐terminal Fe4S4 cluster in vitro, caused mild growth defects and a significant decrease in the activity of cytosolic FeS enzymes such as aconitase and aldehyde oxidases. The DNA glycosylase ROS1 was only partially decreased in activity and xanthine dehydrogenase not at all. Plants with strongly depleted NBP35 protein in combination with Cys14 to Ala substitution had distorted leaf development and decreased FeS enzyme activities. To find protein interaction partners of NBP35, a yeast‐two‐hybrid screen was carried out that identified NBP35 and DRE2 (Derepressed for Ribosomal protein S14 Expression). NBP35 is known to form a dimer, and DRE2 acts upstream in the cytosolic FeS protein assembly pathway. The NBP35–DRE2 interaction was not disrupted by Cys14 to Ala substitution. Our results show that NBP35 has a function in the maturation of FeS proteins that is conserved in plants, and is closely allied to the function of DRE2.  相似文献

Plasminogen activator inhibitor-1 (PAI-1) belongs to the serpin family of serine proteinase inhibitors. Serpins inhibit their target proteinases by an ester bond being formed between the active site serine of the proteinase and the P1 residue of the reactive centre loop (RCL) of the serpin, followed by insertion of the RCL into beta-sheet A of the serpin. Concomitantly, there are conformational changes in the flexible joint region lateral to beta-sheet A. We have now, by site-directed mutagenesis, mapped the epitope for a monoclonal antibody, which protects the inhibitory activity of PAI-1 against inactivation by a variety of agents acting on beta-sheet A and the flexible joint region. Curiously, the epitope is localized in alpha-helix C and the loop connecting alpha-helix I and beta-strand 5A, on the side of PAI-1 opposite to beta-sheet A and distantly from the flexible joint region. By a combination of site-directed mutagenesis and antibody protection against an inactivating organochemical ligand, we were able to identify a residue involved in conferring the antibody-induced conformational change from the epitope to the rest of the molecule. We have thus provided evidence for communication between secondary structural elements not previously known to interact in serpins.  相似文献

The palp morphology of Prionospio fallax Söderström, 1920 from Sweden and Prionospio cf. saldanha Day, 1961 from Thailand was examined with a scanning electron microscope. Prionospio fallax was also studied in vivo using light-microscopy. Both species have grooved feeding palps, adorned with up to five ciliary characters: frontal cilia, transverse ciliary bands (or bandlets), latero-frontal cirri, lateral cilia and randomly distributed non-motile cirri. All, except the frontal cilia and non-motile cirri, are asymmetrically arranged relative to the long axis of the palps. Prionospio fallax possesses transverse bandlets and the other four groups, while P. cf. saldanha has transverse bands (consisting of several contiguous bandlets), frontal cilia and some randomly scattered cirri. Asymmetrical palp ciliation was previously only known in Marenzelleria viridis (Verril, 1873) and the genus Scolelepis Blainville, 1828. The newly recognised transverse ciliary bands and bandlets are considered to be homologous with the transverse ciliary rows found basally on the palps of Paraprionospio pinnata (Ehlers, 1901). This multistate character (named transverse cilia) may prove useful in elucidating the phylogeny of the Prionospio-complex of genera.  相似文献

Patch use under predation risk often results in a change of feeding behaviour in the prey animals. However, such changes only appear if the animals are able to assess under which predation pressure they live. We investigated patch use of Mastomys natalensis under different conditions of avian predation pressure.
In replicated maize field plots in Morogoro, Tanzania, avian predators were allowed under natural conditions (control), attracted with perches and nest boxes or kept out with nets. During four one‐week periods in late 1999, we measured rodent feeding decisions with the giving‐up density (GUD) method. Trays with known amounts of millet seeds in sand were placed in pairs, one of them under a cover, the other one in the open. M. natalensis mice were expected to give up sooner in the open trays than in those with cover. We hypothesised that M. natalensis mice could assess the ambient predation pressure leading to larger difference in GUD between covered and non‐covered trays in the plots where predators were attracted. We also made video recordings of the rodent activity at a pair of trays in each treatment. The GUD‐values were significantly lower for the covered trays but predation pressure did not affect this difference. The video observations showed that in the control and netted plots the animals visited trays equally frequently regardless of the cover, while the visits in the predator‐attracted plots occurred significantly more often in the covered trays. We conclude that M. natalensis can assess the ambient predation pressure and adapt its behaviour at a feeding patch. However, the variation in predation pressure in our experiment was not obvious from the GUD. Moreover, we found a strong relation between rodent density and GUD, which may mask variations in perceived predation pressure. Similar GUD values may be reached in different ways and we present models to investigate whether animals’ decision to forage at a food patch is only affected by the seed density at that patch, not by that at a neighbour patch.  相似文献

