The spectra and kinetics of light-induced absorbance changes in the near-infrared region of subchloroplast fragments enriched by P700 were studied. An increase in absorbancy within the region of 725--900 nm upon illumination was characterized by a maximum around 810 nm and by "shoulders" around 760 and 870 nm. Similar effects of thermal inactivation and low temperatures on the duration of dark recovery of light-induced absorbance changes at 700 nm and within the region of 725--900 nm suggest that the absorbance changes in the near-infrared region are due to photooxidation of P700. The values of P700 differential extinction coefficients at 810 nm are 8,2.10(3) M-1.cm-1 for digitonin fragments and 7,7.10(3) M-1.cm-1 for fragments prepared with the use of diethyl ester. It was shown that the value of midpoint oxidation-reduction potential measured for the absorbance changes at 810 nm (+492 mv) is higher than that measured at 700 nm (+475 mv).  相似文献

In etiolated pea and maize leaves illuminated after incubation at 38 degreesC, a new dark reaction was shown manifested in the bathochromic shift of spectral bands and accompanied by esterification of the product of protochlorophyllide photochemical reduction--Chld 684/676: Chld 684/676 --> Chl 688/680. After completion of the reaction a rapid (20-30 sec) quenching of the fluorescence of the reaction product (Chl 688/680) was observed. The reaction Chld 684/676 --> Chl 688/680 is inhibited under anaerobic conditions and in the presence of cyanide; the reaction accompanied by Chl 688/680 fluorescence quenching is not observed in pea mutants with impaired function of photosystem II reaction centers. The spectral properties of the formed Chl form with the absorption maximum at 680 nm, fluorescence quenching, and simultaneous synthesis of pheophytin suggest that the reaction is connected with the chlorophyll of photosystem II reaction center--P-680.  相似文献

We report a patient who received a stent following intracoronary 3-irradiation. Despite a good initial angiographic result, the stent appeared to be not fully expanded on intravascular ultrasound imaging at 6-month follow-up. Cadmium (Cd) is an industrial and environmental pollutant that produces toxic effects on gametogenesis, pre- and post-implantation embryos, and the placenta. Because the effects of acute Cd intoxication on the placenta are not well understood, we investigated changes in its glycosylated components in Cd treated dams at days 4, 7, 10 and 15 of gestation using lectin histochemistry. CdCl2 was administered to pregnant rats; control animals received sterile normal saline. Placentas were processed for DBA, Con A, SBA, PNA, UEA-I, RCA-I and WGA lectin histochemistry to evaluate changes in the carbohydrate pattern of the placenta that might modify cell interactions and contribute to embryonic alterations. Lectin binding was analyzed in the yolk sac; trophoblast giant cells; trophoblast I, II and III; spongiotrophoblast cells and endovascular trophoblast cells in the chorioallantoic placenta. Our lectin binding patterns showed that Cd caused alteration of SBA and DBA labeling of trophoblast-derived cells, which suggested increased expressions of α and β GalNAc. Cd also caused decreased UEA-1 binding affinity, which indicated fewer α-L-Fuc residues in placentas of Cd treated dams. The nonreactivity in trophoblast I of the control placentas incubated with Con-A contrasted with the labeling in placentas of experimental dams, which indicated increased expression of terminal α-D-Man, and α-D-Glc residues. We found that Cd altered the reactivity of placenta to several lectins, which indicated modification of the glycotype presented by the fetal component of the placenta. We report that Cd exerts a deleterious effect on the glycosylation pattern of the placenta.  相似文献

The interaction of Ethidium Bromide (EtBr) with double-stranded (ds-) and single-stranded (ss-) poly[d(A-T)] was studied in different ionic strengths solutions. Optical spectroscopy and Scatchard analysis results indicate that the ligand interacts to both helix and coiled structures of the polynucleotide by "strong" and "weak" binding modes. The association parameters (binding constant -K- and the number of nucleotides corresponding to a binding site -n) of the strong type of interaction were found to be independent of Na+ concentration. Weak interaction occurs at low ionic strength and/or high EtBr concentration. Estimated binding parameters of EtBr with ss- and ds-polynucleotide are in good agreement with those for EtBr-B-DNA complexes. Data obtained provided an evidence for a stacking interaction of EtBr with single stranded poly[d(A-T)].  相似文献

The influence of unilateral destruction of n. entopeduncularis on learning and retention of simultaneous discrimination in a T-maze and adaptation in the open filed was studied in 29 adult rats. Destruction of n. entopeduncularis in rats resulted in appreciable deviations from the normal behavior during realization of earlier acquired simultaneous discrimination and slowing down of the acquisition of a new habit. Differences in behavioral activity of the lesioned animals in the maze and open field were revealed.  相似文献

The interaction of ethidium bromide with single-stranded synthetic and natural polynucleotides at high temperatures (t = 70 degrees C) and low pH values (pH 3.0) was studied. The isotherms of adsorption of ethidium bromide on single-stranded DNA were obtained. Two modes of binding of single-stranded DNA, strong and weak, were revealed. The values of the corresponding constants of interaction of this ligand and the number of bases per one binding site were determined.  相似文献

The interaction of meso-tetra(4-N-hydroxyethylpyridyl)porphyrin, meso-tetra(3-N-hydroxyethylpyridyl)porphyrin, and their zinc complexes with bovine serum albumin (BSA) was studied by electronic spectroscopy, CD, and equilibrium dialysis at pH 7.2. The titration of the porphyrins with BSA was accompanied by a decrease in light absorption and a bathochromic shift of the Soret band, as well as by the appearance of an isobestic point. The porphyrin interaction with BSA also led to the induction of positive CD spectra in the visible region, which is explained by the porphyrin sorption on the protein globule. The equilibrium dialysis helped in determining the stoichiometry of binding and the binding constants of the porphyrins under study with BSA using Scatchard plots. The structures of limbic system have great integration in vegetative reactions. In the oxygen deficiency conditions we studied influence of irritation of limbical cortex, orbitofrontal cortes, hipotalamys, septum and hippocampus (CA1 and CA3 areas) on the impulse activity of respiratory neurons. Phases of hypoxia were the model of experiment. Irritation some of this structures on normoxia and 4-5 thousand meters higher, we discover inhibiting influense on respiratory neurons; some structures irritation had activating influence. In difficult conditions of hypoxia (7.5-8 thousand meters) on the reduction of the impulse activity of neurons, stimulation induced uncharacteristic reactions. Those different reactions of irritation limbic structures have regulation sense on respitatory neurons.  相似文献

Injection of adrenergic and cholinergic agents to animals in the normal athmospheric conditions did not tigger drastic changes on the electric activity of the brain and heart. Acutehipoxia demands high adaptability from the body. In such conditions stimulation of reticular formation and hypothalamus produces different changes in the EEG and ECG activity whith injecting adrenergic and cholinergic agents. It was determined that cholinergic influence are effective in the regulation of electrical brain activity while adrenergics are more important for the realization of descending influences of the truncus cerebri vegetative centers and are less active in the modulation of the cerebral cortex activity.  相似文献
