The position of the gametophytic bud was examined in relationto the development of protonema in the moss, Physcomitrium sphaericum. Positions of protrusion formation, of the development of protrusionsinto lateral filaments, and of the differentiation of protrusionsinto buds are restricted within the narrow regions of the filaments.The number of cells from the apical cell of the filament tothese positions are constant in any size filament. The growth pattern of the protonema is shown as follow. As afilament grows one-dimensionally through divisions of the apicalcell, new protrusions are produced successively on the 5th cellfrom the apical cell or on its vicinity. The cells which intervenebetween the apical cell and this protrusion increase in numberas the apical cell divides. When this protrusion is positionedat the 8th or 9th cell from the apex, it differentiates intoa bud or a lateral filament. This growth pattern is common toboth the main and lateral filaments. Buds are differentiated not only on caulonema cells in the mainand lateral filament, but also on chloronema cells at the baseof the lateral filaments. Poly(A)-positive mRNA extracted from tonsillar mononuclear cells stimulated with phytohemagglutinin-M and 12-o-tetradecanoyl phorbol 13-acetate was successfully translated into biologically active interleukin 2 (IL-2) in Xenopus laevis oocytes, and secreted into the incubation medium. In control experiments, the extract of oocytes injected with either poly(A)-negative RNA or buffer did not show any IL-2 activity. We developed a simple method for freezing anchorage-dependent cells, including primary cultured rat liver parenchymal cells, without detaching the cells from the culture dish. The method consists of preculture of the cells to confluence, changing the growth medium to a conventional freezing medium, packaging in a container, and storage at –80°C. We have established a novel cell line from a Vero cell derivative that is useful for expression of exogenous genes and protein production. Parental Vero-317 cells can grow in biotin-containing Eagle's MEM without supplements. By transforming this cell line with replication origin-defective SV40 DNA, which contains a temperature-sensitive tsA58 large T antigen gene, we established the Verots S3 cell line that amplified a SV40-origin containing plasmid. The cell line expressed a human growth hormone (hGH) gene insert with higher efficiency than COS-7 cells in 5% serum-containing MEM and could grow and continue hGH expression in protein-free MEM. However, temperature-sensitive shut down of hGH production was observed not immediately but 3 days after the temperature shift from 33°C to 39.5°C. 相似文献

Puf5, a Puf-family RNA-binding protein, binds to 3´ untranslated region of target mRNAs and negatively regulates their expression in Saccharomyces cerevisiae. The puf5Δ mutant shows pleiotropic phenotypes including a weakened cell wall, a temperature-sensitive growth, and a shorter lifespan. To further analyze a role of Puf5 in cell growth, we searched for a multicopy suppressor of the temperature-sensitive growth of the puf5Δ mutant in this study. We found that overexpression of CLB2 encoding B-type cyclin suppressed the temperature-sensitive growth of the puf5Δ mutant. The puf5Δ clb2Δ double mutant displayed a severe growth defect, suggesting that Puf5 positively regulates the expression of a redundant factor with Clb2 in cell cycle progression. We found that expression of CLB1 encoding a redundant B-type cyclin was decreased in the puf5Δ mutant, and that this decrease of the CLB1 expression contributed to the growth defect of the puf5Δ clb2Δ double mutant. Since Puf5 is a negative regulator of the gene expression, we hypothesized that Puf5 negatively regulates the expression of a factor that represses CLB1 expression. We found such a repressor, Ixr1, which is an HMGB (High Mobility Group box B) protein. Deletion of IXR1 restored the decreased expression of CLB1 caused by the puf5Δ mutation and suppressed the growth defect of the puf5Δ clb2Δ double mutant. The expression of IXR1 was negatively regulated by Puf5 in an IXR1 3´ UTR-dependent manner. Our results suggest that IXR1 mRNA is a physiologically important target of Puf5, and that Puf5 and Ixr1 contribute to the cell cycle progression through the regulation of the cell cycle-specific expression of CLB1. 相似文献

Measuring mechanical parameters in tissues, such as the elastic modulus of cell-cell junctions, is essential to decipher the mechanical control of morphogenesis. However, their in vivo measurement is technically challenging. Here, we formulated an image-based statistical approach to estimate the mechanical parameters of epithelial cells. Candidate mechanical models are constructed based on force-cell shape correlations obtained from image data. Substitution of the model functions into force-balance equations at the cell vertex leads to an equation with respect to the parameters of the model, by which one can estimate the parameter values using a least-squares method. A test using synthetic data confirmed the accuracy of parameter estimation and model selection. By applying this method to Drosophila epithelial tissues, we found that the magnitude and orientation of feedback between the junction tension and shrinkage, which are determined by the spring constant of the junction, were correlated with the elevation of tension and myosin-II on shrinking junctions during cell rearrangement. Further, this method clarified how alterations in tissue polarity and stretching affect the anisotropy in tension parameters. Thus, our method provides a novel approach to uncovering the mechanisms governing epithelial morphogenesis. 相似文献

The effects of local population density, sex morph [protogynous (PG) or protandrous (PA)], and individual tree size on the
demographic processes of seed production were investigated in a heterodichogamous maple, Acer mono Maxim. var. Marmoratum (Nichols.) Hara f. dissectum, in a temperate forest of Japan. As the distance from conspecific reproductive adults increased, the percentage of immature
seed fall and empty seeds increased significantly, indicating higher pollination success along with local population density.
Although the difference was not distinct, pollination success was affected by the local population density of the reciprocal
sex morph rather than that of both sex morphs. The trees at higher local population density sites suffered higher seed mortality
due to predation and decay, and tended to produce smaller seeds. Thus, the impacts of local population density operated both
positively and negatively on reproduction. As a factor of individual traits, tree size scarcely affected any demographic processes.
On the other hand, sex morph did affect pollination success. Trees of PG type had lower immature seed fall than those of PA
type, suggesting that the former has higher efficiency of pollen acceptance than the latter. The results on seed demography
presented here partly support previous suggestions that heterodichogamous plants exhibit reciprocal cross-pollination and
gender specialization as reproductive traits. 相似文献
