We studied Coleoptera emerging from deadwood in a broadleaved forest that has been managed with nature-oriented forestry since 1990 and in which an accumulation of 10m3 deadwood/ha has been achieved. We sampled a total of 11m3 of deadwood from oak and beech trees and yielded 32,477 individuals pertaining to 342 species. The number of species emerging from oak deadwood was significantly higher than that from beech. Significantly more species emerged from deadwood with medium size (15–21cm) diameter than from smaller (5–7cm) or larger (40–60cm) diameters. The stage of decay did not influence the number of species during the first 6 years of decomposition. Species richness strongly increased with the amount of deadwood sampled. Species–area regressions yielded z-values of 0.81 for oak and 0.87 for beech deadwood. The beetle assemblage was characterized by a low number of xylophagous species and a high number of mobile zoophagous species. However, two-thirds of the individuals sampled pertained to nine species of Scolytidae. This is why Shannon–Wiener diversity (H) and evenness-indices (E) greatly decreased with increasing number of scolytids. The species and individuals yielded from the sun-exposed deadwood outnumbered those from shaded areas. Namely the scolytid Taphrorychus bicolor reached remarkably high densities in the sun-exposed beech deadwood (>800 individuals/m2). In contrast, oak deadwood never harboured such high numbers of scolytids. Felling of moribund trees had decreased the number of scolytids but it had also considerably reduced the number of rare and endangered species.  相似文献

Integase interactor 1 (INI1), also known as hSNF5, is a protein that interacts with HIV-1 integrase. We report here that a cytoplasmically localized fragment of INI1 (S6; aa183-294) containing the minimal integrase-interaction domain potently inhibits HIV-1 particle production and replication. Mutations in S6 or integrase that disrupt integrase-INI1 interaction abrogated the inhibitory effect. An integrase-deficient HIV-1 transcomplemented with integrase fused to Vpr was not affected by S6. INI1 was specifically incorporated into virions and was required for efficient HIV-1 particle production. These results indicate that INI1 is required for late events in the viral life cycle, and that ectopic expression of S6 inhibits HIV-1 replication in a transdominant manner via its specific interaction with integrase within the context of Gag-Pol, providing a novel strategy to control HIV-1 replication.  相似文献

Oreochromis aureus was imported from Israel into South Africa in 1959 but data on its current status in South Africa are lacking. Genomic DNA was extracted and the COI gene amplified at the South African Institute for Aquatic Biodiversity. The identity of the sequences and specimens was determined using the Barcode of Life Data Systems and GenBank. Morphological and genetic assessment demonstrated that 11 specimens collected from two farm dams in the Eerste River System, Western Cape province, were Oreochromis aureus. A MaxEnt model compiled using global distribution, rainfall and temperature data predicted that large areas of southern Africa were climatically suitable for this species, indicating considerable invasion debt in southern Africa. As a result, surveys to assess for the extent of the invasion in South Africa and eradication of existing populations, if feasible, are recommended management actions.  相似文献

Lentiviral vectors are widely used as effective gene-delivery vehicles. Optimization of the conditions for efficient lentiviral transduction is of a high importance for a variety of research applications. Presence of positively charged polycations reduces the electrostatic repulsion forces between a negatively charged cell and an approaching enveloped lentiviral particle resulting in an increase in the transduction efficiency. Although a variety of polycations are commonly used to enhance the transduction with retroviruses, the relative effect of various types of polycations on the efficiency of transduction and on the potential bias in the determination of titer of lentiviral vectors is not fully understood. Here, we present data suggesting that DEAE-dextran provides superior results in enhancing lentiviral transduction of most tested cell lines and primary cell cultures. Specific type and source of serum affects the efficiency of transduction of target cell populations. Non-specific binding of enhanced green fluorescent protein (EGFP)-containing membrane aggregates in the presence of DEAE-dextran does not significantly affect the determination of the titer of EGFP-expressing lentiviral vectors. In conclusion, various polycations and types of sera should be tested when optimizing lentiviral transduction of target cell populations.  相似文献

