The mitogen-activated protein kinases (MAPKs) play essential roles during oocyte maturation and egg activation and are also active in somatic cell cycle regulation in many animals. In clams, starfish, ascidians, mice, and frogs, the species-specific timing of MAPK activity during oocyte maturation and egg activation correlates with the different meiotic arrest points of these various organisms. Furthermore, MAPKs have been shown to regulate the meiotic cell cycle in marine invertebrates and vertebrates. The initial trigger for egg activation in insects is different from that of marine invertebrates and vertebrates, and it was not previously known whether changes in MAPK activity accompany egg activation in insects. To examine the regulation of MAPKs during Drosophila egg activation and early embryogenesis, we quantified the levels of phosphorylated (active) forms of ERK, p38 and JNK by western blotting with antibodies specific to the phospho-forms of these kinases. Levels of phospho-ERK, phospho-p38 and phospho-JNK are high in Drosophila oocytes. Upon egg activation, levels of all these phospho- (active) forms of MAPKs decrease. Fertilization is not required for this decrease, consistent with the independence of egg activation from fertilization in Drosophila. The decrease in levels of phospho-MAPK occurs normally in embryos laid by sterile females mutant in the egg activation genes cortex, sarah, and prage. We present a model in which the decrease in MAPK activity is an intermediate step in the pathway leading from the calcium signal that initiates egg activation to the downstream events of activation.  相似文献

In studies investigating adverse reproductive outcomes associated with video display terminal (VDT) usage, amounts of time spent in front of a VDT or magnetic field (MF) levels in front of the VDT are used as surrogate measures of subject's MF exposure. However, the relevance of such surrogates to actual exposures has not been demonstrated, and the validity of the use of such measures as a surrogate for the actual MF exposure is only speculative. This study examines 1) measurements of MFs at frequencies of approximately 30–1000 Hz at a fixed distance from the VDTs, 2) reported hours of VDT use, and 3) reported distance between the VDT and the subject's waist as surrogate measures for the average MF exposure level of a VDT user during one 8 h workday. The results showed a weak correlation between the average exposure level of a VDT user and the MF 46 cm from a VDT (R = 0.52, n = 67, P < 0.001). This study showed no association between self-reported hours of VDT usage, or self-reported distance between waist and VDT, and the average MF exposures. Moreover, individuals' average MF exposures did not seem to be affected by other variables, such as position of a VDT on the desk, hours of desk use, and the VDT type (color vs. monochrome). These findings indicate that VDT exposures within office settings are complex and cannot be easily predicted by surrogates. © 1996 Wiley-Liss, Inc.  相似文献

Immunization with radiation-attenuated sporozoites (RAS) can confer sterilizing protection against malaria, although the mechanisms behind this protection are incompletely understood. We performed a systems biology analysis of samples from the Immunization by Mosquito with Radiation Attenuated Sporozoites (IMRAS) trial, which comprised P. falciparum RAS-immunized (PfRAS), malaria-naive participants whose protection from malaria infection was subsequently assessed by controlled human malaria infection (CHMI). Blood samples collected after initial PfRAS immunization were analyzed to compare immune responses between protected and non-protected volunteers leveraging integrative analysis of whole blood RNA-seq, high parameter flow cytometry, and single cell CITEseq of PBMCs. This analysis revealed differences in early innate immune responses indicating divergent paths associated with protection. In particular, elevated levels of inflammatory responses early after the initial immunization were detrimental for the development of protective adaptive immunity. Specifically, non-classical monocytes and early type I interferon responses induced within 1 day of PfRAS vaccination correlated with impaired immunity. Non-protected individuals also showed an increase in Th2 polarized T cell responses whereas we observed a trend towards increased Th1 and T-bet+ CD8 T cell responses in protected individuals. Temporal differences in genes associated with natural killer cells suggest an important role in immune regulation by these cells. These findings give insight into the immune responses that confer protection against malaria and may guide further malaria vaccine development.Trial registration: ClinicalTrials.gov NCT01994525.  相似文献

There is increasing interest in developing better predictive tools and a broader conceptual framework to guide the restoration of degraded land. Traditionally, restoration efforts have focused on re-establishing historical disturbance regimes or abiotic conditions, relying on successional processes to guide the recovery of biotic communities. However, strong feedbacks between biotic factors and the physical environment can alter the efficacy of these successional-based management efforts. Recent experimental work indicates that some degraded systems are resilient to traditional restoration efforts owing to constraints such as changes in landscape connectivity and organization, loss of native species pools, shifts in species dominance, trophic interactions and/or invasion by exotics, and concomitant effects on biogeochemical processes. Models of alternative ecosystem states that incorporate system thresholds and feedbacks are now being applied to the dynamics of recovery in degraded systems and are suggesting ways in which restoration can identify, prioritize and address these constraints.  相似文献

An effective approach for enhancing the selectivity of beta-site amyloid precursor protein cleaving enzyme (BACE 1) inhibitors is developed based on the unique features of the S1' pocket of the enzyme. A series of low molecular weight (<600) compounds were synthesized with different moieties at the P1' position. The selectivity of BACE 1 inhibitors versus cathepsin D and renin was enhanced 120-fold by replacing the hydrophobic propyl group with a hydrophilic propionic acid group.  相似文献

Humans and apes are placed together in the superfamily Hominoidea. The evolutionary trajectory of hominoids is intimately bound up with the exploitation of ripe, fleshy fruits. Fermentation of fruit sugars by yeasts produces a number of alcohols, particularly ethanol. Because of their pre-human frugivorous dietary heritage, it has been hypothesized that humans may show pre-existing sensory biases associating ethanol with nutritional rewards. This factor, in turn, could influence contemporary patterns of human ethanol use. At present, there seems little evidence to support a view of selection specifically for ethanol detection or its utilization over the course of hominoid evolution. Ethanol concentration in wild fruits consumed by monkeys and apes is predicted to be low. Wild monkeys and apes avoid consumption of over-ripe fruits, the class showing notable ethanol concentrations, and for this reason, ethanol plumes may act as deterrents rather than attractants. Any energetic benefits to wild primates from ingested ethanol appear negligible, at best. Mice and rats show patterns of ethanol self-administration similar to humans, indicating that a frugivorous dietary heritage is not necessary for such behaviors. In the natural environment, ethanol is predicted to be just one of many alcohols, esters and related compounds routinely encountered by frugivorous primates and of no particular significance. The strong attraction ethanol holds for some individuals could be due to a broad range of genetic and environmental factors. In some humans, the appetite for ethanol appears related to the appetite for sugar. The predisposition some individuals display toward excessive ethanol consumption could involve features of their genetics and biochemical similarities of ethanol and carbohydrate. Regular low ethanol intake is hypothesized to lower the incidence of cardiovascular disease in humans, perhaps through its effects on body fat distribution. Such a benefit, if confirmed, would appear to relate to features of the contemporary human rather than pre-human diet.  相似文献
