Summary Body protein sparing during starvation has been examined in fat and lean Svalbard ptarmigan. Protein utilization was determined from daily N excretion and from the rate of decrease in body mass. Changes in plasma concentrations of -hydroxybutyrate, free fatty acids, glucose, and uric acid were also recorded. When fat birds were starved for 15 days protein catabolism initially fell (phase I) and was thereafter kept low (phase II). This was evident from the temporal pattern in both N excretion and body mass loss. In two birds, N excretion eventually increased, revealing enhanced protein catabolism and thus a third phase of starvation. Changes in protein utilization were paralleled by changes in plasma uric acid. Approximately 9% of the energy demand was covered by breakdown of body protein during phase II. The importance of fat catabolism in providing energy was indicated by markedly elevated plasma levels of -hydroxybutyrate and free fatty acids. When lean birds were starved for 5 days there appeared to be no phase II. The temporal pattern of body mass loss indicated phase I and III but that of N excretion only phase III. The relative contribution of body protein to energy demand increased from 22% at day 2 to 41% at the end of starvation and was paralleled by increased plasma uric acid. When data from lean and fat birds were pooled, the changes in uric acid and N excretion were highly correlated (r=0.92, P<0.001), indicating that plasma uric acid is a reliable index of protein breakdown in starving Svalbard ptarmigan. In conclusion, starving fat Svalbard ptarmigan have a much greater capacity to spare body protein than lean birds. Twenty-four patients were analyzed for the development of HAMA (human antimouse antibodies) after being treated with repeated doses (200–500 mg) of the mouse monoclonal antibody (MAb) 17-1A. All patients developed anti-17-1A IgG antibodies, and most of them also developed IgM antibodies. In only two patients could immune complexes be demonstrated. Allergic reactions were rare (1.9%). In an extended study, a further 19 patient were analyzed for an idiotypic response. Forty-one out of 43 patients developed antiidiotypic antibodies (ab2), and 20 of these also antianti-idiotypic antibodies (ab3). Ab3 + patients responded significantly better (p=0.01) and survived longer (p<0.001) compared to ab3 patients. In this study, we showed that MAb 17-1A could be repeatedly given on a safe basis. The development of high titers of HAMA did not cause significant clinical problems when further repeated infusions of MAb 17-1A were given. The development of an idiotypic response also indicate that the induction of HAMA might be beneficial and not harmful to the patient.  相似文献

In September 1987 the shallow, eutrophic, Lake Mosvatn was treated with rotenone to eliminate planktivorous fish (mainly whitefish,Coregonus lavaretus, L.), and the effects were studied. The first summer after treatment the zooplankton community changed markedly from rotifer dominance and few grazers, to a community with few rotifers and many grazers. Accordingly there was a fivefold increase in the biomass ofDaphnia galeata. Adult females ofD. galeata approximately doubled in weight. The decrease in rotifer biomass was probably mainly due to a loss of food by competition with the daphnids. The phytoplankton community was also markedly affected. Prior to treatment Secchi depth was 1.7 m and Chl-a 23μg l−1 in the summer. After treatment there was an increase in the proportion of small and gelatinous algae and the mean chlorophyll concentration fell to 7μg Chl-a l−1. Secchi depth increased to>2.3 m (bottom-sight most of the season). After the treatment there were also fewer cyanobacterial blooms. This seems to be related to oligotrophication caused indirectly by increased grazing by the zooplankton. Total nutrient concentrations were affected. Prior to treatment the mean summer concentration of total phosphate was 44μg P l−1. This decreased to 29μg P l−1 in the first summer and 23μg P l−1 the second summer after the treatment. Total nitrogen decreased from 0.68 mg N l−1 before treatment to 0.32 mg N l−1 the first summer after the treatment. The phosphate loading was not reduced, therefor it can be concluded that the fish removal provided a biomanipulation which caused the more oligotrophic conditions.  相似文献

