M-Protein (165 kDa) is a structural constituent of myofibrillar M-band in striated muscle. We generated a monoclonal antibody which recognized a 165-kDa protein from chicken pectoralis muscle in immunoblot analysis and stained the M-band under immunofluorescence microscopy. By screening a lambda gt11 cDNA library from chicken embryonic pectoralis muscle with this antibody, we isolated a cDNA clone encoding the M-protein. Northern blot analysis showed that M-protein mRNA is expressed in pectoralis and cardiac muscle but not in gizzard smooth muscle or non-muscle tissues. Moreover, the anterior latissimus dorsi muscle, which consists almost exclusively of slow fiber types, contains no detectable levels of the mRNA. The full-length cDNA sequence predicted a 1,450-amino acid polypeptide with a calculated molecular weight of 163 x 10(3). The encoded protein contains several copies of two different repetitive motifs: five copies of fibronectin type III repeats are in the middle part of the predicted molecule, and two and four copies of the immunoglobulin C2-type repeats are located toward the NH2-terminal and COOH-terminal regions, respectively. This indicates that M-protein, along with other thick filament-associated proteins such as C-protein, twichin, and titin, belongs to the superfamily of cytoskeletal proteins with immunoglobulin/fibronectin repeats. 相似文献

To compare morphological characters and phylogenetic placement between Japanese and European Russula, 32 specimens of 12 species were collected from Japanese subalpine forests and Northern Europe. Several sequences of nrDNA ITS region (ITS) of these Russula species were obtained. High homological similarities were shown between ITS sequences of several Russula samples collected from Japanese subalpine forests, Europe and North America. These facts show distribution of the same Russula species among these areas. From morphological observations and phylogenetic analyses, two same Russula species, R. velenovskyi, and R. decolorans are found in Japan, Europe and North America. Of these, R. velenovskyi collected from Mt. Fuji, Mt. Nyukasa and Mt. Tateshina in mountainous area of central Honshu is reported as a new Japanese record. 相似文献

Savanna vegetation in the northern region of Brazil is jeopardized by several anthropogenic activities including cattle ranching and extensive agriculture, and soil biota of these ecosystems is virtually unknown. The soils in savannas are poor in nutrients, very acidic, and subject to drought, and under these conditions, arbuscular mycorrhizal fungi (AMF) are likely to play a key role on plant nutrition and improving soil structure. In this study, we surveyed AMF communities in five savanna locations in Roraima state, Northern Brazil. AMF species were identified using two approaches: field collected spores and trap cultures. Twenty-three AMF species were identified, including 21 species in field samples, 8 species in trap cultures, of which 15 and 2 were unique to field and trap culture samples, respectively. Gigaspora margarita, Dentiscutata heterogama, and Glomus sp1 were the most frequent species recovered from all locations. AMF communities were dominated by members of Gigasporaceae that accounted for 50 to 87% of the total species richness within each location. Spore numbers differed across locations and ranged from 5 to 25 spores 100 cm?3 soil. Redundancy analysis indicated that soil organic matter was the only selected predictor among soil parameters and correlated positively with occurrence of Glomus heterosporum. We conclude that savannas in Roraima harbor a high sporulating AMF species richness with communities dominated by members of Gigasporaceae and that organic carbon is an important edaphic factor influencing AMF community composition in this ecosystem. 相似文献

The achlorophyllousMonotropastrum globosum was found growing in aFagus crenata forest. Samples ofM. globosum and their interpenetrating root systems ofF. crenata were collected to investigate the mycorrhizal association.Monotropastrum globosum mycorrhizae showed thick sheaths, invasion of the epidermal cells by fungal pegs, and Hartig nets, which reached only the first layer of cortical cells. TheF. crenata mycorrhizae also showed thick sheaths, but Hartig nets penetrated deep into the cortex and intracellular hypha were seen in the outer cortical cells. The effects of pressure on the sol-gel transition of κ- and ι-carrageenans were studied in KCl solutions under high pressures up to 3000 kg/cm2. The carrageenan gels were destabilized by pressure: the pressure depression of melting temperature, (dT/dP)m, was ?5.7 × 10?3 and ?4.0 × 10?3 K cm2/kg independent of KCl concentration for κ- and ι-carrageenans, respectively. The enthalpy, entropy and volume changes accompanying the gel formation were calculated from the Eldridge-Ferry's plots and the Clausius-Clapeyron equation. The volume change per unit cross-link (two disaccharide residues) was estimated to be (2.5 ~ 4.9) and (1.7 ~ 3.4) ml/mol for κ- and ι-carrageenans, respectively. The compressibility of both carrageenan molecules appeared to be larger by (1.6 ~ 2.6) × 10?12 (κ-form) and by (0.8 ~ 1.3) × 10?12cm2/dyn (i-form) in gel state as compared with in sol state These increases in volume and compressibility on gelation were attributed to a reduction of water of hydration from the carrageenan molecules, which is mainly due to a replacement of the polymer-water hydrogen bond by the polymer-polymer hydrogen bond. We have developed an accurate and sensitive system for the measurement of cytoplasmic free calcium concentrations ([Ca++]i) of a single cell by using UV-laser and Indo-1. By this method, we made the first successful measurement of [Ca++]i of single living vascular smooth muscle cells. [Ca++]i in spontaneously hypertensive rats was elevated and maintained after the 6th passage culture. However, [Ca++]i in Goldblatt hypertensive rats was not elevated. Thus, these results suggest that the maintenance of high [Ca++]i levels of vascular smooth muscle cells in spontaneously hypertensive rats is genetically regulated and that it is one of the mechanisms for hypertension. 相似文献
