Articular stress fracture arising from the distal end of the third metacarpal bone (MC3) is a common serious injury in Thoroughbred racehorses. Currently, there is no method for predicting fracture risk clinically. We describe an ex-vivo biomechanical model in which we measured subchondral crack micromotion under compressive loading that modeled high speed running. Using this model, we determined the relationship between subchondral crack dimensions measured using computed tomography (CT) and crack micromotion. Thoracic limbs from 40 Thoroughbred racehorses that had sustained a catastrophic injury were studied. Limbs were radiographed and examined using CT. Parasagittal subchondral fatigue crack dimensions were measured on CT images using image analysis software. MC3 bones with fatigue cracks were tested using five cycles of compressive loading at -7,500N (38 condyles, 18 horses). Crack motion was recorded using an extensometer. Mechanical testing was validated using bones with 3 mm and 5 mm deep parasagittal subchondral slots that modeled naturally occurring fatigue cracks. After testing, subchondral crack density was determined histologically. Creation of parasagittal subchondral slots induced significant micromotion during loading (p<0.001). In our biomechanical model, we found a significant positive correlation between extensometer micromotion and parasagittal crack area derived from reconstructed CT images (SR = 0.32, p<0.05). Correlations with transverse and frontal plane crack lengths were not significant. Histologic fatigue damage was not significantly correlated with crack dimensions determined by CT or extensometer micromotion. Bones with parasagittal crack area measurements above 30 mm2 may have a high risk of crack propagation and condylar fracture in vivo because of crack micromotion. In conclusion, our results suggest that CT could be used to quantify subchondral fatigue crack dimensions in racing Thoroughbred horses in-vivo to assess risk of condylar fracture. Horses with parasagittal crack arrays that exceed 30 mm2 may have a high risk for development of condylar fracture.  相似文献

In this study, we investigated how neighbors (i.e., competitors) altered resistance phenotypes, namely plant size and levels of secondary compounds (iridoid glycosides), of individual plants and specifically tested whether neighbor identity mattered. We conducted a greenhouse experiment with Plantago lanceolata and Plantago major (Plantaginaceae) in which each species served as focal plants as well as neighbors in a factorial design. In addition, we harvested plants six and nine weeks after transplantation to test whether effects changed as plants grew. In both species, competition reduced plant size, and this effect increased over time. Plantago lanceolata neighbors suppressed growth of both focal plant species more than P. major neighbors. Effects of competition on levels of secondary compounds were more complex. Concentrations of iridoid glycosides were increased by competition in both species at harvest one. By the second harvest, an effect of competition on iridoid glycosides was found only in P. major. Neighbor identity influenced levels of iridoid glycosides in P. lanceolata at harvest one; concentrations were higher in plants grown with P. lanceolata neighbors than in plants grown with P. major neighbors. We also tested whether there was a trade-off between growth (biomass) and defense (levels of iridoid glycosides). Biomass and iridoid glycoside content were significantly correlated only in plants grown with competition and harvested at nine weeks, and this relationship was positive in both species, indicating that there was no trade-off between growth and defense. This study suggests that neighbor identity could play an important role in interspecific interactions, including the interactions of plants with other trophic levels.  相似文献

In species that reproduce into uncertain environments, the relationship between mean reproductive success (the abundance of new recruits) and the variance in reproductive success (whether adults contribute disproportionally more offspring) may not be straightforward because of stochastic environmental processes that create high variance in reproductive success among adults. In this study, we investigated the relationships between oceanography, reproductive success and reproductive variance in the black rockfish, Sebastes melanops, a long‐lived temperate reef fish with pelagic larvae. We quantified black rockfish recruitment, genetic diversity and growth rates from otolith microstructure over 5 years (2005–2009) during which oceanographic conditions differed. We used cross‐correlations to determine windows of time during which oceanographic variables were significantly correlated with the resulting abundance or genetic diversity of recruits. We found that warmer ocean temperatures were positively correlated with the abundance of recruits, as well as the effective number of breeders. In contrast, the strength of coastal upwelling during settlement was positively correlated with the annual abundance of new recruits, but was negatively correlated with the effective number of breeders. Larval growth rates were explained substantially more by temperature than by upwelling and suggested that temperature affected survival through growth, while upwelling affected survival through transport. Our results indicated that cold ocean temperatures and intense upwelling caused sweepstakes‐like processes to operate on black rockfish populations, despite high abundances of recruits. We propose that a decoupling of the mean and variance in reproductive success may be characteristic of organisms that reproduce into uncertain environments.  相似文献

