Nerve cell bodies in the spiral and vestibular ganglia of the adult rat are surrounded by thin (about ten lamellae) myelin sheaths which differ in several respects from typical axonal myelin. In some instances lamellae surrounding perikarya appear as typical major dense lines, and in others as thin Schwann cell sheets in which cytoplasm persists. Discontinuities and irregularities appear in the structure of perikaryal myelin. Lamellae may terminate anywhere within the sheaths; they may bifurcate; they may reverse their direction; or they may merge with each other. The number of lamellae varies from one part of a sheath to another. In addition, the myelin of a single perikaryal sheath may receive contributions from more than one Schwann cell, which overlap and interleave with each other. The ganglion cells are of two types: those which are densely packed with the usual cytoplasmic organelles but have few neurofilaments (granular neurons), and those which exhibit large areas containing few organelles but have a high concentration of neurofilaments (filamented neurons). The latter cell type is ensheathed by myelin which is generally more compact that that surrounding the former. The formation and the physiologic significance of perikaryal myelin are discussed. 相似文献
Subsurface cisterns (SSC's) are large, flattened, membrane-limited vesicles which are very closely apposed to the inner aspect of the plasma membranes of nerve cell bodies and the proximal parts of their processes. They occur in a variety of vertebrate and invertebrate neurons of both the peripheral and central nervous systems, but not in the surrounding supporting cells. SSC's are sheet-like in configuration, having a luminal depth which may be less than 100 A and a breadth which may be as much as several microns. They are separated from the plasmalemma by a light zone of ~50 to 80 A which sometimes contains a faint intermediate line. Flattened, agranular cisterns resembling SSC's, but structurally distinct from both typical granular endoplasmic reticulum (ER) and from Golgi membranes, also occur deep in the cytoplasm of neurons. It is suggested that membranes which are closely apposed may interact, resulting in alterations in their respective properties. The patches of neuronal plasmalemma associated with subsurface cisterns may, therefore, have special properties because of this association, resulting in a non-uniform neuronal surface. The possible significance of SSC's in relation to neuronal electrophysiology and metabolism is discussed. 相似文献
Somatic muscle cells of Ascaris lumbricoides consist of three differently specialized components referred to as the fiber, which contains the contractile apparatus (described previously), the belly, and the arm. The belly is shown to be a sac of glycogen, which is depleted during starvation of the animal. The arm extends to a nerve cord where it establishes a myoneural junction characterized by giant mitochondria and clusters of vesicles in the nerve fibers and by a 500 A neuromuscular gap. The arms, which have been shown to be "electrically interconnected" in the vicinity of the nerve cord, form "tight junctions" with one another in just this region. At high magnification, these junctions can be resolved into several types. In some there is fusion of the outer leaflets of the membranes with formation of an intermediate line. Others resemble septate desmosomes in that a residual extracellular space ~20 A in width remains between the membranes, but the outer leaflets are interconnected across the gap. It is suggested that the term "tight junction" encompasses a variety of structures distinguishable only at high magnification and that the different variations are not necessarily equivalent functionally. 相似文献
This article is a detailed case study of a particular FMS that will be operational in 1989. It describes the daily planning and operating problems that will need to be addressed. The algorithms that will operate this system are presented. Given the daily changing production requirements, the algorithms begin with an aggregate planning feasibility check. Then planning, scheduling, inventory management, and breakdowns are addressed. The key problems in operating this system are tool management problems. Recent studies have demonstrated that, under certain circumstances, the diaphragm does not contract as a homogeneous unit. These observations suggest that motor units may not be randomly distributed throughout the muscle but confined to localized subvolumes. In the present study, electromyographic (EMG) and glycogen depletion methods were combined to investigate the organization of motor units supplied by the primary branches of the phrenic nerve in the cat. Four primary branches are generally present, one branch to the crus and three branches to the sternocostal region. The gross motor-unit territory of each of the four phrenic primary branches was determined by stimulating each nerve separately, while recording from nine EMG electrodes distributed over the hemidiaphragm. Stimulation of the crural branch evoked activity in the ipsilateral crus, whereas stimulation of each of the remaining branches evoked activity in discrete but overlapping areas of the sternocostal diaphragm. A more precise analysis of the distribution and borders of the motor territories was obtained by mapping regions depleted of muscle glycogen due to stimulation of each primary branch for 90 min. Glycogen depletion results closely matched the EMG findings of a localized distribution of motor units served by single primary branches. Stimulation of the crural branch typically caused depletion of the ipsilateral crus, whereas the sternocostal branches each served a striplike compartment. In the majority of cases, the borders of the sternocostal compartments were relatively abrupt and consisted of a 1- to 2-mm transition zone of depleted and nondepleted fibers. These studies demonstrate that motor unit territories of the primary branches of the phrenic nerve are highly delineated. This compartmentalization provides the central nervous system with the potential for a more precise regional motor control of costal and crural diaphragm than previously suspected. 相似文献
1. 1.|The mitochondrial population in hypothalamic and hypophysial brain tissue from warm (30°C) and cold (5°C) acclimated goldfish (Carassius auralus L.) was analyzed using sterological techniques.
2. 2.|It was revealed that there is a significantly larger volume density (Vv) in the cold acclimated tissue, with no significant difference in either of the surface densities (Svext and Svint) from either of the brain areas.
3. 3.|The hypothalamic brain tissue has a significantly lower specific surface (S/V) in the cold acclimated tissue but there is not a significant difference in this parameter for the hypophysial brain tissue.
4. 4.|The values for these three parameters (Vv, Svext and SVint, and S/V) indicate that mitochondria from acclimated brain tissue undergo shape changes in response to thermal stress.
5. 5.|We suggest that the shape changes may be related to the change in the phospholipid composition of the inner mitochondrial membrane with acclimation temperature.
Summary The immunogenicity of the disialoganglioside, GD3, a melanoma-tumor-associated antigen, has been evaluated in non-human primates. Sera from four chimpanzees and two monkeys were evaluated for anti-GD3 antibody activity by solid-phase radioimmunoassay using GD3 and control gangliosides as targets. Serum from one monkey, immunized with cells from a melanoma cell line, was strongly reactive with GD3, having a titer of >2500. In contrast, serum from this animal was non-reactive with several other gangliosides including the structurally similar GM3. Anti-GD3 reactivity was also demonstrable, albeit in low titer, in the sera of an additional monkey and a chimpanzee. Each of these animals had likewise been immunized using cells from melanoma cell lines. On the basis of these observations, suggestive of a primate anti-GD3 antibody response, we initiated a series of immunizations of chimpanzee using purified GD3 bound to Salmonella minnesota, R595. IgG reactive with melanoma cells in the cell-binding assay was first detected in sera collected after 4 immunizations and increased in titer against each reactive melanoma cell line during the immunizations. Reactivity of this serum with melanoma cell lines demonstrated a direct correlation with the expression of GD3 by the respective cell line. Anti-GD3 reactivity was evident in solid-phase radioimmunoassay against purified GD3 beginning with serum collected after 11 immunizations. By comparison with its binding to the control ganglioside panel, this serum demonstrated strong specificity for GD3 (titer=640) while having only marginal reactivity with GM3 (titer=40). Immune serum from this animal was also able specifically to block subsequent binding of a murine IgM anti-GD3 antibody (DMab7) to target GD3 in solid-phase radioimmunoassay.