Abstract

A directed PCR‐based iterative screening protocol was developed to isolate cosmids containing microsatellite markers linked to chromosomal regions of interest, such as those near the ends of linkage groups or quantitative trait loci. This method was optimized for large‐scale screening of total genomic libraries and used to purify bovine cosmids that anchor the ends of the bovine linkage group 28 (BTA28). Cosmids containing ms markers RBP3 and BMS2060 were purified for fluorescence in situ hybridization and assigned to 28ql8‐ql9 and 28ql2, respectively. These assignments indicated that approximately 73% of BTA28 (90% excluding the centromere) is covered by the current linkage map. Since this method is applicable to any target gene sequence suitable for PCR amplification, it may be extended to comparative mapping of genes.  相似文献

Colonial nesting seabirds frequently drive island ecosystem biodiversity by maintaining ecosystem functioning and community dynamics. Invasive mammal introductions to most of the world’s islands have ravaged insular seabird populations and had associated devastating ecosystem-wide effects. Eradication programs remove invasive mammals from islands, with the goal of conserving and restoring island species and systems. However, most eradication programs rely almost exclusively on passive seabird recovery to achieve these goals. Unfortunately, the life histories of most seabird species are not conducive to passive recovery within a contemporary timeframe. Seabird restoration techniques can effectively overcome life history related issues and significantly reduce recovery times for insular seabird populations, thereby reducing associated ecosystem-wide recovery times. By integrating seabird restoration and eradication programs, practitioners can maximize conservation gains, expand funding opportunities, and restore island ecosystems and the biodiversity they support.  相似文献

Previous studies have shown that human immunodeficiency virus (HIV) protease cleaves procaspase 8 to a fragment, termed Casp8p41, that lacks caspase activity but nonetheless contributes to T cell apoptosis. Herein, we show that Casp8p41 contains a domain that interacts with the BH3-binding groove of pro-apoptotic Bak to cause Bak oligomerization, Bak-mediated membrane permeabilization, and cell death. Levels of active Bak are higher in HIV-infected T cells that express Casp8p41. Conversely, targeted mutations in the Bak-interacting domain diminish Bak binding and Casp8p41-mediated cell death. Similar mutations in procaspase 8 impair the ability of HIV to kill infected T cells. These observations support a novel paradigm in which HIV converts a normal cellular constituent into a direct activator that functions like a BH3-only protein.  相似文献

We investigated the distribution pattern of litter dwelling beetles in four primeval forests of the Western Carpathians. The forests are situated in two mountain ranges and are either southerly exposed oak forests or northerly exposed beech forests. Beetles were extracted from leaf litter of plots close to coarse woody debris (c-CWD) and distant from coarse woody debris (d-CWD). We collected 2946 individuals of 172 species. Plots close to CWD usually were hot spots of species richness and beetle density, which could be increased two-fold and five-fold, respectively, when compared to d-CWD sites. The influence of CWD within each forest type was stronger than that of any other environmental factor. The c-CWD plots were characterised by both a higher presence of common species and a higher number of rare species. Especially, zoophagous and mycetophagous beetles were enhanced. The assemblage wide carrying capacity at c-CWD sites varied in respect to the gradients of temperature and moisture within a forest. A longer gradient provided a higher carrying capacity. We assume that the influence of downed deadwood on litter dwelling beetles will also be effective in commercial forests and stop the downward spiral with continuing loss of species.  相似文献

Microsatellite Evolution: Testing the Ascertainment Bias Hypothesis   总被引:5,自引:0,他引:5  
Previous studies suggest the median allele length of microsatellites is longest in the species from which the markers were derived, suggesting that an ascertainment bias was operating. We have examined whether the size distribution of microsatellite alleles between sheep and cattle is source dependent using a set of 472 microsatellites that can be amplified in both species. For those markers that were polymorphic in both species we report a significantly greater number of markers (P < 0.001) with longer median allele sizes in sheep, regardless of microsatellite origin. This finding suggests that any ascertainment bias operating during microsatellite selection is only a minor contributor to the variation observed.
