The in vivo dynamics of selenium (Se) and mercury (Hg) interaction was studied in mouse tissues using direct visualization
of individual Se, Hg, and SeHg particles on the surface of circulating erythrocytes. This high-resolution detection of Se
and Hg was obtained by scanning electron microscopy coupled to X-ray microanalysis. BALB/c mice were injected in the peritoneal
cavity with Se and Hg salts, and the animals were sacrificed 3 min after the Hg injection. Only a minority (9%) of the metal
dots seen on mouse liver erythrocytes were SeHg complexes when Se and Hg salts were mixed together before injection. In contrast,
the majority (73%) of metal dots on liver erythrocytes were SeHg complexes if Se was injected at least 5 min before Hg injection.
All metal dots on liver erythrocytes were of SeHg complexes if Se was injected 9 or 12 min before the Hg injection. We conclude
that the formation of stable in vivo SeHg complexes requires preliminary interaction of Se with a putative serum factor before
complexes between Se and Hg are formed and are bound to the erythrocyte cell surface. 相似文献

By using a series of aquaria which were more or less separated into two compartments by a "gate" made of plastic or glass strips, we studied how paradise fish changed their patterns of movement relative to the nature of the gates. We found that opaque plastic gates were well recognized by the paradise fish (Macropodus opercularis) even if the strips were very thin, but that they only changed their pattern of movement when the strips exceeded a certain width. Thus, paradise fish appear to be able to distinguish a physical space of the same size as consisting of either one or two compartments, depending on the material (transparency) and width of the dividing strips. 相似文献

Protein kinase D1 (PKD1) plays a vital role in signal transduction, cell proliferation, membrane trafficking, and cancer; however, the majority of the studies up to date had centered primarily on PKD1 functions in interphase, very little is known about its role during cell division. We previously demonstrated that during mitosis PKD1 is activated and associated with centrosomes, spindles, and midbodies. However, these observations did not address whether PKD1 was associated with mitosis regulation. Accordingly, we used rapidly acting PKD-specific inhibitors to examine the contribution of PKD1 the sequence of events in mitosis. We found that although PKD1 overexpression did not affect mitosis progression, suppression of its catalytic activity by two structurally unrelated inhibitors (kb NB 142-70 and CRT 0066101) induced a significant delay in metaphase to anaphase transition time. PKD1 inhibition during mitosis also produced the appearance of abnormal spindles, defects in chromosome alignment, and segregation as well as apoptosis. Experimental residual dipolar couplings (RDCs) in combination with structural models have the potential for accelerating the
protein backbone resonance assignment process because RDCs can be measured accurately and interpreted quantitatively. However,
this application has been limited due to the need for very high-resolution structural templates. Here, we introduce a new
approach to resonance assignment based on optimal agreement between the experimental and calculated RDCs from a structural
template that contains all assignable residues. To overcome the inherent computational complexity of such a global search,
we have adopted an efficient two-stage search algorithm and included connectivity data from conventional assignment experiments.
In the first stage, a list of strings of resonances (CA-links) is generated via exhaustive searches for short segments of
sequentially connected residues in a protein (local templates), and then ranked by the agreement of the experimental 13Cα chemical shifts and 15N-1H RDCs to the predicted values for each local template. In the second stage, the top CA-links for different local templates
in stage I are combinatorially connected to produce CA-links for all assignable residues. The resulting CA-links are ranked
for resonance assignment according to their measured RDCs and predicted values from a tertiary structure. Since the final
RDC ranking of CA-links includes all assignable residues and the assignment is derived from a “global minimum”, our approach
is far less reliant on the quality of experimental data and structural templates. The present approach is validated with the
assignments of several proteins, including a 42 kDa maltose binding protein (MBP) using RDCs and structural templates of varying
quality. Since backbone resonance assignment is an essential first step for most of biomolecular NMR applications and is often
a bottleneck for large systems, we expect that this new approach will improve the efficiency of the assignment process for
small and medium size proteins and will extend the size limits assignable by current methods for proteins with structural

