IL-7 promotes thymocyte proliferation and maintains immunocompetent thymocytes bearing alpha beta or gamma delta T-cell receptors in vitro: synergism with IL-2

IL-7 induced the proliferation of normal thymocytes and the effect was synergistically potentiated by a small dose of IL-2, which by itself hardly affected thymocyte proliferation. No synergism was observed between IL-7 and any one of the other lymphokines including IL-1, IL-3, and IL-4. The thymocyte culture stimulated with IL-7 and IL-2 consisted of single positive (CD4+CD8- and CD4-CD8+) and double negative (CD4-CD8-) populations, and double positive (CD4+CD8+) cells were completely deleted. Both single positive and double negative thymocytes expressed CD3, but only the former exhibited V beta 8 and V beta 6 in an expected proportion (approximately 30% in BALB/c mice) and the latter none at all. Immunoprecipitation of the cultured thymocytes by anti-TCR gamma antibody, on the other hand, revealed the presence of a TCR gamma chain. Taken together, these results indicated that the thymocyte cultured with IL-7 and IL-2 consisted of mature T cells bearing alpha beta or gamma delta TCR. Experiments using preselected thymocyte subpopulations indicated that double negative cells responded to both IL-7 and IL-2 with positive synergism when combined, while thymocytes enriched for single positive cells preferentially responded to IL-7 with little response to IL-2 and no detectable synergism. Double positive thymocytes showed no proliferation in response to IL-7 and IL-2. In contrast to single positive thymocytes, splenic T cells hardly responded to IL-7, although significant proliferation was induced in the presence of a low dose of IL-2. Thymocytes cultured with IL-7 and IL-2 showed little nonspecific cytotoxic activity, but responded to Con A or alloantigen, whereas those stimulated with a high dose of IL-2 alone exhibited potent cytotoxic activity. These results indicated that IL-7 was involved in the generation of immunocompetent T cells in the thymus in concert with IL-2.     

Three fluorescent probes for the flow-cytometric assessment of membrane potential inSaccharomyces cerevisiœ

Three fluorescent probes, tetramethyl rhodamine ethyl ester (TMRE), 3,3′-dipropylthiacarbocyanine iodide (diS-C₃(3)) and 3,3′-dipropyloxacarbocyanine iodide (diO-C₃(3)), were tested for their suitability as fluorescent indicators of membrane potential inSaccharomyces cerevisiœ in studies performed by flow cytometry. For all these dyes the intensity of fluorescence of stained cells increased with probe concentration in the range of 60–3000 nmol/L. The optimum staining period was 15–20 min for diS-C₃(3). Depolarization of cells by increased extracellular potassium level and by valinomycin elicited with all probes a drop in fluorescence intensity. In some yeast batches this depolarization was accompanied by a separation of subpopulations with different fluorescence properties.     

Letters to the metanarrative: Totalitarian “enjoyment” and self representation in postcommunist Serbia

Conclusion This outline of the instrumentality of the technology/technique of representation in postcommunist Serbia is but a brief attempt to address the modes of production of identity in postmodernism, its legitimation in/of dominant discourse, and their embedding in common sense language and practice. Postcommunist Serbia is obviously a special situation:²³ a particular case of the social appropriation and construction of enjoyment and desire, intimately related to the desires and various enjoyments of the members of the community, precarious in its collective affirmation. Based on the idea of contingent collectivity, faction-fiction, in its meta-critique seems to signal the emergence of a new régime in the relation between historical reality and its representation in historical narratives.This new régime, obviously, cannot be exhaustively (or even extensively) discussed in a brief paper like this one. Nevertheless, some (three) points of departure for the general discussion of postmodern modes of representation are still possible. One such point of departure is the contingence of representation, another the apparent paradigmatic similarity between trash talk-shows and Echoes and Reactions. Oxidation and amidation of salicylate by Streptomyces species

Seven streptomycete strains were tested for biotransformation of salicylate. The products were identified by nuclear magnetic resonance spectroscopy and three types of conversion were found. Streptomyces cinnamonensis and Streptomyces spectabilis formed gentisate and salicylamide concurrently. Streptomyces rimosus transformed salicylate to salicylamide. Streptomyces lividans, Streptomyces coelicolor, Streptomyces griseus and Streptomyces avermitilis produced only gentisate. Time course studies of salicylate conversion by thin-layer chromatography and high pressure liquid chromatography showed that salicylamide was accumulated in the culture broth, whereas gentisate was further metabolized.Key words: salicylate, gentisate, salicylamide, biotransformation, Streptomyces spp.     

Variance deficit is not reliable evidence for niche limitation

Variance deficit in the number of species per sampling unit, as compared to a null model with species occurring independently of each other has been considered by various authors to be evidence for niche limitation. I show that simpler mechanisms could explain this phenomenon. Of those, the competition of individuals for space limits the number of individuals per sampling unit, resulting in decrease of variance in species richness. Testing the proportions of species belonging to various guilds seems to be more ecologically interesting, although here also the variance deficit can hardly be taken as reliable evidence for niche limitation.
