The transmission/disequilibrium test (TDT) developed by Spielman et al. can be a powerful family-based test of linkage and, in some cases, a test of association as well as linkage. It has recently been extended in several ways; these include allowance for implementation with quantitative traits, allowance for multiple alleles, and, in the case of dichotomous traits, allowance for testing in the absence of parental data. In this article, these three extensions are combined, and two procedures are developed that offer valid joint tests of linkage and (in the case of certain sibling configurations) association with quantitative traits, with use of data from siblings only, and that can accommodate biallelic or multiallelic loci. The first procedure uses a mixed-effects (i.e., random and fixed effects) analysis of variance in which sibship is the random factor, marker genotype is the fixed factor, and the continuous phenotype is the dependent variable. Covariates can easily be accommodated, and the procedure can be implemented in commonly available statistical software. The second procedure is a permutation-based procedure. 2-[4'-Maleimidylanilino]naphthalene 6-sulfonic acid (MIANS) irreversibly inactivates Na,K-ATPase in a time- and concentration-dependent manner. Inactivation is prevented by 3 mM ATP or low K(+) (<1 mM); the protective effect K(+) is reversed at higher concentrations. This biphasic effect was also observed with K(+) congeners. In contrast, Na(+) ions did not protect. MIANS inactivation disrupted high affinity ATP binding. Tryptic fragments of MIANS-labeled protein were analyzed by reversed phase high performance liquid chromatography. ATP clearly protected one major labeled peptide peak. This observation was confirmed by separation of tryptic peptides in SDS-polyacrylamide gel electrophoresis revealing a single fluorescently-labeled peptide of approximately 5 kDa. N-terminal amino acid sequencing identified the peptide (V(545)LGFCH...). This hydrophobic peptide contains only two Cys residues in all sodium pump alpha-subunit sequences and is found in the major cytoplasmic loop between M4 and M5, a region previously associated with ATP binding. Subsequent digestion of the tryptic peptide with V8 protease and N-terminal amino acid sequencing identified the modified residue as Cys(577). The cation-dependent change in reactivity of Cys(577) implies structural alterations in the ATP-binding domain following cation binding and occlusion in the intramembrane domain of Na,K-ATPase and expands our knowledge of the extent to which cation binding and occlusion are sensed in the ATP hydrolysis domain. 相似文献

The proton magnetic resonance spectra of the dihydronicotinamide ring of αNADH3 and the nicotinamide ring of αNAD+ are reported and the proton absorptions assigned. The absolute assignment of the C4 methylene protons of αNADH is based on the generation of specifically deuterium-labeled (pro-S) B-deuterio-αNADH from enzymatically prepared B-deuterio-βNADH. The C4 proton absorption of αNAD+ is assigned by oxidation of B-deuterio-αNADH by the A specific, yeast alcohol dehydrogenase to yield 4-deuterio-αNAD+.The epimerization of either αNADH or βNADH yields an equilibrium ratio of approximately 9:1 βNADH to αNADH. The rate of epimerization of αNADH to βNADH at 38 °C in 0.05, pH 7.5, phosphate buffer is 3.1 × 10?3 min?1, corresponding to a half-life of 4 hr. Four related dehydrogenases, yeast and horse liver alcohol dehydrogenase and chicken M4 and H4 lactate dehydrogenase, are shown to oxidize αNADH to αNAD+ at rates three to four orders of magnitude slower than for βNADH. By using specifically labeled B-deuterio-αNADH the enzymatic oxidation by yeast alcohol dehydrogenase has been shown to occur with the identical stereospecificity as the oxidation of βNADH. The nonenzymatic epimerization of αNADH to βNADH and the enzymatic oxidation αNADH are discussed as a possible source of αNAD+in vivo. 相似文献

The concepts of adaptive/fitness landscapes and adaptive peaks are a central part of much of contemporary evolutionary biology;
the concepts are introduced in introductory texts, developed in more detail in graduate-level treatments, and are used extensively
in papers published in the major journals in the field. The appeal of visualizing the process of evolution in terms of the
movement of populations on such landscapes is very strong; as one becomes familiar with the metaphor, one often develops the
feeling that it is possible to gain deep insights into evolution by thinking about the movement of populations on landscapes
consisting of adaptive valleys and peaks. But, since Wright first introduced the metaphor in 1932, the metaphor has been the
subject of persistent confusion, from equivocation over just what the features of the landscape are meant to represent to
how we ought to expect the landscapes to look. Recent advances—conceptual, empirical, and computational—have pointed towards
the inadequacy and indeed incoherence of the landscapes as usually pictured. I argue that attempts to reform the metaphor
are misguided; it is time to give up the pictorial metaphor of the landscape entirely and rely instead on the results of formal
modeling, however difficult such results are to understand in ‘intuitive’ terms.

