To reveal the general cytogeographical pattern of Cyanus section Protocyanus in Europe, DNA ploidy and/or chromosome numbers were newly examined for 160 populations by flow cytometry (450 plants) and/or chromosome counting (30 plants). Furthermore, previously published karyological data were revised (236 records). Our analyses confirmed chromosome counts of 2n = 22 for all newly investigated samples of the C. triumfetti group (the records for C. semidecurrens and C. ternopoliensis are new), C. diospolitanus and C. achtarovii; 2n = 44 for C. montanus and C. mollis; and 2n = 20 for C. lingulatus, C. napulifer, C. nissanus, C. orbelicus, C. thirkei, C. tuberosus and C. velenovskyi. The chromosome count of 2n = 20 is the first report for C. epirotus. The cytotype 2n = 40 was newly recorded for the Crimean endemic C. fuscomarginatus and Calabrian and Greek populations of C. graminifolius. The cytotypes 2n = 20 and 2n = 40 were confirmed for C. pindicola. For the first time triploidy (2n~3x~30) was found in C. nissanus, C. thirkei and in a newly discovered hybrid, C. epirotus × C. graminifolius. Two contrasting ecogeographical patterns emerged: cytotypes derived from the base chromosome number x = 11 (2n = 22, 44) are widespread in northern latitudes and ecologically diverse, whereas cytotypes with x = 10 (2n = 20, 30, 40) are confined to mountains in southern Europe. In general, tetraploids have smaller ranges than diploids. The new combinations Cyanus section Protocyanus (Dobrocz.) Ol?avská comb. nov. and Cyanus ternopoliensis (Dobrocz.) Mode of reproduction and presence of reproductive barriers were studied in two closely related members of the genus Cyanus: the C. triumfetti (diploid 2n?=?22) and C. montanus (tetraploid 2n?=?44) groups. Based on results from isolation and emasculation experiments, taxa of these groups can be considered allogamous with a low selfing rate (0.07 %–0.21 % of achenes developed after selfing). Taxa of the C. triumfetti group hybridize easily and produce viable progeny. Differences in the percentage of well-developed achenes per capitulum obtained from interspecific crosses between members of the C. triumfetti group suggested different levels of reproductive isolation. The percentage of well-developed achenes of most homoploid crosses was 3.47 %–8.87 %. Higher percentages of well-developed achenes were obtained from crosses between Eastern Carpathian C. pinnatifidus and Alpine C. triumfetti s. str. (18.36 % ; 26.56 %) and between geographically close taxa in Central Europe (C. dominii, C. strictus and ‘intermediate morphotype’; 12.75 % –17.60 %), which indicate their overall close relatedness. Crossing geographically remote C. strictus and C. triumfetti s. str. yielded no or only few (0.99 %) well-developed achenes, indicating an increased degree of incompatibility in allopatry. The success of heteroploid crosses between plants belonging to different groups was reduced (2.96 %) and suggested reproductive incompatibilities between ploidy levels. The progeny of heteroploid crosses comprised 63 % of triploids of presumable hybrid origin on tetraploid as well as diploid maternal plants. Another 15.22 % of progeny had the maternal cytotype, probably resulting from selfing. Low viability of heteroploid hybrids supports the existence of post-zygotic mechanisms.  相似文献

Background aimsThe effect of granulocyte–colony-stimulating factor (G-CSF) and/or the cytokine fms-like thyrosin kinase 3 (Flt3) ligand on functional outcome and tissue regeneration was studied in a rat model of spinal cord injury (SCI).MethodsRats with a balloon-induced compression lesion were injected with G-CSF and/or Flt3 ligand to mobilize bone marrow cells. Behavioral tests (Basso-Beattie-Bresnahan and plantar test), blood counts, morphometric evaluation of the white and gray matter, and histology were performed 5 weeks after SCI.ResultsThe mobilization of bone marrow cells by G-CSF, Flt3 ligand and their combination improved the motor and sensory performance of rats with SCI, reduced glial scarring, increased axonal sprouting and spared white and gray matter in the lesion. The best results were obtained with a combination of G-CSF and Flt3. G-CSF alone or in combination with Flt3 ligand significantly increased the number of white blood cells, but not red blood cells or hemoglobin content, during and after the time–course of bone marrow stimulation. The combination of factors led to infiltration of the lesion by CD11b+ cells.ConclusionsThe observed improvement in behavioral and morphologic parameters and tissue regeneration in animals with SCI treated with a combination of both factors could be associated with a prolonged time–course of mobilization of bone marrow cells. The intravenous administration of G-CSF and/or Flt3 ligand represents a safe and effective treatment modality for SCI.  相似文献

The synthesis of conjugates consisting of two or three mannose units interconnected by a 1,2,3-triazole linker installed by the "click" reaction is reported. These conjugates were evaluated in mycobacterial mannosyltransferase (ManT) assay. Detailed analysis of the reaction products showed that these compounds with triazole linker between sugar moieties were tolerated by the enzyme, which elongated them by one or two sugar units with α-(1→6) linkage. The effectiveness of this transfer was reduced in comparison to that observed for the acceptor analogues containing a glycosidic linkage, but still, this is the first report on such unnatural compounds serving as substrates for mycobacterial ManT. The ability of the studied compounds to function as acceptors for the ManT suggests that the relative distance and spatial orientation of acceptor octyl hydrophobic aglycone (optimal length for the ManT) and free primary C-6 hydroxy group of the nonreducing terminal mannose unit (to which glycosyl residue is transferred by the mycobacterial ManT) are important for ManT activity, but at the same time, their variations are tolerated by the enzyme in a relatively wide range.  相似文献

