Conifers produce terpenoid-rich oleoresin in specialized resin ducts as a main line of defence against pests and pathogens. In spruce species (Picea spp.), axial resin ducts are either present constitutively in the cortex tissue (cortical resin ducts, CRDs) or are formed de novo as traumatic resin ducts (TRDs) in the cambial zone upon attack by insects, fungi or treatment with methyl jasmonate (MeJA). Using immunofluorescence localization we tested if previously formed CRDs respond to MeJA treatment with increased capacity for diterpenoid biosynthesis. We also tested the dynamics of diterpene synthase localization in the cambial zone. Immunofluorescence localization was performed using an antibody against a diterpene synthase, levopimaradiene/abietadiene synthase (LAS), in stem cross-sections of untreated and 0.1% MeJA-treated 4-year old Sitka spruce (P. sitchensis) trees. No fluorescence signal was observed in untreated stem cross-sections; however, signal was present 2 days after treatment with MeJA exclusively in the epithelial cells of CRDs. Fluorescence steadily increased in the CRD epithelial cells 4 and 8 days after treatment. At 8 days, additional fluorescence was observed in developing epithelial cells of traumatic resin ducts TRDs in the cambial zone. These results confirm that resin duct epithelial cells are the main site of diterpene biosynthesis in Sitka spruce, diterpenoid biosynthesis is induced in CRD epithelial cells early upon treatment with MeJA, and immature developing TRD epithelial cells produce diterpene synthase enzyme. Overall, the results of this work improve our understanding of spatial and temporal patterns of induced diterpene resin acid biosynthesis in conifers.  相似文献

The C4 protein of beet curly top virus [BCTV-B (US:Log:76)] induces hyperplasia in infected phloem tissue and tumorigenic growths in transgenic plants. The protein offers an excellent model for studying cell cycle control, cell differentiation, and plant development. To investigate the role of the C4 protein in plant development, transgenic Arabidopsis thaliana plants were generated in which the C4 transgene was expressed under the control of an inducible promoter. A detailed analysis of the developmental changes that occur in cotyledons and hypocotyls of seedlings expressing the C4 transgene showed extensive cell division in all tissues types examined, radically altered tissue layer organization, and the absence of a clearly defined vascular system. Induced seedlings failed to develop true leaves, lateral roots, and shoot and root apical meristems, as well as vascular tissue. Specialized epidermis structures, such as stomata and root hairs, were either absent or developmentally impaired in seedlings that expressed C4 protein. Exogenous application of brassinosteroid and abscisic acid weakly rescued the C4-induced phenotype, while induced seedlings were hypersensitive to gibberellic acid and kinetin. These results indicate that ectopic expression of the BCTV C4 protein in A. thaliana drastically alters plant development, possibly through the disruption of multiple hormonal pathways.  相似文献

Background

Pneumonia and malaria, two of the leading causes of morbidity and mortality among children under five in Zambia, often have overlapping clinical manifestations. Zambia is piloting the use of artemether-lumefantrine (AL) by community health workers (CHWs) to treat uncomplicated malaria. Valid concerns about potential overuse of AL could be addressed by the use of malaria rapid diagnostics employed at the community level. Currently, CHWs in Zambia evaluate and treat children with suspected malaria in rural areas, but they refer children with suspected pneumonia to the nearest health facility. This study was designed to assess the effectiveness and feasibility of using CHWs to manage nonsevere pneumonia and uncomplicated malaria with the aid of rapid diagnostic tests (RDTs).

Methods and Findings

Community health posts staffed by CHWs were matched and randomly allocated to intervention and control arms. Children between the ages of 6 months and 5 years were managed according to the study protocol, as follows. Intervention CHWs performed RDTs, treated test-positive children with AL, and treated those with nonsevere pneumonia (increased respiratory rate) with amoxicillin. Control CHWs did not perform RDTs, treated all febrile children with AL, and referred those with signs of pneumonia to the health facility, as per Ministry of Health policy. The primary outcomes were the use of AL in children with fever and early and appropriate treatment with antibiotics for nonsevere pneumonia. A total of 3,125 children with fever and/or difficult/fast breathing were managed over a 12-month period. In the intervention arm, 27.5% (265/963) of children with fever received AL compared to 99.1% (2066/2084) of control children (risk ratio 0.23, 95% confidence interval 0.14–0.38). For children classified with nonsevere pneumonia, 68.2% (247/362) in the intervention arm and 13.3% (22/203) in the control arm received early and appropriate treatment (risk ratio 5.32, 95% confidence interval 2.19–8.94). There were two deaths in the intervention and one in the control arm.