The requirement of the inorganic carbon (Ci) transport system for light in cyanobacteria was investigated in Anabaena variabilis by the filtering centrifugation technique and in a mutant (E1) isolated from Anacystis nidulans using a gas exchange system. Ci transport capability increased with time of preillumination and decreased following darkening. Full activity could not be obtained by operating either photosystem II (PSII) or photosystem I alone. 3(3,4 Dichlorophenyl)-1,1 dimethylurea strongly inhibited Ci uptake. Very low activity of PSII was sufficient to activate Ci uptake. However, in the presence of dithiothreitol PSII activity was not required. We conclude that light may be required to activate as well as to energize Ci uptake in cyanobacteria. 相似文献

In acute experiments on immobilized carps, under dark adaptation conditions, studies have been made on the electrical activity of the telencephalon and midbrain tectum. It was found that in the latter photostimulation evokes local rhythmic activity which includes fast (30-50 Hz) phasic (200-300 ms) and slow (8-14 Hz) tonic (up to 3-4 s) components. High rhythm of this activity may be observed only in "healthy" preparations, coinciding with on-rhythm in the midbrain tectum. Low rhythm does not depend on the presence of rhythmic activity in the tectum. Various aspects of the development of the thalamo-telencephalic system in vertebrates are discussed. 相似文献

B-Raf, a member of the Raf family of serine/threonine kinases, is expressed primarily in the brain and in the nervous system. In this study, the biochemical properties of the B-Raf protein were investigated in nerve growth factor (NGF)-responsive cell lines and in brain tissues. B-Raf was identified by using phosphopeptide mapping analysis and cDNA analysis as a 95-kDa protein which is primarily localized in the cytosol. NGF rapidly stimulated both serine and threonine phosphorylation in vivo and autophosphorylation activity in vitro of the B-Raf protein. In PC12 cells, B-Raf autokinase activity was induced by both differentiation factors and mitogens, with maximal activity observed after 5 min of factor addition. B-Raf kinase activity was also observed following NGF treatment of SH-SY5Y neuroblastoma cells and in adult mouse brain and hippocampus. Induction of B-Raf kinase activity in NGF-treated PC12 cells required expression of kinase-active trk receptors. Exogenous substrates or a peptide containing the autophosphorylation site became phosphorylated when added to immune complex kinase assays and reduced the in vitro autophosphorylation activity of B-Raf, suggesting that in vitro autophosphorylation sites and exogenous substrates compete for active sites of the B-Raf kinase. Finally, the major in vitro autophosphorylation site of B-Raf was identified as threonine 372 in the conserved region 2 domain. A threonine residue is present at similar positions in all three mammalian Raf family members and may represent a regulatory site for these proteins. 相似文献

Recent admixture between genetically differentiated populations can result in high levels of association between alleles at loci that are <=10 cM apart. The transmission/disequilibrium test (TDT) proposed by Spielman et al. (1993) can be a powerful test of linkage between disease and marker loci in the presence of association and therefore could be a useful test of linkage in admixed populations. The degree of association between alleles at two loci depends on the differences in allele frequencies, at the two loci, in the founding populations; therefore, the choice of marker is important. For a multiallelic marker, one strategy that may improve the power of the TDT is to group marker alleles within a locus, on the basis of information about the founding populations and the admixed population, thereby collapsing the marker into one with fewer alleles. We have examined the consequences of collapsing a microsatellite into a two-allele marker, when two founding populations are assumed for the admixed population, and have found that if there is random mating in the admixed population, then typically there is a collapsing for which the power of the TDT is greater than that for the original microsatellite marker. A method is presented for finding the optimal collapsing that has minimal dependence on the disease and that uses estimates either of marker allele frequencies in the two founding populations or of marker allele frequencies in the current, admixed population and in one of the founding populations. Furthermore, this optimal collapsing is not always the collapsing with the largest difference in allele frequencies in the founding populations. Uncultivated plants growing on disturbed sites may be useful for assessing the bioavailability of some metals in soils, and thus the potential for metal mobilization up the terrestrial food chain, an important element in ecological risk assessment. A planted chicory cultivar (Cichorium intybus L. var. foliosum Hegi.) and the uncultivated plants horseweed (Canada fleabane) (Erigeron canadensis L.) and dogfennel (Eupatorium capillifolium (Lam.) Small) were evaluated for their ability to act as index plant species for soil Cd, Cr, Ni, and V at two field sites where these metals had been applied five yr previously to two highly weathered sandy Ultisols. Soil Cd was available to all analyzed plant tissues of all three plant species at both sites, particularly on the sandier Blanton soil. Chicory was an effective index plant for Cd on the finer textured Orangeburg soil but functioned as an indicator plant (toxicity symptoms were observed) on the sandier Blanton soil. Horseweed and dogfennel were effective index plants for Cd in both contaminated soils. Soil Cr, Ni, and V were less bioavailable than soil Cd and plant metal uptake was more sensitive to residual soil Cr, Ni, and V than was soil extraction with double acid. Horseweed and chicory may have potential as index plants for soil Cr. Chicory may have potential as a Ni index plant. Chicory and dogfennel may have potential as V index plants. 相似文献