Background

Despite men being less prone to develop autoimmune diseases, male sex has been associated with a more severe disease course in several systemic autoimmune diseases. In the present study, we aimed to investigate differences in the clinical presentation of primary Sjögren’s syndrome (pSS) between the sexes and establish whether male sex is associated with a more severe form of long-term pSS.

Methods

Our study population included 967 patients with pSS (899 females and 68 males) from Scandinavian clinical centers. The mean follow-up time (years) was 8.8 ± 7.6 for women and 8.5 ± 6.2 for men (ns). Clinical data including serological and hematological parameters and glandular and extraglandular manifestations were compared between men and women.

Results

Male patient serology was characterized by more frequent positivity for anti-Ro/SSA and anti-La/SSB (p = 0.02), and ANA (p = 0.02). Further, men with pSS were more frequently diagnosed with interstitial lung disease (p = 0.008), lymphadenopathy (p = 0.04) and lymphoma (p = 0.007). Conversely, concomitant hypothyroidism was more common among female patients (p = 0.009).

Conclusions

We observe enhanced serological responses and higher frequencies of lymphoma-related extraglandular manifestations in men with pSS. Notably, lymphoma itself was also significantly more common in men. These observations may reflect an aggravated immune activation and a more severe pathophysiological state in male patients with pSS and indicate a personalized managing of the disease due to the influence of the sex of patients with pSS.

Recent marine planktonic diatom assemblages from 47 sediment surface samples retrieved from the eastern equatorial Pacific yielded four significant Q-mode factors that are related to surface water properties. They include a northern subtropical (Factor 1), a southern subtropical (Factor 2), a Peru Current (Factor 3) and an eastern Equatorial Pacific (Factor 4) factor.A transfer function (based on 42 surface sediment samples) with a productivity range of 109 to 210 gC m–2 y–1 was defined using the geographic distribution of the northern and southern subtropical assemblages (Factor 1 and 2) and their relation to mapped surface water primary productivity. This transfer function has a standard error of estimate of 11.9 gC m–2 y–1 and it is used to estimate late Pleistocene paleo-productivity changes at ODP Leg 111 Site 677 off Ecuador.The Site 677 samples (usually spaced 10 cm apart and with a linear sedimentation rate of 4.3 cm y–3) were the same samples that were used for stable oxygen/carbon isotopic analysis from planktonic Foraminifera. The well constrained oxygen isotope stratigraphy permitted assignment of accurate ages to downcore samples.The interval between oxygen isotope stages 3 and 9 had variable productivity that is generally higher than present values of 175 gC m–2 y–1 A major decline in paleo-productivity below the present value occurred in stage 9, the early part of stage 8 and in the early and late stage 5.  相似文献

The relative contribution of each parent when providing for the fledglings has been recorded in only a few raptor species. We studied prey deliveries by Eurasian Sparrowhawk Accipiter nisus parents to fledglings at seven nests in southern Norway. Parents and young were fitted with radio-transmitters. Males delivered a larger number of prey to the young than did females throughout the post-fledging period (on average c.  80% of the deliveries). Two females were never observed to deliver food to the offspring, and their mates apparently raised the young to independence alone. The duration of the post-fledging period was positively related to per-capita delivery rate in the late stage.  相似文献
