Elucidation of the antigenic determinant in lipotropin for two monoclonal antibodies

Two monoclonal antibodies were found to give enhanced affinity for β-lipotropin when mixed, as evidenced by competitive radioimmunoassay. Both monoclonals were found to react with a pentapeptide Ala-Glu-Leu-Glu-Tyr, which is a sequence of high local hydrophilicity within the N-terminal section of β-lipotropin.     

Survival of micro-organisms in space

Dried suspensions ofPenicillium roqueforti Thom, Coliphage T-1,Bacillus subtilis and tobacco mosaic virus were exposed to space on board the Gemini-IX-A and XII earth satellites and the Agena-VIII space rocket. All micro-organisms tested survived the direct exposure during the Gemini-IX-A experiment. In the Gemini-XII experiment only the T-1 phage survived the direct exposure. The survival was influenced by the suspending medium and depended on the species of the microorganism. After four months of space flight on the Agena-VIII space rocket surviving fractions between 2×10^–3 and 1.0 were found in the unopened flight container. However, micro-organisms exposed on the cover of the container during this period were completely inactivated. Shielding against solar ultraviolet radiation during flight resulted in survival of micro-organisms exceeding to that of the transport controls, and the survival was considered complete.Sterile methylcellulose collection surfaces were exposed to space on board the Gemini-IX-A and XII satellites in an attempt to collect viable micro-organisms in space. None of the collection surfaces yielded viable micro-organisms.     

Die Ausdrucksbewegungen der Sichelente,Anas falcata L.

Zusammenfassung Die Ausdrucksbewegungen der Sichelente,Anas (Eunetta) falcata Georgi, werden beschrieben, soweit möglich vorläufig analysiert und mit denen nächstverwandter Arten, vor allem denen vonAnas (Nettium) crecca crecca L.,Anas (Chaulelasmus) strepera L. undAnas (Mareca) penelope L. verglichen. Dieser Vergleich ergibt, genau wie der morphologischer Merkmale des Gefieders und der Knochentrommel, eine systematische Stellung vonfalcata genau zwischen den drei genannten Arten, näher den beiden erstgenannten als der dritten. Die Verteilung der Merkmal-Gemeinsamkeiten einerseits mitcrecca, andererseits mitstrepera, läßt den Schluß zu, daß die drei Arten divergent aus einer gemeinsamen Ahnenform entstanden seien. Keine gemeinsamen Merkmale, die bei anderen Arten fehlen, verbinden die drei genannten Formen zu einer Gruppe.Beim Gesellschaftsspiel der Sichelerpel sind, im Gegensatz zu dem aller anderen bisher daraufhin untersuchten Schwimmentenarten, alle beteiligten Bewegungsweisen, einschließlich des einleitenden Schüttelns, zur Ente hin orientiert, die hier, wie beicrecca undstrepera, am Spiel der Erpel sehr regen Anteil nimmt. Dagegen fehlen solche Bewegungen, die durch Ritualisierung aus Angriffsverhalten entstanden sind, beim Gesellschaftsspiel völlig, spielen aber eine große Rolle, analog dem Triumphgeschrei der Gänse und Tadorninen, beim Zusammenhalt der bereits verpaarten Tiere.     

Some effects of air ions on the activity of rats

A duct system was arranged through which flowed air of controlled ion concentration. In series was a chamber with a flexible dielectric floor which made possible electrical detection of activity.Exposure to various ion concentrations of either polarity produced pronounced effects.In addition to gross motor activity six other behavioural parameters were recorded. The lowest ion concentration were most effective. In addition to presenting a controlled ambient ion concertration the ion current drawn by the animal was measured. This varied markedly from rat to rat.

Zusammenfassung Ein Kanalsystem zum Durchfluss von Luft bekannter Ionenkonzentration wurde eingerichtet. Angeschlossen war eine Kammer mit einem beweglichen dielektrischen Boden, der die Registrierung der Aktivität von Ratten ermöglichte. Neben der motorischen Aktivität wurden 6 andere Verhaltensformen der Tiere registriert.Die niedrigste Ionenkonzentration war am wirksamsten zur Anregung der Aktivität. Neben der Ionenkonzentration in der Kammer wurde der von den Tieren eingeatmete Ionenstrom gemessen; er variierte stark von Tier zu Tier.

