Summary The major organelles within the cells of maleBangia atropurpurea (Roth) C. Ag. filaments undergo a series of ultrastructural transformations during the production of spermatia. Initially, thylakoids within the large axial chloroplast develop a reticulate pattern commencing at the central pyrenoid region. Subsequent changes involve loss of lobes and diminution of volume through division; chloroplasts in final stages contain a few dilated, distorted thylakoids and many plastoglobuli. During differentiation the large nucleolus disappears from the nucleus and four masses of chromatin aggregate near the nuclear envelope. Furrows originating from the nuclear envelope form double membranes around each of the chromatin masses and most of the nucleoplasm is eliminated. Several types of fibrillar vesicles are formed during the process and large floridean starch reserves are utilized. Multilamellar bodies and microbody-like structures occur within the cells during certain phases of spermatiogenesis.  相似文献

Slow fluorescence transients in Chlamydomonas reinhardi arise after transitions from high light intensities to low light or dark conditions. Characteristics of the newly described transient phenomena include: (a) A slow biphasic decrease in fluorescence yield occurs in the dark, followed by an even slower, hour long, increase in fluorescence. (b) A similar, but faster, fluorescence yield decrease and subsequent increase also occurs during low intensity illumination periods separating high light intervals, or after transitions from high intensity to low intensity light. (c) Short (several seconds) flashes of light given during a dark period have no effect on the dark fluorescence decay, regardless of the flash frequency. Such flash regimes accurately monitor the dark decline of the M2 level by tracing the parallel decay of flash-generated P2 (Kautsky) peaks. However, flashes during a low light illumination period do influence the decay kinetics. Frequent flashes allow decay similar to that occurring in dark, but less frequent flashes inhibit the decrease in fluorescence yield.  相似文献

From studies of the oxygenation rate of red blood cells (RBC) using rapid-mix techniques, it has been suggested that RBC are surrounded by a stagnant layer of water that does not (or cannot) mix with the rest of the water. A consideration of the appropriate hydrodynamics and convective diffusion rates shows that a mixer can reduce the resolution time to approximately 1 ms (or possibly less) and give a diffusion layer around the TBC that is approximately 1 micron thick. In stopped flow equipment it expands to approximately 4 micron over approximately 10 ms, whereas in continuous flow work the diffusion layers expands slightly less rapidly and less far. Thus the rate of oxygenation of TBC should be slower when measured by stopped flow techniques than by continuous flow apparatus for which the rate will depend weakly on the Reynolds number of the flow in the interrogation tube.  相似文献

Both muscular and respiratory biofeedback procedures have been employed in attempts to reduce symptoms of bronchial asthma. Research relating to these approaches is reviewed in the present article. Biofeedback training both for facial muscle relaxation and for respiratory resistance decrease improves short-term pulmonary function in asthmatic individuals. These forms of training represent promising avenues for the management of asthma. However, unqualified endorsement of these procedures is premature, at the present time, since their influence on asthma-related variables other than pulmonary function has not been determined and since their long-term effects have not been investigated.  相似文献

The four pectate lyases (EC 4.2.2.2) secreted by Erwinia chrysanthemi EC16 have been individually produced as recombinant enzymes in Escherichia coli. Oligogalacturonates formed from polygalacturonic acid during reactions catalyzed by each enzyme have been determined by high-performance liquid chromatography analysis. PLa catalyzes the formation of a series of oligomers ranging from dimer to dodecamer through a random endolytic depolarization mechanism. PLb and PLc are trimer- and tetramer-generating enzymes with an identical combination of endolytic and exolytic mechanisms. PLe catalyzes a nonrandom endolytic depolymerization with the formation of dimer as the predominant product. The pectate lyases secreted by E. chrysanthemi EC16 represent a battery of enzymes with three distinct approaches to the depolymerization of plant cell walls.  相似文献

Using 3-D searching techniques based on algorithms derived from graph theory we have established a striking structural similarity between the structure of bovine carboxypeptidase A and that of the C-terminal domain of bovine leucine aminopeptidase. There is no significant sequence homology between the aminopeptidases and the carboxypeptidases but the strong structural relationship detected in this complex fold suggests that there may be a very remote divergent evolutionary relationship between these two enzyme classes.  相似文献

Summary Chaos game representation (CGR) is a novel holistic approach that provides a visual image of a DNA sequence quite different from the traditional linear arrangement of nucleotides. Although it is known that CGR patterns depict base composition and sequentiality, the biological significance of the specific features of each pattern is not understood. To systematically examine these features, we have examined the coding sequences of 7 human globin genes and 29 relatively conserved alcohol dehydrogenase (Adh) genes from phylogenetically divergent species. The CGRs of human globin cDNAs were similar to one another and to the entire human globin gene complex. Interestingly, human globin CGRs were also strikingly similar to human Adh CGRs. Adh CGRs were similar for genes of the same or closely related species but were different for relatively conserved Adh genes from distantly related species. Dinucleotide frequencies may account for the self-similar pattern that is characteristic of vertebrate CGRs and the genome-specific features of CGR patterns. Mutational frequencies of dinucleotides may vary among genome types. The special features of CG dinucleotides of vertebrates represent such an example. The CGR patterns examined thus far suggest that the evolution of a gene and its coding sequence should not be examined in isolation. The continuous addition of toluene as a solute of treated ballast water from oil tankers into a well-defined estuary facilitated the study of the dynamics of dissolved hydrocarbon metabolism in seawater. Most rates of toluene oxidation were in the range of 1 to 30 pg/liter per h at 0.5 μg of toluene per liter. Near the ballast water injection point, a layer of warm ballast water, rich in bacteria, that was trapped below the less-dense fresh surface water was located. Toluene residence times were approximately 2 weeks in this layer, 2 years elsewhere in Port Valdez, and 2 decades in the surface water of a more oceanic receiving estuary adjacent. Mixing was adequate for a steady-state treatment which showed that 98% of the toluene was flushed from Port Valdez before metabolism and gave a steady-state concentration of 0.18 μg/liter. Total bacterial biomass from direct counts and organism size data was usually near 0.1 mg/liter, but ranged up to 0.8 mg/liter in the bacteria-rich layer. The origin of bacteria in this layer was traced to growth in oil tanker ballast during shipments. The biomass of toluene oxidizers in water samples was estimated from the average affinity of pure-culture isolates for toluene (28 liters per g of cells per h) and observed toluene oxidation kinetics. Values ranged from nearly all of the total bacterial biomass within the bacteria-rich layer down to 0.2% at points far removed. Because the population of toluene oxidizers was large with respect to the amount of toluene consumed and because water from a nearby nonpolluted estuary was equally active in facilitating toluene metabolism, we searched for an additional hydrocarbon source. It was found that terpenes could be washed from spruce trees by simulated rainfall, which suggested that riparian conifers provide an additional and significant hydrocarbon source to seawater.  相似文献
