The water soluble polysaccharide produced by the green variant of tetrasporic Gigartina skottsbergii was found to be composed of D-galactose and sulfate groups in a molar ratio of 1.0:0.65. (1)H and (13)C NMR spectroscopy studies of the desulfated polysaccharide showed a major backbone structure of alternating 3-linked β-D-galactopyranosyl and 4-linked α-D-galactopyranosyl units, and minor signals ascribed to 3-O-methyl-substitution on the latter unit. Ethylation analysis of the polysaccharide indicated that the sulfate groups are mainly located at position O-2 of 4-linked α-D-galactopyranosyl residue and partially located at positions O-6 of the same unit and at position O-2 of 3-linked β-D-galactopyranosyl residue, and confirmed the presence of 3-O-methyl-galactose in minor amounts (4.4%). The sulfated d-galactan presents a similar structure to λ carrageenan but with much lower sulfation at position O-6 of the α-residue and at position O-2 of β-residue. The antioxidant capacity of the sulfated d-galactan was evaluated by the peroxyl radicals (ORAC method), hydroxyl radicals, chelating activity, and ABTS(+) assays. Kinetic results obtained in these assays were compared with those obtained for the commercial λ carrageenan. The antioxidant activity toward peroxyl radicals was higher for commercial λ carrageenan, this agrees with its higher content of sulfate group. The kinetics of the reaction of both polysaccharides with hydroxyl and ABTS(+) radicals showed a complex mechanism, but the antioxidant activity was higher for the polysaccharide from the green variant of tetrasporic Gigartina skottsbergii.  相似文献

Experimental evidence suggests that spontaneous neuronal activity may shape and be shaped by sensory experience. However, we lack information on how sensory experience modulates the underlying synaptic dynamics and how such modulation influences the response of the network to future events. Here we study whether spike-timing-dependent plasticity (STDP) can mediate sensory-induced modifications in the spontaneous dynamics of a new large-scale model of layers II, III and IV of the rodent barrel cortex. Our model incorporates significant physiological detail, including the types of neurons present, the probabilities and delays of connections, and the STDP profiles at each excitatory synapse. We stimulated the neuronal network with a protocol of repeated sensory inputs resembling those generated by the protraction-retraction motion of whiskers when rodents explore their environment, and studied the changes in network dynamics. By applying dimensionality reduction techniques to the synaptic weight space, we show that the initial spontaneous state is modified by each repetition of the stimulus and that this reverberation of the sensory experience induces long-term, structured modifications in the synaptic weight space. The post-stimulus spontaneous state encodes a memory of the stimulus presented, since a different dynamical response is observed when the network is presented with shuffled stimuli. These results suggest that repeated exposure to the same sensory experience could induce long-term circuitry modifications via 'Hebbian' STDP plasticity.  相似文献

ObjectivesTestosterone replacement therapy improves bladder capacity in urinary tract dysfunction. There is no information, however, about the role of this steroid hormone on the muscle tension of the bladder outflow region. The current study investigated the mechanisms underlying the testosterone-induced action in the pig bladder neck.MethodsUrothelium-denuded bladder neck strips were mounted in myographs for isometric force recordings and for simultaneous measurements of intracellular Ca2+ concentration ([Ca2+]i) and tension. The relaxations to testosterone, the non-aromatizable metabolite 4,5α-dihydrotestosterone (DHT) and electrical field stimulation (EFS) were carried out on phenylephrine (PhE)-precontracted strips.ResultsTestosterone and DHT evoked similar concentration-dependent relaxations only at very high pharmacological concentrations. The presence of the urothelium and the inhibition of intracellular androgenic receptor (AR), aromatase, 5α-reductase, nitric oxide (NO) synthase, guanylyl cyclase, cyclooxygenase (COX), large-, intermediate- and small-Ca2+-activated K+ channels or ATP-dependent K+ channels failed to modify the testosterone relaxations. Neuronal voltage-gated Ca2+ (VOC) channels and voltage-gated K+ (KV) channel blockers potentiated these responses. EFS evoked frequency-dependent relaxations, which were not changed by threshold concentrations of testosterone. In Ca2+-free potassium rich physiological saline solution, testosterone inhibited the contractions induced by CaCl2 and the L-type VOC channel activator (±)-BAY K 8644. Relaxations elicited by testosterone were accompanied by simultaneous decreases in smooth muscle [Ca2+]i.ConclusionsTestosterone produces relaxation of the pig urinary bladder neck through mechanisms independent of urothelium, AR, aromatase, 5α-reductase, NO synthase, guanylyl cyclase, COX and K+ channels. Testosterone-induced relaxation is produced via the inhibition of the extracellular Ca2+ entry through L-type VOC channels.  相似文献

Owl monkey kidney cell cultures yielded a viral agent 32 days after initiation of the culture. The virus was identified as a herpesvirus by physico-chemical, cultural, and morphological features. Serologically this herpesvirus was found to be unrelated to other members of this family, as well as to Herpesvirus aotus type 1 and 2. Based on these findings the name Herpesvirus aotus type 3 is suggested for this herpesvirus.  相似文献

Motility is a key trait for rhizosphere colonization by Pseudomonas fluorescens. Mutants with reduced motility are poor competitors, and hypermotile, more competitive phenotypic variants are selected in the rhizosphere. Flagellar motility is a feature associated to planktonic, free‐living single cells, and although it is necessary for the initial steps of biofilm formation, bacteria in biofilm lack flagella. To test the correlation between biofilm formation and rhizosphere colonization, we have used P. fluorescens F113 hypermotile derivatives and mutants affected in regulatory genes which in other bacteria modulate biofilm development, namely gacS (G), sadB (S) and wspR (W). Mutants affected in these three genes and a hypermotile variant (V35) isolated from the rhizosphere were impaired in biofilm formation on abiotic surfaces, but colonized the alfalfa root apex as efficiently as the wild‐type strain, indicating that biofilm formation on abiotic surfaces and rhizosphere colonization follow different regulatory pathways in P. fluorescens. Furthermore, a triple mutant gacSsadBwspR (GSW) and V35 were more competitive than the wild‐type strain for root‐tip colonization, suggesting that motility is more relevant in this environment than the ability to form biofilms on abiotic surfaces. Microscopy showed the same root colonization pattern for P. fluorescens F113 and all the derivatives: extensive microcolonies, apparently held to the rhizoplane by a mucigel that seems to be plant produced. Therefore, the ability to form biofilms on abiotic surfaces does not necessarily correlates with efficient rhizosphere colonization or competitive colonization.  相似文献
