This pilot study was carried out to evaluate the occurrence of Clostridium difficile in marine environments and in edible shellfish. Samples of seawater, sediment, and zooplankton were collected at five sampling
stations in the Gulf of Naples. Six samples of edible shellfish, furthermore, were obtained: two from mussel farms and four
from wholesalers. The isolation and the characterization of C. difficile strains were carried out using selective media and molecular techniques, respectively. C. difficile was isolated from nine of the 21 samples investigated. Shellfish and zooplankton showed the highest prevalence of positive
samples. No C. difficile was detected in marine sediment. Majority of the C. difficile isolates were toxin A/B positive. Six known different PCR ribotypes (003, 005, 009, 010, 056, and 066) were identified, whereas
one strain may represent a new PCR ribotype. C. difficile may be present in the marine environment in Southern Italy, including shellfish and zooplankton. This study is reporting

Comparative mineral and hormonal analyses were made on tissue culture derived truncated leaf syndrome and wild type oil palm seedlings. Mineral analysis confirmed that Boron, Zinc and chlorophyll levels were significantly lower in truncated leaf syndrome leaves than those of wild type. Hormonal analysis also revealed various cytokinin derivatives such as trans-zeatin, trans-zeatin riboside, trans-zeatin O-glucoside and trans-zeatin riboside 5??mono phosphate were significantly higher in truncated leaf syndrome leaves compared to wild type leaves. Brassinolide level was also significantly higher in truncated leaf syndrome leaves than those of the wild type. These observations suggest that the truncated leaf syndrome abnormality could be associated to high cytokinin and brassinosteroid production which affects the uptake of Boron and Zinc. 相似文献

The effect of divalent Mg2+ and Mn2+ cations on the elongation of ApU, UpA and their 3'-O- and 5'-O-phosphonylmethyl analogues by RNA polymerase holoenzyme to the corresponding trinucleo-tides on a poly(dA-dT) template was investigated. In contrast to Mgz+ ions, Mn2+ ions enhance abortive trinucleotide synthesis. This effect is more pronounced with phosphonylmethyl analogues. The core enzyme cannot catalyze the elongation of either (2'-5') UpA or phosphonylmethyl analogues. The localization of the divalent cation activator, as well as the role of the σ subunit at the catalytic centre of the holoenzyme, is discussed. 相似文献

A randomisation test is described for assessing relative abundance predictions from the maximum entropy approach to biodiversity. The null model underlying the test randomly allocates observed abundances to species, but retains key aspects of the structure of the observed communities; site richness, species composition, and trait covariance. Three test statistics are used to explore different characteristics of the predictions. Two are based on pairwise comparisons between observed and predicted species abundances (RMSE, RMSESqrt). The third statistic is novel and is based on community‐level abundance patterns, using an index calculated from the observed and predicted community entropies (EDiff). Validation of the test to quantify type I and type II error rates showed no evidence of bias or circularity, confirming the dependencies quantified by Roxburgh and Mokany (2007) and Shipley (2007) have been fully accounted for within the null model. Application of the test to the vineyard data of Shipley et al. (2006) and to an Australian grassland dataset indicated significant departures from the null model, suggesting the integration of species trait information within the maximum entropy framework can successfully predict species abundance patterns. The paper concludes with some general comments on the use of maximum entropy in ecology, including a discussion of the mathematics underlying the Maxent optimisation algorithm and its implementation, the role of absent species in generating biased predictions, and some comments on determining the most appropriate level of data aggregation for Maxent analysis. 相似文献

A previously isolated cDNA molecule from Gerbera hybrida (Asteraceae) codes for a new chalcone synthase-like polyketide synthase, 2-pyrone synthase (2PS). 2PS is able to synthesise 4-hydroxy-6-methyl-2-pyrone (triacetolactone), a putative precursor for gerberin and parasorboside, two abundant glucosides in gerbera. In this study, we show that gerbera plants transformed with the gene for 2PS in an antisense orientation and unable to synthesise gerberin and parasorboside are susceptible to Botrytis cinerea infection. In addition to the preformed glucosides, the transgenic plants also lack several compounds that are induced in control plants when infected with the mould. Some of these induced substances are effective in inhibiting fungal growth both in vitro and in vivo. Two of the phytoalexins were identified as the aglycones of gerberin and trans-parasorboside. The third phytoalexin is a rare coumarin, 4-hydroxy-5-methylcoumarin; however, it is typical of many plants of the sunflower family Asteraceae. The coumarin cannot be structurally derived from either gerberin or parasorboside, but may be derived from a related polyketide intermediate. 相似文献

