Dexamethasone or Zn++ increase the rate of synthesis of the metal-binding protein metallothionein in hepatocyte cultures. Dexamethasone induction of the capacity to synthesize metallothionein is not blocked by cycloheximide. In contrast, the dexamethasone stimulated increase in Zn++ uptake is inhibited by cycloheximide. Like Zn++, dexamethasone is a “primary inducer” of metallothionein. The glucocorticoid induction of metallothionein in primary cultures of rat hepatocytes is not mediated through elevation of Zn++ uptake.  相似文献

The adaptation of Klebsiella aerogenes to high levels of cadmium was studied in continuous culture under conditions of glucose limitation. When up to 6 × 10−4 M cadmium was added to a culture in steady state, growth ceased instantaneously but resumed within 5 h (dilution rate, 0.1 h−1). When again in steady state, these adapted cells exhibited a far greater tolerance to cadmium than did unadapted cells (not previously exposed to cadmium) when tested on solid media containing different concentrations of cadmium. This relative insensitivity of adapted cells to cadmium was subsequently lost in continuous culture within 5 days after omitting cadmium from the influent medium. Thus, the phenomenon was an inducible physiological process. Adapted cells contained substantial amounts of cadmium (up to 2.4% of the bacterial dry weight). The cadmium content of the cells was dependent on growth conditions and was found to be proportional to the inorganic sulfide content of the cells in all cases. This suggested that formation of CdS is probably the most important mechanism of detoxification in this organism. The presence of large numbers of electron-dense granules on the cell surface (absent in cultures without added cadmium) provided additional support for this conclusion.  相似文献

The benthic deep-water siphonophore Rhodalia miranda was collected for the first time in 1876 by H.M.S. Challenger off the Rio de la Plata estuary beneath the Subtropical Convergence at about 1000 m depth. Rhodalia was reported again about 100 years later from certain distant localities in the subantarctic region of the south-west Atlantic. Hydrographic and topographic features that may be involved in creating this peculiar distribution pattern are discussed together with a likely mode of dispersal of benthic coelenterates. The disjunct distribution of Rhodalia and other benthos of the upper Argentine Slope suggests the subsurface oceanographic régime to be quite different from what we know of the surface layers.  相似文献

Many sialic acid-containing oligosaccharides and five acidic monosaccharides have been separated by high-performance anion-exchange chromatography using a Dionex AS6 ion-exchange column eluted with aqueous 50 mM NaOH plus 50-175 mM sodium acetate. Using a pulsed amperometric detector, as little as 50 pmol oligosaccharide can be detected. Many factors, such as the presence of fucosyl groups or sialyl groups, glycosidic linkage positions, and branching structure, can have a tremendous influence on overall acidity of the oligosaccharide, which can lead to excellent separations and make this method an important addition to existing alternatives for the separation of sialic acid-containing oligosaccharides.  相似文献

The chemotactic responses by starved cells of marine Vibrio sp. strain S14 differed from those elicited by cells that were not nutrient limited. The rate of chemotaxis at different concentrations of several attractants varied for starved and growing cells. Vibrio sp. strain S14 showed positive chemotaxis to leucine, valine, arginine, and glucose at the onset of energy and nutrient deprivation. A continued, though decreased, positive response was demonstrated fro leucine, arginine, and glucose at 10 h of starvation. Cells starved for 3 h displayed a stronger response to glucose than those starved for shorter or longer times. However, cells starved for 5 and 10 h responded more strongly to a lower concentration of glucose than did cells starved for 0 and 3 h. Starvation for 24 h elicited no measurable chemotaxis to leucine, arginine, or glucose. The motility decreased by over 95% in the cell population after 24 h of starvation, which resulted in a low sensitivity in the chemotaxis assay. A switch in the response to valine was observed by 3 h of starvation. The addition of nutrients of 22-h-starved cells elicited a temporary positive chemotactic response to leucine by 2 and 4 h of nutrient recovery, while cells at 1 and 6 h of recovery showed no response. At 2 h of recovery, the greatest response was recorded to 10−4 M leucine, whereas at 4 h it was to 10−2 M leucine. Ten to fifty percent of the 22-h-starved cell population regained their motility after 4 h of nutrient-aided recovery. It is possible that two types of chemosensory systems exist in marine bacteria. Starved and growing cells responded to different concentrations of the attractant, and growing cells displayed a saturated chemotactic system with leucine as the attractant, unlike the response during starvation.  相似文献

Dermorphin and its Hyp6 analogue are opiate-like heptapeptides originally discovered in frog skin and characterized by the presence of a D-Ala2 residue in their sequence. They were assayed for their capacity to compete with [3H]Leu-enkephalin for binding to opioid receptors in membranes of neuroblastoma x glioma hybrid cells. In the presence of 7 nM-[3H]Leu-enkephalin, the concentrations at which they caused 50% inhibition of [3H]enkephalin binding (IC50 values) are 0.1 micro M and 0.3 micro M, respectively. In contrast, the synthetic L-Ala2-dermorphin shows very low affinity for the opioid receptors. In addition, like other opioid peptides, dermorphin and hyp6-dermorphin inhibit the elevation by prostaglandin E1 (PGE1) of the level of adenosine 3':5'-cyclic monophosphate (cyclic AMP) (IC50 values 0.2 micro M and 0.4 micro M, respectively). The inhibition is prevented by the opiate antagonist naloxone, L-Ala2-dermorphin is at least three orders of magnitude less potent in inhibiting the PGE1-evoked increase in the level of cyclic AMP. The results show that peptides with an amino acid sequence quite different from that of the enkephalins can bind to opioid receptors of the hybrid cells.  相似文献
