We have examined the effects of a variety of chemical modifications to recombinant human interleukin-2 (rIL-2) on its pharmacokinetic behavior in rats. Unmodified rIL-2 is cleared from plasma with half-lives of 3 and 44 min for the alpha and beta phases. Modification of rIL-2 with monomethoxy polyethylene glycol or polyoxyethylated glycerol increased the half-lives as much as 20-fold, although the volume of distribution remained unchanged at 88 +/- 13 ml/kg. The clearance rates correlated with the effective molecular size of the modified protein determined by size exclusion chromatography. Clearance decreased rapidly as the effective molecular size increased from 19.5 to 70 kDa, whereas above 70 kDa the clearance decreased more slowly. This abrupt change at 70 kDa may be related to the permeability threshold of the kidney glomerular membrane which retains proteins larger than albumin in the plasma. Using the relationship between clearance and effective molecular size, the clearance rates of mixtures of modified rIL-2 could be predicted based on their average effective molecular size. Since the effectiveness of rIL-2 therapy is likely to be related to its pharmacokinetic behavior, the ability to design a molecule with a predictable time course in plasma provides a means to study this relationship.  相似文献

The aim of the study was to investigate the relationships between specific anthropometric (9 skinfolds, 13 girths, 8 lengths and 8 breadths), body composition (body fat %, fat free mass [FFM], fat mass [FM]) parameters and bone mineral parameters (bone mineral density [BMD], bone mineral content [BMC) in young rhythmic gymnasts and same age controls. Eighty nine 7-8-year-old girls participated in this study and were divided to the rhythmic gymnast's (n = 46) and control (n = 43) groups. Body composition was determined by dual energy X-ray absorptiometry (FFM, FM, body fat %, BMD and BMC). Body fat % and FM were lower and BMD and BMC values at lumbar spine (L2-L4) and femoral neck were higher in rhythmic gymnasts compared with controls. All measured skinfold thicknesses were thicker in controls. In girths, lengths and widths there were only few significant differences between the groups. Stepwise multiple regression analysis indicated that skinfold thicknesses (supraspinale and medial calf) influenced L2-L4 BMD only in controls 38.2% (R2x100). Supraspinale and iliac crest skinfold thicknesses characterised L2-L4 BMC 43.9% (R2x100). Calf girths influenced BMD in L2-L4 52.3% (R2x100) in controls. BMC in L2-L4 was dependent only on mid-thigh girths 35.9% (R2x100). BMD in L2-L4 was dependent on tibiale-laterale height 30.0% (R2x100). Biiliocristal breadths together with sitting height characterised BMC in L2-L4 BMD 62.3% (R2x100). In conclusion, we found that the relationships between anthropometry, body composition and bone parameters in young rhythmic gymnasts are weak. In control group first of all lower body anthropometric parameters significantly correlated with BMD and BMC in spine.  相似文献

Background

The genetics of osteogenesis imperfecta (OI) have not been studied in a Vietnamese population before. We performed mutational analysis of the COL1A1 and COL1A2 genes in 91 unrelated OI patients of Vietnamese origin. We then systematically characterized the mutation profiles of these two genes which are most commonly related to OI.

Methods

Genomic DNA was extracted from EDTA-preserved blood according to standard high-salt extraction methods. Sequence analysis and pathogenic variant identification was performed with Mutation Surveyor DNA variant analysis software. Prediction of the pathogenicity of mutations was conducted using Alamut Visual software. The presence of variants was checked against Dalgleish’s osteogenesis imperfecta mutation database.

Results

The sample consisted of 91 unrelated osteogenesis imperfecta patients. We identified 54 patients with COL1A1/2 pathogenic variants; 33 with COL1A1 and 21 with COL1A2. Two patients had multiple pathogenic variants. Seventeen novel COL1A1 and 10 novel COL1A2 variants were identified. The majority of identified COL1A1/2 pathogenic variants occurred in a glycine substitution (36/56, 64.3 %), usually serine (23/36, 63.9 %). We found two pathogenic variants of the COL1A1 gene c.2461G?>?A (p.Gly821Ser) in four unrelated patients and one, c.2005G?>?A (p.Ala669Thr), in two unrelated patients.

Conclusion

Our data showed a lower number of collagen OI pathogenic variants in Vietnamese patients compared to reported rates for Asian populations. The OI mutational profile of the Vietnamese population is unique and related to the presence of a high number of recessive mutations in non-collagenous OI genes. Further analysis of OI patients negative for collagen mutations, is required.