Euglena gracilis possessing chloroplasts of secondary green algal origin and parasitic trypanosomatids Trypanosoma brucei, Trypanosoma cruzi and Leishmania major belong to the protist phylum Euglenozoa. Euglenozoa might be among the earliest eukaryotic branches bearing ancestral traits reminiscent of the last eukaryotic common ancestor (LECA) or missing features present in other eukaryotes. LECA most likely possessed mitochondria of endosymbiotic ??-proteobacterial origin. In this study, we searched for the presence of homologs of mitochondria-targeted proteins from other organisms in the currently available EST dataset of E. gracilis. The common motifs in predicted N-terminal presequences and corresponding homologs from T. brucei, T. cruzi and L. major (if found) were analyzed. Other trypanosomatid mitochondrial protein precursor (e.g., those involved in RNA editing) were also included in the analysis. Mitochondrial presequences of E. gracilis and these trypanosomatids seem to be highly variable in sequence length (5?C118 aa), but apparently share statistically significant similarities. In most cases, the common (M/L)RR motif is present at the N-terminus and it is probably responsible for recognition via import apparatus of mitochondrial outer membrane. Interestingly, this motif is present inside the predicted presequence region in some cases. In most presequences, this motif is followed by a hydrophobic region rich in alanine, leucine, and valine. In conclusion, either RR motif or arginine-rich region within hydrophobic aa-s present at the N-terminus of a preprotein can be sufficient signals for mitochondrial import irrespective of presequence length in Euglenozoa.  相似文献

Summary During microbial degradation of polychlorinated biphenyls (PCB) in liquid media two processes take place simultaneously: elimination of PCB and evaporation of PCB. The physical loss of PCB due to evaporation causes frequently false positive results in long-term biodegradation experiments. Therefore, if only the PCB concentration is to be measured, its determination in both liquid and gaseous phase is essential for a correct appraisal of biodegradation. The kinetics of PCB evaporation have been monitored and the evaporation rate constants for individual PCB congeners have been determined. The values of evaporation rate constants show a good corelation with the values of the 1-octanol/water partition coefficient.  相似文献

Summary Crayfish haemolymph contains three types of haemocytes with cytoplasmic granules: coagulocytes, granulocytes and amoebocytes. Muscle degeneration was induced by either a gross mechanical injury or a mild puncture injury of m. extensor carpopoditi. Granulocytes and amoebocytes were involved in the phagocytosis of disintegrating muscle fibres. Within three weeks after the gross injury the first myotubes were found. The formation of regenerated fibres started before the degenerating material was removed completely. Mild injury resulted in the formation of contraction clots, localized at the ends of a fibre and connected to a persistent external lamina in the form of an empty sheath. The external lamina sheaths were invaded by amoebocytes. They arranged themselves into a superficial layer similar to an epithelium, formed gap junctions and zonulae adherentes, and showed an increase in the number of cytoplasmic microtubules. These transformed haemocytes retained their ability to engulf material of the disintegrating fibre. In about three weeks the number of microtubules in the transformed haemocytes decreased, and newly formed contractile filaments appeared. Satellite cells are present along the normal crayfish muscle fibres. Following their activation in degenerated material, they might conceivably induce the transformation of haemocytes into myogenic cells.  相似文献

The gene encoding β-mannanase (EC 3.2.1.78) BaMan26A from the bacterium Bifidobacterium adolescentis (living in the human gut) was cloned and the gene product characterized. The enzyme was found to be modular and to contain a putative signal peptide. It possesses a catalytic module of the glycoside hydrolase family 26, a predicted immunoglobulin-like module, and two putative carbohydrate-binding modules (CBMs) of family 23. The enzyme is likely cell attached either by the sortase mechanism (LPXTG motif) or via a C-terminal transmembrane helix. The gene was expressed in Escherichia coli without the native signal peptide or the cell anchor. Two variants were made: one containing all four modules, designated BaMan26A-101K, and one truncated before the CBMs, designated BaMan26A-53K. BaMan26A-101K, which contains the CBMs, showed an affinity to carob galactomannan having a dissociation constant of 0.34 μM (8.8 mg/liter), whereas BaMan26A-53K did not bind, showing that at least one of the putative CBMs of family 23 is mannan binding. For BaMan26A-53K, kcat was determined to be 444 s−1 and Km 21.3 g/liter using carob galactomannan as the substrate at the optimal pH of 5.3. Both of the enzyme variants hydrolyzed konjac glucomannan, as well as carob and guar gum galactomannans to a mixture of oligosaccharides. The dominant product from ivory nut mannan was found to be mannotriose. Mannobiose and mannotetraose were produced to a lesser extent, as shown by high-performance anion-exchange chromatography. Mannobiose was not hydrolyzed, and mannotriose was hydrolyzed at a significantly lower rate than the longer oligosaccharides.  相似文献
