We aim at finding the smallest set of genes that can ensure highly accurate classification of cancers from microarray data by using supervised machine learning algorithms. The significance of finding the minimum gene subsets is three-fold: 1) it greatly reduces the computational burden and "noise" arising from irrelevant genes. In the examples studied in this paper, finding the minimum gene subsets even allows for extraction of simple diagnostic rules which lead to accurate diagnosis without the need for any classifiers, 2) it simplifies gene expression tests to include only a very small number of genes rather than thousands of genes, which can bring down the cost for cancer testing significantly, 3) it calls for further investigation into the possible biological relationship between these small numbers of genes and cancer development and treatment. Our simple yet very effective method involves two steps. In the first step, we choose some important genes using a feature importance ranking scheme. In the second step, we test the classification capability of all simple combinations of those important genes by using a good classifier. For three "small" and "simple" data sets with two, three, and four cancer (sub)types, our approach obtained very high accuracy with only two or three genes. For a "large" and "complex" data set with 14 cancer types, we divided the whole problem into a group of binary classification problems and applied the 2-step approach to each of these binary classification problems. Through this "divide-and-conquer" approach, we obtained accuracy comparable to previously reported results but with only 28 genes rather than 16,063 genes. Age-related disease, not aging per se, causes most morbidity in older humans. Here we report that skeletal muscle respiratory uncoupling due to UCP1 expression diminishes age-related disease in three mouse models. In a longevity study, median survival was increased in UCP mice (animals with skeletal muscle-specific UCP1 expression), and lymphoma was detected less frequently in UCP female mice. In apoE null mice, a vascular disease model, diet-induced atherosclerosis was decreased in UCP animals. In agouti yellow mice, a genetic obesity model, diabetes and hypertension were reversed by induction of UCP1 in skeletal muscle. Uncoupled mice had decreased adiposity, increased temperature and metabolic rate, elevated muscle SIRT and AMP kinase, and serum characterized by increased adiponectin and decreased IGF-1 and fibrinogen. Accelerating metabolism in skeletal muscle does not appear to impact aging but may delay age-related disease. 相似文献

A series of Mn2+-doped zinc germinate ZGO:xMn2+ (x = 0–0.05) nanorods was synthesized successfully using a hydrothermal method. XRD revealed that crystal phases of the ZGO:xMn2+ were rhombohedral and in the R-3 space group. The Williamson–Hall equation was also used to explain the strain, nanocrystalline size, and stacking fault. Green LEDs were successfully fabricated by coating ZGO:Mn2+ nanorods onto UV-LED chips. For high color purity, CIE of the fabricated green LEDs were (0.2404, 0.5428), which made this material a promising candidate for fabrication of UV-based green LEDs. 相似文献

The maximum quantum yield of photosystem II (as reflected by variable to maximum chlorophyll a fluorescence, Fv/Fm) is regarded as one of the most important photosynthetic parameters. The genetic basis underlying natural variation in Fv/Fm, which shows low level of variations in plants under non‐stress conditions, is not easy to be exploited using the conventional gene cloning approaches. Thus, in order to answer this question, we have followed another strategy: we used genome‐wide association study (GWAS) and transgenic analysis in a rice mini‐core collection. We report here that four single‐nucleotide polymorphisms, located in the promoter region of β‐glucosidase 5 (BGlu‐5), are associated with observed variation in Fv/Fm. Indeed, our transgenic analysis showed a good correlation between BGlu‐5 and Fv/Fm. Thus, our work demonstrates the feasibility of using GWAS to study natural variation in Fv/Fm, suggesting that cis‐element polymorphism, affecting the BGlu‐5 expression level, may, indirectly, contribute to Fv/Fm variation in rice through the gibberellin signaling pathway. Further research is needed to understand the mechanism of our novel observation. 相似文献

Global demand to increase food production and simultaneously reduce synthetic nitrogen fertilizer inputs in agriculture are underpinning the need to intensify the use of legume crops. The symbiotic relationship that legume plants establish with nitrogen‐fixing rhizobia bacteria is central to their advantage. This plant–microbe interaction results in newly developed root organs, called nodules, where the rhizobia convert atmospheric nitrogen gas into forms of nitrogen the plant can use. However, the process of developing and maintaining nodules is resource intensive; hence, the plant tightly controls the number of nodules forming. A variety of molecular mechanisms are used to regulate nodule numbers under both favourable and stressful growing conditions, enabling the plant to conserve resources and optimize development in response to a range of circumstances. Using genetic and genomic approaches, many components acting in the regulation of nodulation have now been identified. Discovering and functionally characterizing these components can provide genetic targets and polymorphic markers that aid in the selection of superior legume cultivars and rhizobia strains that benefit agricultural sustainability and food security. This review addresses recent findings in nodulation control, presents detailed models of the molecular mechanisms driving these processes, and identifies gaps in these processes that are not yet fully explained. 相似文献

Microbially induced calcite precipitation (MICP) can reduce the permeability of soil by reducing the pore volumes. A MICP-based soil improvement method to control water leakage in irrigation channels and reservoirs built on sandy soil grounds is presented in this article. Using this method, a low-permeable hard crust can be formed at the soil surfaces. An experimental study was carried out to evaluate the effect of this method. Sandy soil samples were treated using four different schemes, namely, (1) surface spray, (2) surface spray with the addition of fibers, (3) surface spray and bulk stabilization, and (4) immersion stabilization. By applying around 2.6?L treatment liquid (consisting of ureolytic bacteria, 0.5?mol/L calcium chloride and 0.5?mol/L urea) to the top 2-cm thick soil, the seepage rates of the samples treated by the four different schemes could be reduced by up to 379 times. The conversion rates of calcium source in the tests were up to 89.7%. The results showed that a method of treating the soil in bulk before the formation of a crust on top of the soil layer was effective in reducing the seepage rates. After the bio-treatment, the formed low-permeable hard crust layer was 10 to 20?mm thick with a calcite content higher than 5%. Below the hard crusts, the calcite content was less than 5% and the soil was not properly cemented. Using the mercury intrusion test, it was found that both pore volumes and pore sizes of the bio-treated soil reduced significantly as compared with the untreated soil. Penetration tests using a flat-bottom penetrometer were used to assess the mechanical behavior of the bio-treated soil. The results indicated that the penetration resistance of the bio-treated soil layer was much higher than that of the untreated soil. 相似文献
