The whitefly Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) is a species complex, and its systematic classification requires controlled crossing experiments among its genetic groups. Accurate information on pre‐copulation intervals, copulation frequencies, and initial frequency of egg fertilization of newly emerged adults is critical for designing procedures for collecting the virgin adults necessary for these experiments. In the literature, considerable variation is reported between B. tabaci populations, with respect to the length of the pre‐copulation interval and the initial frequency of egg fertilization. Here, we used a video‐recording method to observe continuously the copulation behaviour of the Mediterranean/Asia Minor/Africa (B biotype) and the Asia II (ZHJ1 biotype) groups of B. tabaci. We also recorded the initial frequency of egg fertilization, as determined by the sex of the progeny. When adults were caged in female–male pairs on leaves of cotton plants, the earliest copulation events occurred 2–6 h after emergence; at 12 h after emergence 56–84% of the females had copulated at least once, and nearly all (92–100%) had copulated at least once by 36 h after emergence. Both females and males copulated repeatedly. Approximately 80 and 20% of copulation events occurred during the photophase and scotophase, respectively. By 72 h post‐emergence, the females of the B and ZHJ1 biotypes had copulated on average 6.1 and 3.9 times, respectively. When adults were caged in groups on plants 1–13 h after emergence, 30–35% of the eggs deposited during this period were fertilized, and approximately 90% of females were fertilized by the end of the 13 h. Although timing of copulation differed in detail between the two genetic groups, the results demonstrate that B. tabaci adults can start to copulate as early as 2–6 h post‐emergence and the majority of females can become fertilized on the day that they emerge.  相似文献

Heat shock resulted in rapid accumulation of large amounts of trehalose in Saccharomyces cerevisiae. In cultures growing exponentially on glucose, the trehalose content of the cells increased from 0.01 to 1 g/g of protein within 1 h after the incubation temperature was shifted from 27 to 40 degrees C. When the temperature was readjusted to 27 degrees C, the accumulated trehalose was rapidly degraded. In parallel, the activity of the trehalose-phosphate synthase, the key enzyme of trehalose biosynthesis, increased about sixfold during the heat shock and declined to the normal level after readjustment of the temperature. Surprisingly, the activity of neutral trehalase, the key enzyme of trehalose degradation, also increased about threefold during the heat shock and remained almost constant during recovery of the cells at 27 degrees C. In pulse-labeling experiments with [14C]glucose, trehalose was found to be turned over rapidly in heat-shocked cells, indicating that both anabolic and catabolic enzymes of trehalose metabolism were active in vivo. Possible functions of the heat-induced accumulation of trehalose and its rapid turnover in an apparently futile cycle during heat shock are discussed.  相似文献

P700 is rapidly, but only transiently photooxidized upon illuminating dark-adapted leaves. Initial oxidation is followed by a reductive phase even under far-red illumination which excites predominantly photosystem (PS) I. In this phase, oxidized P700 is reduced by electrons coming from PSII. Charge separation in the reaction center of PSI is prevented by the unavailability of electron acceptors on the reducing side of PSI. It is subsequently made possible by the opening of an electron gate which is situated between PSI and the electron acceptor phosphoglycerate. Electron acceptors immediately available for reduction while the gate is closed corresponded to 10 nmol · (mg chlorophyll)–1 electrons in geranium leaves, 16 nmol · (mg chlorophyll)–1 in sunflower and 22 nmol · (mg chlorophyll)–1 in oleander. Reduction of NADP during the initial phase of P700 oxidation showed that the electron gate was not represented by ferredoxin-NADP reductase. Availability of ATP indicated that electron flow was not hindered by deactivation of the thylakoid ATP synthetase. It is concluded that NADP-dependent glyceraldehydephosphate dehydrogenase is completely deactivated in the dark and activated in the light. The rate of activation depends on the length of the preceding dark period. Oscillations in the rate of photosynthesis of sunflower (Helianthus annuus L.) leaves were induced by subjecting leaves, whose photosynthetic apparatus had been activated, to a sudden transition from darkness or low light to high-intensity illumination, or by transfering them in the light from air to an atmosphere containing saturating CO2. It was found that at the first maximum, light-and CO2-saturated photosynthesis can be much faster than steady-state photosynthesis. Both QA in the reaction center of PS II and P700 in the reaction center of PS I of the chloroplast electron-transport chain were more oxidized during the maxima of photosynthesis than during the minima. Maxima of P700 oxidation slightly preceded maxima in photosynthesis. During a transition from low to high irradiance, the assimilatory force FA, which was calculated from ratios of dihydroxyacetone phosphate to phosphoglycerate under the assumption that the reactions catalyzed by NADP-dependent glyceraldehydephosphate dehydrogenase, phosphoglycerate kinase and triosephosphate isomerase are close to equilibrium, oscillated in parallel with photosynthesis. However, only one of its components, the calculated phosphorylation potential (ATP)/(ADP)(Pi), paralleled photosynthesis, whereas calculated NADPH/NADP ratios exhibited antiparallel behaviour. When photosynthetic oscillations were initiated by a transition from low to high CO2, the assimilatory force FA declined, was very low at the first minimum of photosynthesis and increased as photosynthesis rose to its second maximum. The observations indicate that the minima in photosynthesis are caused by lack of ATP. This leads to overreduction of the electron-transport chain which is indicated by the reduction of P700. The efficacy of 2‐furfuraldehyde for control of Sclerotium rolfsii was studied in laboratory and greenhouse experiments. Mycelial growth of the fungus was reduced proportionally with concentrations of 0.1–0.5 ml furfuraldehyde l‐1 agar medium, and viability of sclerotia diminished on exposure to 2‐furfuraldehyde vapours. Detectable populations of bacteria and fungi, including Trichoderma spp., were reduced significantly (9=0.05) when furfuraldehyde was added to the agar used for soil dilution plates of untreated soil. Repeated treatments of natural soil with the fumigant significantly increased populations of Trichoderma spp. and bacteria, but diminished numbers of actinomycetes. Increasing dosages applied to soil artificially infested with S. rolfsii caused a reduction of disease on lentil, Lens culinaris. Results indicate that the compound, when applied to field soil, changes the composition of soil microflora and has potential for integrated control of S. rolfsii.  相似文献