Conclusions

The potential for CHWs to use RDTs, AL, and amoxicillin to manage both malaria and pneumonia at the community level is promising and might reduce overuse of AL, as well as provide early and appropriate treatment to children with nonsevere pneumonia.

Organ formation at shoot and flower meristems in plants requires the maintenance of a population of centrally located stem cells and the differentiation of peripherally located daughter cells. The CLAVATA (CLV) gene products in Arabidopsis, including the CLV1 receptor-kinase, regulate this process by promoting the differentiation of stem cells on the meristem flanks. Here, we have analyzed the developmental roles of the CLV1-related BAM1 (derived from barely any meristem 1), BAM2 and BAM3 receptor-like kinases. Loss-of-function alleles of these receptors lead to phenotypes consistent with the loss of stem cells at the shoot and flower meristem, suggesting that their developmental role is opposite to that of CLV1. These closely related receptors are further distinguished from CLV1, whose expression and function is highly specific, by having broad expression patterns and multiple developmental roles. These include a requirement for BAM1, BAM2 and BAM3 in the development of high-ordered vascular strands within the leaf and a correlated control of leaf shape, size and symmetry. In addition, BAM1, BAM2 and BAM3 are required for male gametophyte development, as well as ovule specification and function. Significantly, the differing roles of CLV1 and BAM receptors in meristem and organ development are largely driven by differences in expression patterns.  相似文献

Sardines (Sardinops spp.) occupy temperate upwelling zones in the coastal regions of the Indian and Pacific Oceans, including locations in Japan, California, Chile, Australia, and South Africa. East and West Pacific populations are separated by vast expanses of open ocean, and northern and southern hemisphere populations are separated by tropical waters which are lethal to sardines. The relative importance of these barriers has been the focus of a longstanding debate between vicariance and dispersal schools in biogeography. Comparisons of a 500 bp fragment of the mitochondrial (mt) DNA control region reveal strong geographic structuring of mtDNA lineages but shallow divergence both within and between regional populations. Regional populations are related to one another in a stepping-stone pattern, the apparent result of a series of Pleistocene dispersal events around the continental margins of the Indian-Pacific Basin. These mtDNA data, combined with an electrophoretic survey of variability at 34 nuclear loci (Grant and Leslie 1996), indicate that the five regional forms of Sardinops (considered separate taxa by most authorities) probably diverged within 500,000 years BP, a much shorter timeframe than predicted by vicariance models based on plate tectonics. High mtDNA haplotype diversity, coupled with an excess of rare alleles in the protein electrophoretic dataset, may indicate exponential growth from a small ancestral population. The mtDNA and allozyme data are concordant with climate records and fossil evidence in portraying regional populations as recent, unstable, and ephemeral. Regional populations of sardines have probably been extinguished and recolonized over short evolutionary timescales in response to changes in climate and the oceanography of coastal upwelling zones.  相似文献

Although the small GTPase Ran is best known for its roles in nucleocytoplasmic transport, mitotic spindle assembly, and nuclear envelope formation, recent studies have demonstrated the overexpression of Ran in multiple tumor types and that its expression is correlated with a poor patient prognosis, providing evidence for the importance of this GTPase in cell growth regulation. Here we show that Ran is subject to growth factor regulation by demonstrating that it is activated in a serum-dependent manner in human breast cancer cells and, in particular, in response to heregulin, a growth factor that activates the Neu/ErbB2 tyrosine kinase. The heregulin-dependent activation of Ran requires mTOR (mammalian target of rapamycin) and stimulates the capped RNA binding capability of the cap-binding complex in the nucleus, thus influencing gene expression at the level of mRNA processing. We further demonstrate that the excessive activation of Ran has important consequences for cell growth by showing that a novel, activated Ran mutant is sufficient to transform NIH-3T3 cells in an mTOR- and epidermal growth factor receptor-dependent manner and that Ran-transformed cells form tumors in mice.  相似文献