The aim of our longitudinal study was to investigate the relationships between physical activity and bone mass in boys with different body mass status during the years surrounding pubertal growth spurt. Two hundred and six boys entering puberty took part in this study. The subjects were divided into underweight (), normal weight (), overweight () and obese () groups at baseline according to age related categories. Whole-body DXA scans were performed at baseline, after 12 and 24 months to assess body composition (lean body mass, fat mass), and total body (TB), lumbar spine (LS) and femoral neck (FN) bone mineral density (BMD) parameters. Physical activity was measured by 7- day accelerometry. For longitudinal analysis, multilevel fixed effects regression models were constructed. Biological age, height and lean body mass had an effect for explanation of TB BMD, FN BMD and LS BMD. Moderate to vigorous physical activity (MVPA), vigorous physical activity (VPA) and sedentary time (SED) had the significant effect only on FN BMD. Being an underweight boy at the baseline indicated greater chance (p<0.01) to have lower TB BMD in the future (2 years at follow up) development, compared to normal weight (estimates = −0.038), overweight (estimates = −0.061) and obese boys (estimates = −0.106).  相似文献

The uncoupler of mitochondrial oxidative phosphorylation, 2-nitro-4-azido-carbonylcyanide phenylhydrazone (N3CCP) which is capable of photoaffinity labeling has been used to examine the effect of uncouplers on the energy conserving membrane of Paracoccus denitrificans and Tetrahymena pyriformis. The N3CCP uncouples respiration in P. denitrificans and T. pyriformis cells with U1/2 values of 1.05 microM and 0.24 microM, respectively. Binding studies show the presence of 0.65 +/- 0.05 high affinity sites per cytochrome alpha with Kd of 0.5 +/- 0.1 microM in P. denitrificans membranes and 1.4 +/- 0.2 sites per cytochrome alpha 2 with a Kd of 0.4 +/- 0.1 microM in T. pyriformis membranes. Irradiation of [3H]-N3CCP bound to the membranes leads to a covalent linking of the radioactive uncoupler to a peptide of 10--15 kdaltons as analyzed by SDS-polyacrylamide gel electrophoresis. It is concluded that these two microbial systems contain a specific high affinity uncoupler binding site very similar to that of mammalian mitochondria (Katre, N.V. and Wilson, D.F. (1978) Arch. We located a heavy metal label, mercurilated phenylglyoxal, in both the primary sequence and in the tertiary structure of bacteriorhodopsin. This label modified arginines 225 and 227, which are on the COOH-terminal helix (G). In the projected electron potential difference map, the major site is close to the central inner helix. From this result we conclude that helix 1 could not be the COOH-terminal helix G. We tested the multiple isomorphous replacement method for obtaining phases for purple membrane by electron diffraction.  相似文献

2-Nitro-4-azidocarbonylcyanide phenylhydrazone (N3CCP), a potent water-soluble uncoupler at pH 6–8, was used to determine the nature of binding of the uncoupler to the mitochondrial membrane. Equilibrium binding studies with N3CCP showed that isolated pigeon heart mitochondria contain 1.6 ± 0.3 high-affinity binding sites per cytochrome a. Several different types of chemical uncouplers were also found to bind to the same high-affinity site as evidenced by their observed competition with N3CCP. The potassium ionophore valinomycin and the respiratory inhibitor antimycin A did not affect uncoupler binding to the high-affinity sites nor did active respiration of the mitochondria. The number of high-affinity binding sites was essentially unchanged by extraction of 80% of the mitochondrial phospholipids. The ability of the uncouplers to bind to the high-affinity binding sites is proportional to the uncoupler activities. These data support the idea that the high-affinity binding sites of mitochondria are protein(s) which are involved in the coupling reactions of oxidative phosphorylation and that uncoupler bound at these sites is responsible for the uncoupling activity.  相似文献

Elvers of Anguilla nebulosa were reared at 23°C in confined waters and their food intake, growth and conversion efficiency (K1:%) were studied. Elvers weighing 278.33±14.35 mg were fed on an ad libitum diet of the oligochaete worm Tubifex tubifex for 60 days. On an average, the test individuals consumed 99.55±19.81 mg food/elver/day amounting to 35.76% of the initial biomass; during the corresponding period the elvers exhibited a growth rate of 5.03±2.42 mg gain in weight/elver/day which is equivalent to 5.06% of the consumed food. Thus the 'housekeeping' of these elvers may be regarded as established. In 50 days, the food required amounts to 0.358±50 = 17.90 times the initially stocked biomass. That is, 1 kg of initially stocked elvers may need 17.90 kg of T. tubifex to produce 0.912 kg of elver meat. A poor conversion ratio of 20 : 1 may be one reason for the slow growth of the elvers.  相似文献