Agrotis segetum nuclear polyhedrosis virus (AsNPV) and granulosis virus (AsGV), propagated in laboratory cultures of A. segetum in England and A. ipsilon in Spain, respectively, were applied to plots of maize plants at the one‐ to four‐leaf stage of growth. Plots were arranged in a 6 x 6 Latin square design and infested with second‐instar A. segetum larvae (the common cutworm). Each virus was applied in separate treatments by two application methods; as an aqueous spray containing 0.1% Agral as a wetting agent, and as a bran bait. The NPV was applied at a rate of 4 X 1012 polyhedra/ha, and the GV at 4 X 1013 granules/ha. Soil and plants were sampled for larvae on three occasions following virus treatment: 24 h, 4 days and 11 days. The larvae were reared on diet in the laboratory, until death or pupation, to examine the rate and level of viral infection. Infection data showed 87.5% and 91% NPV infection and 12.5% and 55% GV infection in spray and bait treatments, respectively, in larvae sampled 24 h after treatment. In larvae sampled 4 days after treatment, the results were 78% and 100% NPV infection, and 13% and 6% GV infection. A total of only six larvae were retrieved on day 11. In both treatments larvae infected with AsNPV died significantly more rapidly and at an earlier instar than those infected with AsGV, indicating that AsNPV appears to have better potential as a control agent for A. segetum.  相似文献

Zusammenfassung Die Kinetik der Ionenaufnahme durch junge und alte Sprosse von Mnium cuspidatum wurde untersucht. Die verschieden alten Sprosse unterscheiden sich vor allem durch das Vorhandensein einer aktiven Gipfelknospe bei den jungen Gametophyten, die bei den alten offenbar ihre Tätigkeit eingestellt hat. Im niedrigen Konzentrationsbereich (0–0,5 mM) haben jungen und alte Sprosse hyperbolische Isothermen der Ionenaufnahme, die sich etwas hinsichtlich der apparenten Michaeliskonstanten und der Maximalgeschwindigkeit unterscheiden. Im hohen Konzentrationsbereich (1–10 mM) ist der Unterschied qualitativ. Mit jungen Sprossen erhält man eine lineare oder exponentielle Isotherme, mit alten Sprossen eine hyperbolische Kurve. Der vermutete Einfluß der Gipfelknospe kann mit Effekten von Wuchsstoffen auf den Stofftransport zusammenhängen und läßt einen Einfluß dieser Regulationssysteme auf die Membranfunktion vermuten.
The kinetics of ion uptake by young and old branches of mnium cuspidatum
Summary Isotherms of K(Rb)-, Cl- and SO4-uptake by young and old branches of the moss Mnium cuspidatum were investigated. Old moss gametophytes from the 1966 vegetation period were collected in the forests surrounding Darmstadt from February to mid-April 1967 and from the 1967 season in late September 1967. Young plants were sampled from mid-April to the end of May 1967 and they were also grown by water culture of old plants.Both young and old branches have hyperbolic isotherms of ion uptake in the low concentration range (0–0.5 mM) (Fig. 1–3), which slightly differ in K mand V max (Table). Isotherms in the high range (1–10 mM), however, are drastically different, changing from linear or exponential with young moss branches to hyperbolic with old gametophytes (Figs. 1–3).The linear or exponential high-range isotherm obtained with young moss plants is compared with other examples reported in the literature (Fig. 4). As the leaflets of the moss plants, which constitute 2/3 of the fresh weight of the material used in the experiments, have well developed vacuoles, the correlation between hyperbolic isotherms and vacuolation does not apply here (Fig. 4a, Torii and Laties, 1966).The change in shape of the high-range moss isotherm with age resembles the change from exponential to hyperbolic kinetics in isolated potato discs during washing (Fig. 4b, Laties, Macdonald and Dainty, 1964). The events triggered by isolation of potato discs from the interior of the tuber may be similar to the changes in the moss material under the control of the terminal bud, which is only active in the young branches.The suggested influence of the active terminal bud of young moss plants on the ion absorption process of cells in the tissue may be related to effects of growth substances on translocation reported in the literature and may point to a direct effect of these regulatory systems on membrane function.In this respect the comparison of corn root stele and cortex is of interest. Isolated steles, both freshly isolated and after washing, have exponential isotherms in the high range (Fig. 4c), whereas cortex displays a hyperbolic isotherm which changes little with ageing (Lüttge and Laties, 1967). In contrast to the case in potato and moss materials, this phenomenon is not simply due to ageing but involves morphogenetic differences.Temperature is another factor which influences the shape of the high range isotherm. All examples discussed so far refer to experiments at room temperature. At low temperatures high-range isotherms for proximal root tissue or aged potato discs have an exponential shape (Torii and Laties, 1966; Laties, Macdonald and Dainty, 1964). It thus appears that the exponential isotherm of young moss branches indicates that as in freshly isolated potato discs or in corn root stele the metabolic high-range uptake system is not developed.