Summary Cells immunoreactive with an anti-LHRH serum were visualized in the brain of the three-spined stickleback, Gasterosteus aculeatus, by means of the PAP technique. Positive cells were found in a periventricular position in the nucleus praeopticus pars magnocellularis, the nucleus dorsomedialis thalami, the nucleus ventromedialis thalami, the nucleus periventricularis posterior, and in the periventricular dorsomedian tegmentum. K+-stimulated ATPase activity of a plasmalemma-enriched fraction from excised roots of oat was triphasic in the range 5 to 80 millimolar KCl. The phases obeyed Michaelis-Menten kinetics and were separated from each other by jumps or sharp breaks at about 10 and 20 millimolar. Stimulation by alkali cations was in the order K+ > Rb+ > Na+ > Cs+ > Li+ or in a closely related sequence. The specificity reflected differences in Vmax, not in affinity (Km−1). Stimulation by the organic cations ethanolamine and choline in the interval 11 to 80 millimolar appeared monophasic rather than biphasic. Substitution on the quaternary nitrogen of the amino alcohols decreased their effectiveness, as did extension and branching of the chain. Stimulation was maximal at about pH 7 both for K+ and choline.  相似文献

Total thyroxine (TT4) and free thyroxine index (FT4I) were measured in peripheral plasma of cows. The samples were collected at the time of insemination from 66 cows showing pronounced signs of the heat and from 56 cows showing weak or silent heat. Kidney β-glucuronidase activity in C57BL/Kl and DBA/2/Kl male mice differs about tenfold, C57 giving low and DBA high values. Another C57 subline, C57BL/6J, has slightly higher activity than C57BL/Kl. There is an association between the kidney glucuronidase activity and coat color determined by the buff locus, which indicates that part of the variation is due to differences at the Gur locus. The bf allele per se raises the activity of the enzyme. The backcross distributions give evidence that at least one more locus is involved.  相似文献

Summary The purpose of this study was to try to differentiate histochemically between the various enzymes which may catalyze the hydrolysis of ATP in developing rat dental tissues. Freeze cut and freeze dried sections of molar and incisor teeth were incubated in lead capture-based media at pH 5.0, 7.2 or 9.4 with one of the following substrates: -glycerophosphate, AMP, ADP, ATP, AMP-PNP and tetrasodium pyrophosphate. To establish the enzymatic nature of the hydrolysis parallel sections were incubated after prior fixation in either formaldehyde or glutaraldehyde.By comparing the enzymatic stainings obtained with the various substrates and at the different pH: s, it was concluded that ATP can be visibly hydrolyzed in rat dental tissues by alkaline phosphatase (stratum intermedium, apical part of maturation ameloblasts, basal part of all ameloblasts, odontoblasts and subodontoblastic layer), specific ATPase (apical and basal parts of secretory ameloblasts) and ATP pyrophosphatase and/or adenylate cyclase (stratum intermedium, odontoblasts). Acid phosphatase, specific ADPase, 5-nucleotidase, inorganic pyrophosphatase, 3:5-cyclic-AMP-phosphodiesterase and adenylate kinase on the other hand, seem not to be engaged in the ATP hydrolysis to such a degree as to complicate the interpretation of the histochemical staining. The alkaline phosphatase part of the ATP hydrolysis appeared to be rather insensitive to aldehyde fixation, while the hydrolysis effected by specific ATPase and ATP pyrophosphatase and/or adenylate cyclase was extinguished after fixation with formaldehyde for 4 h or glutaraldehyde for 10 min.  相似文献

Summary Nitroso-naphthol reacts with tyrosine residues of peptides (and probably also proteins) to yield intensely fluorescent condensation products. This reaction forms the basis of a fluorescence histochemical procedure designed to demonstrate cells that are rich in tyrosine-containing peptides or proteins. In models the method was found to be specific for p-hydroxylated phenolic compounds. Fluorescence was induced also following formaldehyde vapour fixation. With the nitroso-naphthol technique the zymogen granules of gastric chief cells, intestinal Paneth cells, pancreatic acinar cells and certain peptide hormone-secreting cells such as the GH cells in the adenohypophysis, the insulin cells of the pancreatic islets and the calcitonin cells of the thyroid gave intense fluorescence with spectral characteristics indistinguishable from those of the fluorophores of tyrosine-containing peptides.
