Tannase producing fungal strains were isolated from different locations including garbages, forests and orchards, etc. The strain giving maximum enzyme yield was identified to be Aspergillus ruber. Enzyme production was studied under solid state fermentation using different tannin rich substrates like ber leaves (Zyzyphus mauritiana), jamun leaves (Syzygium cumini), amla leaves (Phyllanthus emblica) and jawar leaves (Sorghum vulgaris). Jamun leaves were found to be the best substrate for enzyme production under solid-state fermentation (SSF). In SSF with jamun leaves, the maximum production of tannase was found to be at 30 °C after 96 h of incubation. Tap water was found to be the best moistening agent, with pH 5.5 in ratio of 1:2 (w/v) with substrate. Addition of carbon and nitrogen sources to the medium did not increase tannase production. Under optimum conditions as standardized here, the enzyme production was 69 U/g dry substrate. This is the first report on production of tannase by A. ruber, giving higher yield under SSF with agro-waste as the substrate. 相似文献

Proteome alterations in the metal-reducing bacterium Shewanella oneidensis MR-1 in response to different acute dose challenges (0.3, 0.5, or 1 mM) of the toxic metal chromate [Cr(VI)] were characterized with multidimensional HPLC-MS/MS. Proteome measurements were performed and compared on both quadrupole ion traps as well as linear trapping quadrupole mass spectrometers. We have found that the implementation of multidimensional liquid chromatography on-line with the rapid scanning, high throughput linear trapping quadrupole platform resulted in a dramatic increase in the number of measured peptides and, thus, the number of identified proteins. A total of 2406 functionally diverse, nonredundant proteins were identified in this study, representing a relatively deep proteome coverage for this organism. The core molecular response to chromate challenge under all three concentrations consisted predominantly of proteins with annotated functions in transport and binding (e.g., components of the TonB1 iron transport system, TonB-dependent receptors, and sulfate transporters) as well as a functionally undefined DNA-binding response regulator (SO2426) that might play a role in mediating metal stress responses. In addition, proteins annotated as a cytochrome c, a putative azoreductase, and various proteins involved in general stress protection were up-regulated at the higher Cr(VI) doses (0.5 and 1 mM) only. Proteins down-regulated in response to metal treatment were distributed across diverse functional categories, with energy metabolism proteins dominating. Schizophrenia patients suffer from severe cognitive deficits, such as impaired reality monitoring. Reality monitoring is the ability to distinguish the source of internal experiences from outside reality. During reality monitoring tasks, schizophrenia patients make errors identifying "I made it up" items, and even during accurate performance, they show abnormally low activation of the medial prefrontal cortex (mPFC), a region that supports self-referential cognition. We administered 80 hr of computerized training of cognitive processes to schizophrenia patients and found improvement in reality monitoring that correlated with increased mPFC activity. In contrast, patients in a computer games control condition did not show any behavioral or neural improvements. Notably, recovery in mPFC activity after training was associated with improved social functioning 6 months later. These findings demonstrate that a serious behavioral deficit in schizophrenia, and its underlying neural dysfunction, can be improved by well-designed computerized cognitive training, resulting in better quality of life. 相似文献

Phenstatin analogues were synthesized on steroidal framework, for selective targeting of breast cancer cells. These analogues were evaluated for anticancer efficacy against breast cancer cell lines. Analogues 12 and 19 exhibited significant anticancer activity against MCF-7, hormone dependent breast cancer cell line. While analogues 10-14 exhibited significant anticancer activity against MDA-MB-231, hormone independent breast cancer cell line. Compound 10 showed significant oestrogen antagonistic activities with low agonistic activity in in vivo rat model. These analogues also retain tubulin polymerization inhibition activity. The most active analogue 10 was found to be non-toxic in Swiss albino mice up to 300 mg/kg dose. Gallic acid based phenstatin analogues may further be optimized as selective anti-breast cancer agents. 相似文献

Uridine diphosphate glucose pyrophosphorylase (UDP-Glc PPase, EC2.7.7.9) was purified 65 fold from immature grains of wheat (Triticum aestivum L. cv, WH-147) by ammonium sulphate fractionation, DEAE-cellulose anion exchange chromatography and Sephadex G-100 permeation chromatography. The partially purified enzyme, having molecular weight of 72 kD, exhibited broad pH optimum between 8 and 9 and was stable at 4°C for 15 days. At pH 8.5, the enzyme followed typical hyperbolic kinetics with respect to UDP-glucose and inorganic pyrophosphate (Km 0.22 mM and 0.66 mM respectively). The enzyme showed absolute requirement for Mg2+ and did not appear to require sulfhydryl groups for its activity.
