Summary Spontaneous chloramphenicol (cap r)- and erythromycin (ery r)-resistant mutants were isolated from strain ade7–50 h - and the antimycin-resistant mutant ana r-8 ade 7–50 h- of Schizosaccharomyces pombe (Sch. p.). By mitotic segregation analysis all 154 cap r- and 120 ery r-mutants derived from ade 7–50 h - proved to be recessive chromosomal, whereas all 108 cap r- and 200 ery r-mutants originating from ana r-8 were extrachromosomally inherited. The rate of spontaneous cap r- and ery r-mutants was about hundredfold in ana r-8 compared to ade 7–50 h -. Growth of cap r-and ery r-mutants was not inhibited by chloramphenicol or erythromycin, respectively, in glucose-medium and only slightly in glycerol-medium at concentrations which completely inhibited ana r-8. By mitotic segregation-, tetrad-, and mitotic haploidization-analysis the extrachromosomal inheritance of mutants derived from ana r-8 was established. Segregational patterns of cap r- and ery r-determinats during mitosis, meiosis, and mitotic haplidization of diploids are discussed.  相似文献

Enzyme-bound pentadienyl and peroxyl radicals in purple lipoxygenase   总被引:1,自引:0,他引:1

Samples of purple lipoxygenase prepared by addition of either 13-hydroperoxy-9,11-octadecadienoic acid or linoleic acid and oxygen to ferric lipoxygenase contain pentadienyl and/or peroxyl radicals. The radicals are identified by the g values and hyperfine splitting parameters of natural abundance and isotopically enriched samples. The line shapes of their EPR spectra suggest the radicals are conformationally constrained when compared to spectra of the same radicals generated in frozen linoleic acid. Further, the EPR spectra are unusually difficult to saturate. The radicals are stable in buffered aqueous solution at 4 degrees C for several minutes. All of this implies that these species are bound to the enzyme, possibly in proximity to the iron. Only peroxyl radical is seen when the purple enzyme is generated with either hydroperoxide or linoleic acid in O2-saturated solutions. Addition of natural abundance hydroperoxide under 17O-enriched O2 leads to the 17O-enriched peroxyl radical, while the opposite labeling results in the natural abundance peroxyl radical, demonstrating the exchange of oxygen. Both radicals are detected in samples of purple lipoxygenase prepared with either linoleic acid or hydroperoxide under air. Addition of the hydroperoxide in the absence of oxygen favors the pentadienyl radical. We propose that addition of either linoleic acid or hydroperoxide to ferric lipoxygenase leads to multiple mechanistically connected enzyme complexes, including those with (hydro)peroxide, peroxide, peroxyl radical, pentadienyl radical, and linoleic acid bound. This hypothesis is essentially identical with the proposed radical mechanism of oxygenation of polyunsaturated fatty acids by lipoxygenase.  相似文献

The effect of increasing concentration of T3 (10 × 10?12 – 2000 × 10?6 M) on O2-consumption and [14C]-alanine conversion into [14C]-CO2 and [14C]-glucose was investigated in the isolated perfused liver of hypothyroid starved rats. T3 induced within 1 h an increase (i) in the oligomycine-sensitive O2-consumption (+ 50–85 %), (ii) in the [14C]-CO2-production (+55–102 %), and (iii) in the [14C]-glucose synthesis (+ 40?80 %). These effects were dose dependent and significant at a concentration as low as 10 × 10?12 M, reported to represent the free hormone concentration in rat serum under in vivo conditions. The results demonstrate a direct and rapid stimulatory action of T3 in the physiological range on hepatic energy metabolism and glucose production. The effects could not be explained by the thyroid hormone induced nuclear activity.  相似文献

Force exertion against different mechanical environments can affect motor control strategies in order to account for the altered environmental dynamics and to maintain the ability to produce force. Here, we investigated the change of muscular activity of selected muscles of the lower extremities while the participants interacted with an external mechanical device of variable stability. Twenty-five healthy participants exerted force against the device by performing a unilateral ballistic leg extension task under 1 or 3 degrees of freedom (DoF). Directional force data and electromyographic responses from four leg muscles (TA, VM, GM, PL) were recorded. Muscle responses to the altered experimental conditions were analyzed by calculating time to peak electrical activity (TTP), peak electrical activity (PEA), slope of EMG-signal and muscle activity. It was found that neuromuscular system adjustments to the task are expressed mainly by temporal (TTP) rather than amplitude (PEA) scaling of muscular activity. This change was specific for the investigated muscles. Moreover, a selective increase of muscle activity occurred while increasing external DoF. This scheme was accompanied by a significant reduction of applicable force against the device in the unstable 3 DoF condition. The findings suggest that orchestration of movement control is linked to environmental dynamics also affecting the ability to produce force under dynamic conditions. The adjustments of the neuromuscular system are rather temporal in nature being consistent with the impulse timing hypothesis of motor control.  相似文献
