Allozyme variation in Chondrostoma from Austrian, French and northern Greek rivers was investigated. Populations of C. nasus from the upper Danube catchment are genetically very similar and apparently connected by high level of gene flow. Chondrostoma nasus has conserved all major allozyme polymorphisms during its recent range expansion into the Rhone basin. Chondrostoma vardarense from the Axios and Aliakmonas rivers share most of their allozymes with C. nasus from the upper Danube catchment, but have less genetic variation, a possible consequence of a genetic bottleneck following river capture. 相似文献

Among HIV-infected individuals, co-infection with Mycobacterium tuberculosis is associated with faster progression to AIDS. We investigated the hypothesis that M. bovis BCG and M. tuberculosis (Mtb complex) could enhance susceptibility of CD4+ cells to HIV infection. Peripheral blood mononuclear cells (PBMCs) collected from healthy donors were stimulated with M. bovis BCG, M. tuberculosis CDC1551 and M. smegmatis MC(2)155, and stimulated CD4+ cells were infected with R5-and X4-tropic single replication-competent pseudovirus. CD4+ cells stimulated with Mtb complex showed enhanced infection with R5- and X4-tropic HIV, compared to unstimulated cells or cells stimulated with M. smegmatis (p<0.01). Treatment with TLR2 siRNA reversed the increased susceptibility of CD4+ cells with R5- and X4-tropic virus induced by Mtb complex. These findings suggest that TB infection and/or BCG vaccination may be a risk factor for HIV acquisition. 相似文献

Boron toxicity has been recognised as an important problem limiting production in the low-rainfall regions of southern Australia,
West Asia and North Africa. Genetic variation for boron toxicity tolerance in barley has been characterised but the mode of
inheritance and the location of genes controlling tolerance were not previously known. A population of 150 doubled-haploid
lines from a cross between a boron toxicity tolerant Algerian landrace, Sahara 3771, and the intolerant Australian cultivar
Clipper was screened in four tolerance assays. An RFLP linkage map of the Clipper×Sahara population was used to identify chromosomal
regions associated with boron tolerance in barley. Interval regression-mapping allowed the detection of four chromosomal regions
involved in the boron tolerance traits measured. A region on chromosome 2H was associated with leaf-symptom expression, a
region on chromosome 3H was associated with a reduction of the affect of boron toxicity on root growth suppression, a region
on chromosome 6H was associated with reduced boron uptake, and a region on chromosome 4H was also associated with the control
of boron uptake as well as being associated with root-length response, dry matter production and symptom expression. The benefits
and potential of marker-assisted selection for boron toxicity tolerance are discussed.

The cereal cyst nematode (CCN), Heterodera avenae Woll., is an economically damaging pest of barley in many of the world’s cereal-growing areas. The development of CCN-resistant
cultivars may be accelerated through the use of molecular markers. A number of resistance genes against the pest are well
known; one of them, the single dominant Ha 2 resistance gene, has been shown to be effective against the Australian pathotype and maps to chromosome 2 of barley. Segregation
analysis identified two restriction fragment length polymorphism (RFLP) markers flanking the resistance gene in two doubled-haploid
populations of barley. AWBMA 21 and MWG 694 mapped 4.1 and 6.1 cM respectively from the Ha 2 locus in the Chebec×Harrington cross and 4.0 and 9.2 cM respectively in the Clipper×Sahara cross. Analysis of a further seven
sources of CCN resistance in the form of near-isogenic lines (NILs) indicates that all available sources of resistance to
the Australian pathotype of CCN in barley represent the Ha 2 locus.

The frequency of individual genetic mutations conferring drug resistance (DR) to Mycobacterium tuberculosis has not been studied previously in Central America, the place of origin of many immigrants to the United States. The current gold standard for detecting multidrug-resistant tuberculosis (MDR-TB) is phenotypic drug susceptibility testing (DST), which is resource-intensive and slow, leading to increased MDR-TB transmission in the community. We evaluated multiplex allele-specific polymerase chain reaction (MAS-PCR) as a rapid molecular tool to detect MDR-TB in Panama. Based on DST, 67 MDR-TB and 31 drug-sensitive clinical isolates were identified and cultured from an archived collection. Primers were designed to target five mutation hotspots that confer resistance to the first-line drugs isoniazid and rifampin, and MAS-PCR was performed. Whole-genome sequencing confirmed DR mutations identified by MAS-PCR, and provided frequencies of genetic mutations. DNA sequencing revealed 70.1% of MDR strains to have point mutations at codon 315 of the katG gene, 19.4% within mabA-inhA promoter, and 98.5% at three hotspots within rpoB. MAS-PCR detected each of these mutations, yielding 82.8% sensitivity and 100% specificity for isoniazid resistance, and 98.4% sensitivity and 100% specificity for rifampin resistance relative to DST. The frequency of individual DR mutations among MDR strains in Panama parallels that of other TB-endemic countries. The performance of MAS-PCR suggests that it may be a relatively inexpensive and technically feasible method for rapid detection of MDR-TB in developing countries. 相似文献

To date, no single reported DNA extraction method is suitable for the efficient extraction of DNA from all fungal species. The efficiency of extraction is of particular importance in PCR-based medical diagnostic applications where the quantity of fungus in a tissue biopsy may be limited. We subjected 16 medically relevant fungi to physical, chemical and enzymatic cell wall disruption methods which constitutes the first step in extracting DNA. Examination by light microscopy showed that grinding with mortar and pestle was the most efficient means of disrupting the rigid fungal cell walls of hyphae and conidia. We then trialled several published DNA isolation protocols to ascertain the most efficient method of extraction. Optimal extraction was achieved by incorporating a lyticase and proteinase K enzymatic digestion step and adapting a DNA extraction procedure from a commercial kit (MO BIO) to generate high yields of high quality DNA from all 16 species. DNA quality was confirmed by the successful PCR amplification of the conserved region of the fungal 18S small-subunit rRNA multicopy gene. 相似文献