Predators appear to be less frequently specialised (i.e. adapted to restricted diet) on their prey than herbivores, parasites or parasitoids. Here, we critically evaluate contemporary evolutionary hypotheses that might be used to explain the evolution of specialised foraging in predators. We propose a unifying concept within which we define four types of trophic categories using ecological (diet breadth) and evolutionary (degree of adaptations) contexts. We use data on spiders (Araneae), the most diversified order of terrestrial predators, to assess applicability of frameworks and evolutionary concepts related to trophic specialisation. The majority of spider species are euryphagous but a few have a restricted prey range, i.e. they are stenophagous. We provide a detailed overview of specialisation on different prey types, namely spiders, crustaceans, moths, dipterans, ants, and termites. We also review the available evidence for trophic adaptations, classified into four categories: behavioural, morphological, venomic and metabolic. Finally, we discuss the ecological and evolutionary implications of trophic specialisation and propose avenues for future research. 相似文献

Exercise training has been used for treatment/prevention of many cardiovascular diseases, but the mechanisms need to be clarified. Thus, our aim was to compare oxidative stress parameters between rats submitted to a swimming training and sedentary rats (control). Twelve male rats were divided into two groups: control and exercise training. The exercise training had daily 1 h swimming sessions for 8 weeks and a load (5% of its body mass) was placed in rat's tail. Thereafter the animals were killed, aorta and heart were surgically removed and blood was collected. Body mass gain, thiobarbituric acid reactive species (TBARS), carbonyl content, total reactive antioxidant potential (TRAP), total antioxidant reactivity (TAR), superoxide dismutase (SOD) activity and catalase (CAT) activity were evaluted. The trained rats showed a lower body mass gain and no modifications on heart. An increased SOD activity was observed on aorta after the training, but no changes were seen for CAT activity, which led to an increased SOD/CAT ratio. The arterial TBARS was also increased for trained rats. The decrease in TRAP in exercise training was the single modification on plasma. Our findings suggest that the increased SOD activity could play a role in vascular adaptations to exercise training. Copyright © 2010 John Wiley & Sons, Ltd. 相似文献

An investigation was undertaken to evaluate a set of cyanobacterial strains in terms of production of biocidal compounds exhibiting
allelochemical and fungicidal properties. Two cyanobacterial strains — Anabaena sp. and Calothrix sp. were selected for further investigation, on the basis of their larger inhibition zones on the lawn of Synechocystis and Synechococcus sp. and two phytopathogenic fungi — Rhizoctonia bataticola and Pythium debaryanum. The diameter of the inhibition zone was largest when extracellular filtrates of the two cultures incubated at high light
intensity (90–100 μmol photons m−2 s−1) and temperature (40 ± 2 °C) or grown in medium containing two-folds higher P (1.4 mg/L, as compared to 0.7 mg/L in BG 11
medium) were taken. A pH of 8 was the most optimal for both strains, in terms of growth and biocidal activity. Partial purification
of ethyl acetate extract using TLC, followed by GLC revealed a single peak. This study highlights the importance of environmental
factors in aggravating or reducing the toxic effects of these harmful cyanobacteria and their potential as a biocontrol agent. 相似文献

The activity of the pyridine nucleotide-independent lactate dehydrogenase (iLDH) was characterized in mitochondria isolated from the protist Euglena gracilis. The dissociation constants for L- and D-lactate were similar, but the V(max) was higher with the d isomer. A ping-pong kinetic mechanism was displayed with 2,4-dichlorophenol-indolphenol (DCPIP), or coenzyme Q(1), reacting as the second substrate with the modified, reduced enzyme. Oxamate was a competitive inhibitor against both L- and D-lactate. Oxalate exerted a mixed-type inhibition regarding L- or D-lactate and also against DCPIP. The rate of L-lactate uptake was partially inhibited by mersalyl and lower than the rate of dehydrogenation, which was mersalyl-insensitive. These data suggested that the active site of L-iLDH was orientated toward the intermembrane space. The following observations indicated the existence of two stereo-specific iLDH enzymes in the inner membrane of Euglena mitochondria: a greater affinity of the D-iLDH for both inhibitors, D-iLDH thermo-stability at 70 degrees C and denaturation of L-iLDH, opposite signs in the enthalpy change for the association reaction of the isomers to the enzyme, differential solubilization of both activities with detergents, and different molecular mass. 相似文献
