It was shown using complement-dependent cytolysis and monoclonal antibodies against CD4, CD8, and NK1.1 antigens that the cortisone-resistant CD3+4-8-NK1.1(-)-thymocytes spontaneously secreted a chemotactic transmitter inducing the release and directed migration of bone marrow cells. When estimating the general profile of the cytokines of these thymocytes by PCR with revertase, it was demonstrated the cells in question did not express cytokines with colony stimulating activities (SCF, IL-3, or GM-CSF) or cytokines affecting the migration of bone marrow stem elements (IL-2, 4, or 7). In addition, an active expression of gene bcl-2 was detected. Thus, the chemotactic cytokine inducing the release of bone marrow stem elements is a product of the cortisone-resistant long-living CD3+4-8-NK1.1(-)-T-cells of the thymus. 相似文献

A dynamic model of the channel of an acetylcholine receptor in a closed state has been proposed. The channel is formed by five a-helices of subunit M2 and stabilized by the cyclic hydrocarbon (CH2)105. The migration of charged and unchanged van der Waals particles with a diameter of 7.72 A equivalent to the diameter of a hydrated sodium ion has been studied. The migration occurred by the action of external force applied to the complex along the channel axis. In the closed state, the inhibition of ions is due to two components: electrostatic interaction and steric constraints. The van der Waals channel gate is formed by residues 13'-A-Val255, B-Val261, C-Val269, D-Val255, and E-Ile264, and the negatively changed residues occurring in the upper part of the channel have a great effect on ion selectivity. 相似文献

In 75 female residents of Vladikavkaz, aged 18-45 years, who had applied to the gynecological department with complaints of vaginal discharge, the qualitative and quantitative composition of vaginal microflora was studied. All these women were divided into 2 groups: group 1 including 38 women living in ecologically unfavorable districts of the city and group 2 including 37 women living under ecologically favorable conditions. The relationship between the ecological situation of the districts of residence and vaginal microflora in women of the reproductive age was established. Unfavorable exogenous factors were found to lead to the development of vaginal dysbacteriosis: a sharp decrease in the amount of lactoflora or its complete absence accompanied by increased amount of staphylococci, enterococci, Escherichia coli, Proteus, Klebsiella and yeast-like fungi of the genus Candida. 相似文献

Possible role of catalases in modification of yeast Saccharomyces cerevisiae response to nitrosative stress was studied in the work. Yeast cell of a wild strain and catalase-defective strains were treated with nitric oxide donor S-nitrosoglutathione, then the cell survival rate, and the levels of protein carbonyls and oxidized glutathione were measured. It was shown that S-nitrosoglutathione decreased cells viability of wild and catalase-defective strains. Unexpected, yeast cells defective by both catalases survived successfully as compared with the cells of the wild strain. The intensity of stress was evaluated by measures of oxidative protein modifications and glutathione oxidation. Treatment with S-nitrosoglutathione did not affect the level of protein carbonyls and was lower by about 14 i 22 % in the cells of double mutant strains after treatments with S-nitrosoglutathione in concentrations of 10 and 20 mM. S-nitrosoglutathione induced a strong increase of the level of oxidized glutathione in yeast cells of the wild strain. This stress slightly increased the level of oxidized glutathione in the yeast cells defective by peroxisomal or both catalases. It is interesting, that an increase of oxidized glutathione level was not observed in the yeast cells defective by cytosolic catalase. The obtained results prove that catalases can modify yeast cell response to the nitrosative stress. 相似文献

Effects of substrate phase state and time factor on variability of human fecal microbiota were studied. It was shown that microecological system of native feces was characterized by marked time-dependent variability. It is unstable and begins to destruct after 24 hours of cultivation. The most sensitive elements of the system were bifidobacteria and Escherichia coli. Change of phase state of biotope eliminated the effect of factor limiting the microecosystem development, which allowed species of obligate and transitory microflora to freely colonize the growth substrate and interact with each other. The mentioned facts demonstrate that fecal microbiota exists in the environment of excess of growth substrate, which colonization is limited by cluster structure of biotope of native feces. It was concluded that phase state of growth substrate and duration of cultivation are important factors determining the population variability of fecal microbiota. 相似文献

The study was focused on localization of "free" phenolic compounds in pea Pisum sativum L. seedling roots grown at 22 and 8 degrees C 24 h after their inoculation with Rhizobium leguminosarum bv. viceae bacteria. A comparison of phenolic compound distribution along the root in root tissues, and results of observation of root hair development on the root surface, response of root hairs to inoculation, manifesting itself in various deformation degree (bends, twists, ect.) enabled us to reveal differences between roots grown at different temperatures. These differences are basically referred to a sector localized 0-5 mm away from the root tip containing meristematic and extending cells. A distribution of phenolic compounds in sectors with root hairs responding to inoculation by various degrees of contortion practically did not depend on the temperature of plant growth. The evidence provided in the course of this work enabled us to suggest that inhibition of pea root infection at low temperature is caused by decelerated growth processes characteristic of both the root itself and root hairs, as well by a slow increase in the root hair zone. 相似文献

Crosslinking of mRNA analog, dodecaribonucleotide pUUAGUAUUUAUU derivative carrying a perfluoroarylazido group at the guanine N7, was studied in model complexes with 80S ribosomes involving tRNA and in binary complex (i.e., in the absence of tRNA). It was shown that, irrespectively of complex formation conditions (13 mM Mg2+, or 4 mM Mg2+ in the presence of polyamines), the mRNA analog in binary complex with 80S ribosomes was crosslinked with sequence 1840-1849 of 18S rRNA, but in the complexes formed with participation of Phe-TPHKPhe (where the G residue carrying the arylazido group occupied position-3 to the first nucleotide of the UUU codon at the P site) the analog was crosslinked with nucleotide 1207. The presence and the nature of tRNA at the E site had no effect on the environment of position-3 of the mRNA analog. Efficient crosslinking of the mRNA analog with tRNA was observed in all studied types of complex. Modified codon GUA, when located at the E site, underwent crosslinking with both cognate valine tRNA and noncognate aspartate tRNA for which the extent of binding at the E site of 80S ribosomes was almost the same and depended little on Mg2+ concentration and the presence of polyamines. 相似文献
