In this paper we introduce a preliminary work in which a redox mediator containing polyvinylferrocene immobilized on silica gel particles is synthesized. Carbon paste electrodes prepared with this polymeric mediator were studied for their stability in aqueous media for a month. The results indicated that approximately 70% of the initial activity was retained after a month. A steady state amperometric measurement of this carbon paste electrode containing this mediator and glucose oxidase at different potentials showed a linear relationship at elevated glucose concentration. 相似文献

Hydrogenobacter thermophilus cytochrome c(552) ( Ht cyt c(552)) is a small monoheme protein in the cytochrome c(551) family. Ht cyt c(552) is unique because it is hypothesized to undergo spontaneous cytoplasmic maturation (covalent heme attachment) when expressed in Escherichia coli. This is in contrast to the usual maturation route for bacterial cytochromes c that occurs in the cellular periplasm, where maturation factors direct heme attachment. Here, the expression of Ht cyts c(552) in the periplasm as well as the cytoplasm of E. coli is reported. The products are characterized by absorption, circular dichroism, and NMR spectroscopy as well as mass spectrometry, proteolysis, and denaturation studies. The periplasmic product's properties are found to be indistinguishable from those reported for protein isolated from Ht cells, while the major cytoplasmic product exhibits structural anomalies in the region of the N-terminal helix. These anomalies are shown to result from the retention of the N-terminal methionine in the cytoplasmic product, and not from heme attachment errors. The (1)H NMR chemical shifts of the heme methyls of the oxidized ( S=1/2) expression products display a unique pattern not previously reported for a cytochrome c with histidine-methionine axial ligation, although they are consistent with native-like heme ligation. These results support the hypothesis that proper heme attachment can occur spontaneously in the E. coli cytoplasm for Ht cyt c(552). 相似文献

A chronological relationship between the annual profiles of oxidative stress markers, the key regulator of stress physiology has been sought in a terrestrial mollusc (Nerita articulata) under natural photothermal conditions. The hemolymph samples were collected at two different times in each month (from January to December) and the same was repeated for two consecutive years throughout an annual cycle. The fluctuations in the concentrations of certain heavy and trace metals (zinc, copper, cadmium, mercury, lead, and nickel) in both soil and hemolymph of Nerita are also estimated accordingly. Therefore, the present study aims to explore the rhythmic responses of oxidative stress marker to assess the impact of different trace and heavy metals on selected mollusc species. We tries to develop a realistic conceptual idea to analyze and predict the effect of changing environmental pollution on the possible shift in the rhythmicity of aforesaid antioxidants in terrestrial mollusc and their adaptive responses to thrive in such environment. Our results indicates that the amplitude of circannual rhythms of all the selected stress markers varied accordingly but the pattern of annual fluctuation is noted to be similar, and correlated with the metal accumulation. Mycobacterium tuberculosis decaprenylphosphoryl-β-d-ribose oxidase (MtbDprE1) acts in concert with decaprenylphosphoryl-β-d-ribose 2-epimerase (MtbDprE2) and catalyzes the epimerization of DPR into DPA. DPA is the sole precursor for synthesis of arabinogalactan and lipoarabinomannan in the mycobacterial cell wall. MtbDprE1 is a unique antimalarial drug target and many covalent and non-covalent inhibitors against MtbDprE1 have been studied for their antituberculosis activities. In the current study, we have purified MtbDprE1 enzyme and synthesized six sulfur-rich 2-mercaptobenzothiazole and 1, 2, 3-triazole conjugated ligands and performed binding analysis with MtbDprE1. All ligands have shown competitive binding, as observed for other covalently and noncovalently bound MtbDprE1 inhibitors. Molecular docking analysis of six ligands with MtbDprE1 shows that they occupy the substrate binding pocket of MtbDprE1 and are stabilized by hydrogen bonds and van der Waals interactions. Our study shows that sulfur-rich 2-mercaptobenzothiazole ligands act as specific inhibitors against MtbDprE1 and could be used as antituberculosis agents. 相似文献

The cognitive capacities of juvenile chimpanzees, normal children, and retarded children were evaluated by using a nonverbal,
Piagetian-type multiple classification task. The three groups of subjects were tested with the same two by two stimulus matrix,
which was formed by combining two different colors with two different shapes. On each problem the subjects were required to
select one of four stimulus items from the response tray and place it inside the empty cell of the stimulus matrix. It was
found that the human and nonhuman primates tested were able to select objects with correct color and shape cues (C+/S+)significantly more often than the other objects with only color cues correct (C+/S-), only shape cues correct (C-/S+), or neither cue correct (C-/S-). Although the two groups of human children were able to select the C+/S+ objects about 100% of the time by the end of testing, the normal children required significantly fewer problems than the
retarded children. The juvenile chimpanzees needed significantly more problems than the human subjects before they consistently
selected the C+/S+ object on the first trial and attained a level of correct responding that was above chance. Moreover, their level of performance
did not exceed 70% correct. These data suggest that the human children (both normal and retarded) used a conceptual strategy,
while the juvenile chimpanzees employed a perceptual strategy to solve the multiple classification problems. The relationship
between language and conceptual problem-solving strategies for Piagetiantype tasks is discussed.

Two tetraploid species of Hibiscus section Furcaria, H. acetosella and H. radiatus, have an AABB genomic constitution. The diploid species, H. cannabinus (AA) and H. surattensis (BB), were hybridized to develop interspecific alloploid (AB) hybrids. The synthetic interspecific hybrid exhibited intermediate morphological characters, with expression of domestication-related traits, but exhibited higher genomic association with the B genome donor. Evolution of allopolyploids in section Furcaria was found to be associated with mutations in repetitive sequences, leading to higher variation in the tetraploid genome. Allopolyploidization was observed to be associated with both loss of repetitive sequences and appearance of new alleles. Genetic diversity analysis using ISSR and cross-species SSR markers revealed a closer association of diploid genomes and high variability of tetraploid genomes. The evolution of AABB tetraploids in this section possibly took place by hybridization of the A and B genome in geographically isolated regions. 相似文献