Resume On a construit un canal dans lequel il est possible de faire circuler de l'air dont la concentration en ions est contrôlable. On y a adjoint une cellule dont le sol est constitué par un diélectrique flexible permettant de déceler électriquement l'activité de petits animaux.En exposant des rats à des concentrations ioniques variables et de polarité différente, on peut constater des effets prononcés sur le comportement des animaux. En effet, en plus de l'activité motrice, on a enregistré six autres formes de ce comportement. Ce sont les plus faibles concentrations qui produisent le plus d'effets. A part la concentration en ions à l'intérieur de la cellule, on a également mesuré la quantité d'ions absorbés par les bêtes par voie respiratoire. Cette quantité varie fortement d'un individu à l'autre.

Induction of accessory cell function of human alveolar macrophages by inhalation of human natural interleukin-2

 Accessory function allows antigen-presenting cells to produce sufficient secondary signals for optimum T cell proliferation and interleukin-2 (IL-2) production. Alveolar macrophages are inferior accessory cells compared to monocytes (PBM). We report here that the accessory index (AI) of alveolar macrophages and PBM of patients with lung metastases of solid tumors treated with inhalations of human natural IL-2 (hnIL-2) increased following its administration (P<0.005). The accessory index was significantly elevated from baseline values after 2 weeks of inhalation of 300 000 IU hnIL-2/day (8.2±10.2 compared to 1.1±1; P<0.001). The inhalation of 150 000 IU also induced increases in the index (AI = 2.3±1.9), however, without reaching statistical significance. In addition at 300 000 IU IL-2/day a significant increase in the accessory index was observed for PBM (4±2.5; P<0.05). The indices of PBM and alveolar macrophages prior to inhalation showed a significant negative correlation with the age of the patients (r _s =  – 0.5; r _s =  – 0.8, respectively; P<0.03 for all comparisons). Our data demonstrate that the inhalational application of hnIL-2 enhances the accessory function of alveolar macrophages and, to lesser extent, the accessory index of PBM, indicating the occurrence of pharmacological immunostimulation. The nervous system of Tricladida. III. Neuroanatomy ofDendrocoelum lacteum andPolycelis tenuis (Plathelminthes,Paludicola): an immunocytochemical study

The organization of the nervous system ofDendrocoelum lacteum (Tricladida, Paludicola, Dendrocelidae) andPolycelis tenuis (Tricladida, Paludicola, Planariidae) was studied by immunocytochemical double staining, using neuropeptide RFamide and serotonin (5-HT) antisera on cryosections. The study confirmed the status of the main nerve cords (MCs) as the most important and stable of the longitudinal cords and supported the hypothesis of a common phylogenetic origin of the MCs in flatworms. The ganglion-like structures along the MCs at the beginning of transverse commissures and laterla branches showed a close contact with ventral fibres of the submuscular nerve plexus indicating an origin from crossing points of insunken ring commissures. The distributional pattern and morphology of the RFamide-IR cell bodies inD. lacteum corresponded to that of neurosecretory cells. Most RFamide-IR cells were unipolar and rounded while 5-HT-IR cells were uni- bi- and multipolar. The neutropile consisted of a dense RFamide-IR and a loose 5-HT-IR network. RFamide dominated in all parts of the genital plexus.     

Significance of Ca2+ and K+ in Micrasterias Growth and Morphogenesis

Significance of Ca²⁺ and K⁺ for the complex morphogenesis ofMicrasterias, which takes place through multipolar tip growth,was investigated. Studies with different external Ca²⁺ concentrationsand Ca²⁺ channel inhibitors LaCl₃ and verapamil indicate thatCa²⁺ and Ca²⁺ channels are essential in the development, whiletreatments with different K⁺ concentrations and K⁺ channel inhibitorTEA demonstrate that potassium or K⁺ channels are not neededin the process, albeit the existence of K⁺ channels. K⁺ is notneeded even for the regulation of turgor pressure, which wasfound to decrease clearly during cell development. The plasmamembrane ATPase inhibitors diethylstilbesterol (DES) and Na-orthovanadatestop morphogenesis and indicate the importance of ion pumpsin the developmental process. Both supraoptimal, external K⁺and Ca²⁺ cause abundant Ca²⁺ precipitate formation in chloroplasts,which shows that chloroplasts are important in regulation ofcytoplasmic Ca²⁺ metabolism and that K⁺ activates the uptakeof Ca²⁺ through Ca²⁺ channels. Rapid reactions of spruce cells to elicitors released from the ectomycorrhizal fungus Hebeloma crustuliniforme, and inactivation of these elicitors by extracellular spruce cell enzymes

