Species-specific foraging habits or feeding preferences influence the overall trophic functioning in consumer assemblages. We set out to assess the in situ trophic diversity in a mesograzer assemblage (isopods, amphipods and gastropods) associated with marine littoral macroalgae. More specifically, we set out to establish whether stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) are dependent on primary consumer identity or expressed in relation to the identity of the algal host, i.e. the habitat of the consumer. Consumer locations in bivariate isotope space revealed significant differences among grazer taxa, but no effect of the algal host. This suggests that grazer-specific foraging is more important in driving the qualitative range of feeding in the consumer assemblage than host diversity per se within a macrohabitat, such as a bay or a lagoon. The stable isotope ratios of the mesoherbivores were in line with expectations based on the known feeding ecology of the grazers. However, the trophic diversity suggested by stable isotope analysis implies that even an established concept such as the mesograzer guild may encompass simplifications in terms of functional group membership. 相似文献
We investigated whether maternal prenatal emotions are associated with gestational length and birth weight in the large PREDO Study with multiple measurement points of emotions during gestation.
Methods
Altogether 3376 pregnant women self-assessed their positive affect (PA, Positive and Negative Affect Schedule) and depressive (Center for Epidemiologic Studies Depression Scale, CES-D) and anxiety (Spielberger State Anxiety Scale, STAI) symptoms up to 14 times during gestation. Birth characteristics were derived from the National Birth Register and from medical records.
Results
One standard deviation (SD) unit higher PA during the third pregnancy trimester was associated with a 0.05 SD unit longer gestational length, whereas one SD unit higher CES-D and STAI scores during the third trimester were associated with 0.04–0.05 SD unit shorter gestational lengths (P-values ≤ 0.02), corresponding to only 0.1–0.2% of the variation in gestational length. Higher PA during the third trimester was associated with a significantly decreased risk for preterm (< 37 weeks) delivery (for each SD unit higher positive affect, odds ratio was 0.8-fold (P = 0.02). Mothers with preterm delivery showed a decline in PA and an increase in CES-D and STAI during eight weeks prior to delivery. Post-term birth (≥ 42 weeks), birth weight and fetal growth were not associated with maternal prenatal emotions.
Conclusions
This study with 14 measurements of maternal emotions during pregnancy show modest effects of prenatal emotions during the third pregnancy trimester, particularly in the weeks close to delivery, on gestational length. From the clinical perspective, the effects were negligible. No associations were detected between prenatal emotions and birth weight. 相似文献
The presence of lipoteichoic acid (LTA) in plasma membrane and released adenosine triphosphatase (ATPase) preparations of Streptococcus lactis subsp. cremoris HA was studied by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). The membrane preparation gave two spots with periodic acid Schiff's (PAS) staining. The reference LTA revealed one CBB stainable band with MW 21 500, and was detected as a characteristic yellow spot in the molecular weight area 14 000–20 900 with silver staining and was also stainable with PAS. A slight colour was found with PAS in the F1-ATPase (EC 3.6.1.3) preparation. The results suggest that the carbohydrate component in the membrane preparation and in the solubilized F1-ATPase is LTA. 相似文献
Toll-like receptor 9 (TLR9) recognizes microbial DNA. We show here that TLR9 protein is expressed in human breast cancer cells and clinical breast cancer samples. Stimulation of TLR9-expressing breast cancer cells with the TLR9 agonistic CpG oligonucleotides (1-10 mumol/L) dramatically increased their in vitro invasion in both Matrigel assays and three-dimensional collagen cultures. Similar effects on invasion were seen in TLR9-expressing astrocytoma and glioblastoma cells and in the immortalized human breast epithelial cell line MCF-10A. This effect was not, however, dependent on the CpG content of the TLR9 ligands because the non-CpG oligonucleotides induced invasion of TLR9-expressing cells. CpG or non-CpG oligonucleotide-induced invasion in MDA-MB-231 cells was blunted by chloroquine and they did not induce invasion of TLR9(-) breast cancer cells. Treatment of MDA-MB-231 cells with CpG or non-CpG oligonucleotides induced the formation of approximately 50-kDa gelatinolytic band in zymograms. This band and the increased invasion were abolished by a matrix metalloproteinase (MMP) inhibitor GM6001 but not by a serine proteinase inhibitor aprotinin. Furthermore, CpG oligonucleotide treatment decreased tissue inhibitor of metalloproteinase-3 expression and increased levels of active MMP-13 in TLR9-expressing but not TLR9(-) breast cancer cells without affecting MMP-8. Neutralizing anti-MMP-13 antibodies inhibited the CpG oligonucleotide-induced invasion. These findings suggest that infections may promote cancer progression through a novel TLR9-mediated mechanism. An indicator of nutrient use efficiency through the entire food chain has been lacking. This article proposes a nutrient footprint method to estimate the efficiency of using both nitrogen (N) and phosphorus (P) in animal production chains following Life Cycle Assessment (LCA).