Although the importance of the extracellular signal-regulated kinase (ERK) pathway in regulating the transition from G1 to S has been extensively studied, its role during the G2/M transition is less well understood. Previous reports have shown that inhibition of the ERK pathway in mammalian cells delays entry as well as progression through mitosis, suggesting the existence of molecular targets of this pathway in M phase. In this report we employed 2-DE and MS to survey proteins and PTMs in the presence versus absence of MKK1/2 inhibitor. Targets of the ERK pathway in G2/M were identified as elongation factor 2 (EF2) and nuclear matrix protein, 55 kDa (Nmt55). Phosphorylation of each protein increased under conditions of ERK pathway inhibition, suggesting indirect control of these targets; regulation of EF2 was ascribed to phosphorylation and inactivation of upstream EF2 kinase, whereas regulation of Nmt55 was ascribed to a delay in normal mitotic phosphorylation and dephosphorylation. 2-DE Western blots probed using anti-phospho-Thr-Pro antibody demonstrated that the effect of ERK inhibition is not to delay the onset of phosphorylation controlled by cdc2 and other mitotic kinases, but rather to regulate a small subset of targets in M phase in a nonoverlapping manner with cdc2.  相似文献

Temperature influences the expression of a wide range of behavioral traits in ectotherms, including many involved in the initiation of pair formation and mating. Although opportunities to mate are thought to be greatest when male and female activity overlap, sex-specific behaviors and physiology could result in mismatched thermal optima for male and female courtship. Here, we investigate how conflicts in the thermal sensitivity of male and female courtship activity affect patterns of mating across temperatures in Enchenopa binotata treehoppers (Hemiptera: Membracidae). These plant-feeding insects coordinate mating with plant-borne vibrational signals exchanged in male–female duets prior to pair formation. We manipulated temperature across an ecologically relevant range (18–36ºC) and tested the likelihood of individual male and female E. binotata to engage in courtship activity using vibrational playbacks. We then staged male–female mating interactions across the same temperature range and quantified the thermal sensitivity of mating-related behaviors across stages of mating. Specifically, we measured the timing of duetting, the likelihood for key pre-copulatory behaviors to occur, whether the pair mated, and copulation duration. We found sex-specific thermal sensitivity in courtship activity: Males showed a clear peak of activity at intermediate temperatures (27–30ºC), while females showed highest activity at the hotter thermal extreme. Mating rates, courtship duets, and copulatory attempts were less likely to occur at thermal extremes. Also, duetting occurred earlier and copulation was shortest at higher temperatures. Overall, our data suggest that sexes differ in how temperature affects mating-related activity and some processes involved in mate coordination may be more sensitive than others across variable thermal environments.  相似文献

Background and AimsDisplacement of native plant species by non-native invaders may result from differences in their carbon economy, yet little is known regarding how variation in leaf traits influences native–invader dynamics across climate gradients. In Hawaii, one of the most heavily invaded biodiversity hotspots in the world, strong spatial variation in climate results from the complex topography, which underlies variation in traits that probably drives shifts in species interactions.MethodsUsing one of the most comprehensive trait data sets for Hawaii to date (91 species and four islands), we determined the extent and sources of variation (climate, species and species origin) in leaf traits, and used mixed models to examine differences between natives and non-native invasives.Key ResultsWe detected significant differences in trait means, such that invasives were more resource acquisitive than natives over most of the climate gradients. However, we also detected trait convergence and a rank reversal (natives more resource acquisitive than invasives) in a sub-set of conditions. There was significant intraspecific trait variation (ITV) in leaf traits of natives and invasives, although invasives expressed significantly greater ITV than natives in water loss and photosynthesis. Species accounted for more trait variation than did climate for invasives, while the reverse was true for natives. Incorporating this climate-driven trait variation significantly improved the fit of models that compared natives and invasives. Lastly, in invasives, ITV was most strongly explained by spatial heterogeneity in moisture, whereas solar energy explains more ITV in natives.ConclusionsOur results indicate that trait expression and ITV vary significantly between natives and invasives, and that this is mediated by climate. These findings suggest that although natives and invasives are functionally similar at the regional scale, invader success at local scales is contingent on climate.  相似文献

The nonclassical class I MHC molecule HLA-G is selectively expressed on extravillous cytotrophoblast cells at the maternal-fetal interface during pregnancy. HLA-G can inhibit the killing mediated by NK cells via interaction with the inhibitory NK cell receptor, leukocyte Ig-like receptor-1 (LIR-1). Comparison of the sequence of the HLA-G molecule to other class I MHC proteins revealed two unique cysteine residues located in positions 42 and 147. Mutating these cysteine residues resulted in a dramatic decrease in LIR-1 Ig binding. Accordingly, the mutated HLA-G transfectants were less effective in the inhibition of NK killing and RBL/LIR-1 induced serotonin release. Immunoprecipitation experiments demonstrated the involvement of the cysteine residues in the formation of HLA-G protein oligomers on the cell surface. The cysteine residue located at position 42 is shown to be critical for the expression of such complexes. These oligomers, unique among the class I MHC proteins, probably bind to LIR-1 with increased avidity, resulting in an enhanced inhibitory function of LIR-1 and an impaired killing function of NK cells.  相似文献