Elicitors released from hyphae or cell walls of the ectomycorrhizal fungus Hebeloma crustuliniforme (Bull. ex Fries.) Quél. induced in suspension-cultured cells of Picea abies (L.) Karst. a set of fast reactions: (i) an immediate efflux of Cl^– into the medium, followed by a K⁺ efflux; (ii) an influx of Ca²⁺ (measured as accumulation of ⁴⁵Ca²⁺ in the cells); (iii) a phosphorylation of a 63-kDa protein and dephosphorylation of a 65-kDa protein (detectable by 4 min after elicitor application); (iv) an alkalinization of the medium, and (v) a transient synthesis of H₂O₂. The removal of extracellular Ca²⁺ by EGTA delayed the elicitor-induced alkalinization. A further reduction of this response could be achieved by TMB-8 an inhibitor of Ca²⁺ release from intracellular stores. Moreover, the inhibition of protein kinase activity by staurosporine prevented the extracellular alkalinization completely. However, the effectiveness of the elicitors in inducing the extracellular alkalinization was strongly impaired by constitutively secreted enzymes of spruce cells which cleaved the elicitors to inactive fragments. It is suggested that in ectomycorrhizae the efficacy of elicitors released from fungal cell walls is controlled by apoplastic enzymes of the host; the plant itself is able to reduce the activity of fungal elicitors on their way through the plant cell wall. Stimulus enhancement and spread of a spontaneous tool use in a colony of long-tailed macaques

In a captive group of long-tailed macaques, tool-using behavior by a single competent individual had a significant effect on the synchronous manipulative behavior of naïve animals. Group members engaged in manipulations on the same object class more frequently during times when the model was working than when it was not. The form of their behavior, however, in no way resembled the technique used by the model. All three animals that later became successful tool users were among the few subjects that exhibited a significant increase in manipulations on the same object class while the model was working. Possible causal relationships between this stimulus enhancement and the transmission of the new behavior to other group members are analyzed and discussed.     

Lck-dependent tyrosyl phosphorylation of the phosphotyrosine phosphatase SH-PTP1 in murine T cells.

The phosphorylation and dephosphorylation of proteins on tyrosyl residues are key regulatory mechanisms in T-cell signal transduction and are controlled by the opposing activities of protein tyrosine kinases and phosphotyrosyl phosphatases (PTPs). In T cells, several nontransmembrane protein tyrosine kinases are associated with receptors; for example, Lck is bound to the coreceptors CD4 and CD8 and becomes activated upon their stimulation. In comparison, little is known about the role of nontransmembrane PTPs in early T-cell signaling. SH-PTP1 (PTP1C, HCP, SHP) is a nontransmembrane PTP expressed primarily in hematopoietic cells, including T cells. We have found that SH-PTP1 is basally phosphorylated on serine in resting T cells. Upon stimulation of CD4 or CD8 either in a T-cell hybridoma cell line or in primary thymocytes, SH-PTP1 becomes tyrosyl phosphorylated. Moreover, SH-PTP1 is constitutively phosphorylated on tyrosine in the Lck-overexpressing lymphoma cell line LSTRA. SH-PTP1 is also a good substrate for recombinant Lck in vitro. Comparisons of the tryptic phosphopeptide maps of wild-type SH-PTP1 and deletion and point mutations establish that the two sites (Y-536 and Y-564) which are directly phosphorylated by Lck in vitro are also phosphorylated in vivo in LSTRA cells. One of these sites (Y-564) is phosphorylated in T cells in response to Lck activation. We conclude that SH-PTP1 undergoes Lck-dependent tyrosyl phosphorylation in T cells and likely plays a role in early T-cell signaling.