The reaction of the chelating P,N ligand RNC(But)CH(R)PPh2 (R = SiMe3) (1) with CuCl and CuCl2 (probably by way of reduction to Cu(I) by the phosphine ligand) or Cu(NCCH3)4ClO4 yielded the dimeric 1:1 complex [Cu{PPh2CH(R)C(But)NR}Cl]2 (2) or the monomeric 2:1 complex [Cu{PPh2CH(R)C(But)NR}2]ClO4 (3), respectively. The presence of trace amounts of water during the reaction resulted in the successive cleavage of the two trimethylsilyl groups of the ligand and the formation of the monomeric chelate complexes [Cu{PPh2CH(R)C(But)NH}2]ClO4 (4) and [Cu{PPh2CH2C(But)NH}2]ClO4 (5). Oxidation of 5 by atmospheric oxygen led to small quantities of the blue Cu(II) complex [Cu{(O)PPh2CH2C(But)NH}2](ClO4)2 (6). The dimeric gold complexes [Au{PPh2CH2C(But)NH}]2X2 (X = BF4, ClO4) (7) were similarly obtained from the previously described Au{PPh2CH(R)C(But)NR}Cl by replacing the covalently bound chlorine with the weakly coordinating anions in the presence of small quantities of water. The solution and solid state structures (except 5) of all complexes were determined by NMR spectroscopy and X-ray crystallography.  相似文献

Many plant species with overlapping geographic ranges segregate at smaller spatial scales. This spatial segregation—zonation when it follows an abiotic gradient and habitat partitioning when it does not—has been experimentally investigated for over a century often using distantly related taxa, such as different genera of algae or barnacles. In those foundational studies, trade-offs between stress tolerance and competitive ability were found to be the major driving factors of habitat partitioning for both animals and plants. Yet, the evolutionary relationships among segregating species are usually not taken into account. Since close relatives are hypothesized to compete more intensely and are more likely to interact during mating compared to distant relatives, the mechanisms underlying habitat partitioning may differ depending on the relatedness of the species in question. Here, I propose an integration of ecological and evolutionary factors contributing to habitat partitioning in plants, specifically how the relative contributions of factors predictably change with relatedness of taxa. Interspecific reproductive interactions in particular are understudied, yet important drivers of habitat partitioning. In spatially segregated species, interspecific mating can reduce the fitness of rare immigrants, preventing their establishment and maintaining patterns of spatial segregation. In this synthesis, I review the literature on mechanisms of habitat partitioning in plants within an evolutionary framework, identifying knowledge gaps and detailing future directions for this rapidly growing field of study.  相似文献

Information on the movement ecology of species can assist with identifying barriers to dispersal and appropriate management actions. We focus on the threatened Malleefowl (Leipoa ocellata) whose ability to move and disperse within fragmented landscapes is critical for their survival. We also investigate the possible effects of climate change on Malleefowl movement. We used solar-powered GPS telemetry to collect movement data and determine the influence of breeding status, remnant vegetation patches and environmental variables. Seven Malleefowl were tracked between 1 and 50 months, resulting in 20 932 fixes. While breeding, Malleefowl had significantly smaller home ranges (92 ± 43 ha breeding; 609 ± 708 ha non-breeding), moved shorter daily distances (1283 ± 605 breeding; 1567 ± 841 non-breeding) and stayed closer to the incubation mound (349 ± 324 m breeding; 3293 ± 2715 m non-breeding). Most Malleefowl effectively disassociated from the mound once breeding stopped, with two birds dispersing up to 10.2 km. Movement patterns were significantly correlated with the size of the remnant native vegetation patch, with smaller home ranges being utilized in small patches than in large patches. One male almost exclusively remained within a 107-ha patch for over 4 years, but a female crossed between closely spaced uncleared patches. Long-range movements of nearly 10 km daily displacement were recorded in large remnants almost exclusively when not breeding. Temperature and rain had a significant effect on movement: modelling suggests daily distances decline from 1.3 km at 25°C to 0.9 km at 45°C, with steeper declines over 30°C. The influence of patch size on movement patterns suggests that Malleefowl movement may be governed by the size of remnant patches and that habitat continuity may be important for facilitating recolonization after catastrophic events and maintaining genetic diversity. Climate change may reduce Malleefowl movement during hot, dry periods possibly affecting breeding success.  相似文献