Methods
Following the nutrient footprint method of Grönman et al. (2016), we applied the nutrient footprint method to the Finnish beef production and consumption chain. We defined N and P flows associated with the beef chain from a product-specific point of view. The nutrient footprint is a resource efficiency indicator which combines the amount of nutrients captured for use in the production and consumption chain and their nutrient use efficiency (NutUE) either in the primary product or in both the primary + secondary products.
Results and discussion
Each 1000 kg of Finnish beef consumed requires 1700 kg N and 189 kg P during its life cycle. The percentage of virgin nutrient is more than 50% for N, but only 25% for P. NutUE in the primary product and in both primary + secondary products for N is 1% and 47% and for P is 0.2% and 74%, respectively.
Conclusions
The nutrient footprint offers information about NutUE in a simple and comparable form. In transition towards systems with sustainable nutrient use, it is essential to identify hot spots of nutrient leakage to be able to close them and improve food chains.
The inhalation of asbestos fibers is considered to be highly harmful, and lead to fibrotic and/or malignant disease. Epithelial-to-mesenchymal transition (EMT) is a common pathogenic mechanism in asbestos associated fibrotic (asbestosis) and malignant lung diseases. The characterization of molecular pathways contributing to EMT may provide new possibilities for prognostic and therapeutic applications. The role of asbestos as an inducer of EMT has not been previously characterized. We exposed cultured human lung epithelial cells to crocidolite asbestos and analyzed alterations in the expression of epithelial and mesenchymal marker proteins and cell morphology. Asbestos was found to induce downregulation of E-cadherin protein levels in A549 lung carcinoma cells in 2-dimensional (2D) and 3D cultures. Similar findings were made in primary small airway epithelial cells cultured in 3D conditions where the cells retained alveolar type II cell phenotype. A549 cells also exhibited loss of cell-cell contacts, actin reorganization and expression of α-smooth muscle actin (α-SMA) in 2D cultures. These phenotypic changes were not associated with increased transforming growth factor (TGF)-β signaling activity. MAPK/Erk signaling pathway was found to mediate asbestos-induced downregulation of E-cadherin and alterations in cell morphology. Our results suggest that asbestos can induce epithelial plasticity, which can be interfered by blocking the MAPK/Erk kinase activity. 相似文献
Human leukocyte antigen (HLA) genes, located on chromosome 6p21.3, have a crucial role in susceptibility to various autoimmune
and inflammatory diseases, such as celiac disease and type 1 diabetes. Certain HLA heterodimers, namely DQ2 (encoded by the
DQA1*05 and DQB1*02 alleles) and DQ8 (DQA1*03 and DQB1*0302), are necessary for the development of celiac disease. Traditional
genotyping of HLA genes is laborious, time-consuming, and expensive. A novel HLA-genotyping method, using six HLA-tagging
single-nucleotide polymorphisms (SNPs) and suitable for high-throughput approaches, was described recently. Our aim was to
validate this method in the Finnish, Hungarian, and Italian populations. The six previously reported HLA-tagging SNPs were
genotyped in patients with celiac disease and in healthy individuals from Finland, Hungary, and two distinct regions of Italy.
The potential of this method was evaluated in analyzing how well the tag SNP results correlate with the HLA genotypes previously
determined using traditional HLA-typing methods. Using the tagging SNP method, it is possible to determine the celiac disease
risk haplotypes accurately in Finnish, Hungarian, and Italian populations, with specificity and sensitivity ranging from 95%
to 100%. In addition, it predicts homozygosity and heterozygosity for a risk haplotype, allowing studies on genotypic risk
effects. The method is transferable between populations and therefore suited for large-scale research studies and screening
of celiac disease among high-risk individuals or at the population level.