Capsule Populations of most forest bird species increased between 1982 and 2003, probably due to increased forest cover and changes in forest age-class composition. Aims To determine population changes of forest birds in the Czech Republic and to determine their possible causes. Methods Population data were collected via the Breeding Bird Monitoring Programme, which is based on skilled volunteers counting birds at point transects using a standardized technique. Population trends and indices for the period 1982–2003 were calculated for 47 species using log-linear models. Published data on development of forest cover and forest age composition in the Czech Republic were used to indicate environmental change over the same period. Results Populations of most forest species increased between 1982 and 2003. There was also an increase in forest cover and an increase in the proportion of older forest age-classes. The increase in forest specialist birds was positively correlated with the average increase in forest coverage. Conclusions The populations of Czech forest birds have increased in the last two decades. This contrasts with widely reported declines of farmland bird populations throughout Europe. The correlation between populations of specialized forest species and extent of forest habitat suggests that changes in land-use are an important factor. However, increasing cover of mature forests could have a similar effect on the populations of specialist species. 相似文献

Slowly cooled cells of Streptomyces aureofaciens contained mainly tight-couple ribosomes. Maximum rate of polyphenylalanine synthesis on ribosomes of S. aureofaciens was observed at 40°C, while cultures grew optimally at 28°C. Ribosomes of S. aureofaciens differed from those of E. coli in the amount of poly(U) required for maximum synthetic activity. The polyphenylalanine-synthesizing activity of E. coli ribosomes was about 3-times higher than that of S. aureofaciens ribosomes. The addition of protein S1 of E. coli or the homologous protein from S. aureofaciens had no stimulatory effect on the translation of poly(U). In order to localize alteration(s) of S. aureofaciens ribosomes in the elongation step of polypeptide synthesis we developed an in vitro system derived from purified elongation factors and ribosomal subunits. The enzymatic binding of Phe-tRNA to ribosomes of S. aureofaciens was significantly lower than the binding to ribosomes of E. coli. This alteration was mainly connected with the function of S. aureofaciens 50 S subunits. These subunits were not deficient in their ability to associate with 30 S subunits or with protein SL5 which is homologous to L7/L12 of E. coli. 相似文献

Mustard oil (MO) is a plant-derived irritant that has been extensively used in experimental models to induce pain and inflammation. The noxious effects of MO are currently ascribed to specific activation of the cation channel TRPA1 in nociceptive neurons. In contrast to this view, we show here that the capsaicin receptor TRPV1 has a surprisingly large contribution to aversive and pain responses and visceral irritation induced by MO. Furthermore, we found that this can be explained by previously unknown properties of this compound. First, MO has a bimodal effect on TRPA1, producing current inhibition at millimolar concentrations. Second, it directly and stably activates mouse and human recombinant TRPV1, as well as TRPV1 channels in mouse sensory neurons. Finally, physiological temperatures enhance MO-induced TRPV1 stimulation. Our results refute the dogma that TRPA1 is the sole nocisensor for MO and motivate a revision of the putative roles of these channels in models of MO-induced pain and inflammation. We propose that TRPV1 has a generalized role in the detection of irritant botanical defensive traits and in the coevolution of multiple mammalian and plant species. 相似文献

According to a general paradigm, proper DNA duplication from each replication origin is ensured by two protein complexes termed replisomes. In prokaryotes and in budding yeast Saccharomyces cerevisiae, these two replisomes seem to be associated with one another until DNA replication initiated from the origin has finished. This arrangement results in the formation of the loop of newly synthesized DNA. However, arrangement of replisomes in other eukaryotic organisms including vertebrate cells is largely unknown. Here, we used in vivo labeling of DNA segments in combination with the electron microscopy tomography to describe the organization of replisomes in human HeLa cells. The experiments were devised in order to distinguish between a model of independent replisomes and a model of replisome couples. The comparative analysis of short segments of replicons labeled in pulse-chase experiments of various length shows that replisomes in HeLa cells are organized into the couples during DNA replication. Moreover, our data enabled to suggest a new model of the organization of replicated DNA. According to this model, replisome couples produce loop with the associated arms in the form of four tightly associated 30 nm fibers. 相似文献
