A novel series of tryptase inhibitors with a N-phenylphthalimide skeleton structurally derived from thalidomide (1) has been developed. Structure–activity relationship studies led to a potent and selective tryptase inhibitor, 2-(4-cyanophenyl)isoindole-1,3-dione-5-yl 3-(2-aminopyridin-5-yl)propanoate (7), with the IC50 value of 78 nM.  相似文献

Cells with a genetic defect affecting a biological activity and/or a cell phenotype are generally called "cell mutants" and are a highly useful tool in genetic, biochemical, as well as cell biological research. To investigate peroxisome biogenesis and human peroxisome biogenesis disorders, more than a dozen complementation groups of Chinese hamster ovary (CHO) cell mutants defective in peroxisome assembly have been successfully isolated and established as a model system. Moreover, successful PEX gene cloning studies by taking advantage of rapid functional complementation assay of CHO cell mutants invaluably contributed to the accomplishment of isolation of pathogenic genes responsible for peroxisome biogenesis diseases. Molecular mechanisms of peroxisome assembly are currently investigated by making use of such mammalian cell mutants.  相似文献

To examine species composition and population structures in sand lance (Ammodytidae) along the northern Pacific coast of Japan, genetic analysis were carried out for specimens collected in 2014 from Otsuchi Bay, Iwate, Ishinomaki Bay, Miyagi, off Soma, Fukushima and Ise–Mikawa Bays, Aichi. The samples consisted of Ammodytes japonicus and Ammodytes heian, of which the latter is a recently described species. Neither species exhibited significant genetic differences among localities. Only A. japonicus was found in the most southern locality at Aichi, but it decreased northward to <90% in Miyagi and Fukushima and the two species occurred almost evenly in Iwate suggesting a latitudinal cline in their species composition along the northern Pacific coast of Japan, off Tohoku. The vertebral counts differed between A. japonicus and A. heian with modes of 65 and 63, respectively, but this characteristic did not differ significantly within a locality (Iwate). This suggests that the vertebral counts of Ammodytes spp. in Japanese waters are probably strongly determined by the environment than by a species‐specific genetic trait.  相似文献

This study was performed to determine the effects of Arctium lappa (Al) to protect against cadmium damage in the rat liver. Male rats received a single i.p. dose of CdCl2 (1.2 mg/kg body weight (BW)) with or without Al extract administered daily by gavage (300 mg/kg BW) for 7 or 56 days. After 7 days, Al caused plasma transaminase activity to diminish in groups Al (glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT)) and CdAl (GPT). After 56 days, GOT and GPT plasma activities were reduced in the Cd group. No alteration in plasma levels of creatinine, total bilirubin, and total protein were observed. GOT liver activity increased in the Cd group. No alteration was observed in superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), and malondialdehyde (MDA) dosage. In the Cd group, hepatocyte proportion decreased and sinusoid capillary proportion increased. In the Al and CdAl groups, the nuclear proportion increased and the cytoplasmic proportion decreased. The hepatocyte nucleus density reduced in Cd and increased in the Al group. After 56 days, there was no alteration in the Cd group. In Al and CdAl groups, the nuclear proportion increased without cytoplasmic proportion variation, but the sinusoid capillary proportion was reduced. The hepatocyte nucleus density decreased in the Cd group and increased in the Al and CdAl groups. In conclusion, the liver function indicators showed that A. lappa protected the liver against cadmium toxicity damage.  相似文献

Protein phosphatases are signalling molecules that regulate a variety of fundamental cellular processes including cell growth, metabolism and apoptosis. The aim of this work was to correlate the cytotoxicity of pervanadate and okadaic acid on HL60 cells and their effect on the phosphatase obtained from these cells. The cytotoxicity of these protein phosphatase inhibitors was evaluated on HL60 cells using phosphatase activity, protein quantification and MTT reduction as indices. The major phosphatase presents in the cellular extract showed high activity (80%) and affinity (Km=0.08?mM) to tyrosine phosphate in relation to p-nitrophenyl phosphate (pNPP)—(Km=0.51?mM). Total phosphatase (pNPP) was inhibited in the presence of 10?mM vanadate (98%), 200?μM pervanadate (95%) and 100?μM p-chloromercuribenzoate (80%) but okadaic acid caused a slight increase in enzyme activity (25%). When the HL60 cells were treated with the phosphatase inhibitors (pervanadate and okadaic acid) for 24?hours, only 20% residual activity was observed in presence of 200?μM pervanadate, whereas in the presence of okadaic acid this inhibitory effect was not observed. However, in respect to mitochondrial function, cell viability decreased about 80% in the presence of 100?nM okadaic acid. The total protein content was decreased 25% when the cells were treated with 100?nM okadaic acid in combination with 200?μM pervanadate. Our results suggest that both phosphatase inhibitors presented different mechanisms of action on HL60 cells. However, their effect on the cell redox status have to be considered.  相似文献

After analyzing all the collection data for larvae of the Japanese eel, Anguilla japonica, in the western North Pacific, we found that the spawning site of this species appears to be near three seamounts in the West Mariana Ridge, 2000–3000km away from their freshwater habitats. These seamounts are located in the westward flow of the North Equatorial Current and are hypothesized to provide cues for migrating silver eels and to serve as possible aggregation sites for spawning. Back-calculated birth dates based on otolith microstructure of leptocephali indicate that the Japanese eel does not spawn continuously during the long spawning season from April to November, but is synchronized to spawn periodically once a month during new moon. This lunar periodicity of spawning and the seamount spawning hypothesis are new developments in the millennium-old mystery of eel spawning that has fascinated naturalists since the time of Aristotle.  相似文献

An anticoagulant sulfated galactan isolated from the marine green alga, Codium cylindricum, was shown to have antiangiogeinic activity. This galactan suppressed microvessel formation in an ex vivo serum-free matrix culture model using rat aortic ring. It also inhibited human umbilical vein endothelial cells (HUVEC) tube formation on reconstituted basement membrane gel. These results show the value of algal sulfated galactans in the design of antiangiogenic agents.  相似文献

Previous research has suggested that enhanced biological phosphorus removal (EBPR) from wastewater can be achieved under continuous aerobic conditions over the short term. However, little is known how environmental conditions might affect aerobic EBPR performance. Consequently we have investigated the impact of temperature, pH and dissolved oxygen (DO) concentrations on EBPR performance under strictly aerobic conditions. A sequencing batch reactor (SBR) was operated for 108 days on a six-hour cycle (four cycles a day). The SBR ran under alternating anaerobic-aerobic conditions as standard and then operated under strictly aerobic conditions for one cycle every three or four days. SBR operational temperature (10, 15, 20, 25 and 30°C), pH (6, 7, 8 and 9) and DO concentration (0.5, 2.0 and 3.5mg/L) were changed consecutively during the aerobic cycle. Recorded increases in mixed liquor phosphorus (P) concentrations during aerobic carbon source uptake (P release) were affected by the biomass P content rather than the imposed changes in the operational conditions. Thus, P release levels increased with biomass P content. By contrast, subsequent aerobic P assimilation (P uptake) levels were both affected by changes in operational temperature and pH, and peaked at 20-25°C and pH 7-8. Highest P uptake detected under these SBR operating conditions was 15.4 mg Pg-MLSS(-1) (at 25°C, pH 7 and DO 2.0mg/L). The ability of the community for linked aerobic P release and P uptake required the presence of acetate in the medium, a finding which differs from previous data, where these are reported to occur in the absence of any exogenous carbon source. Fluorescence in situ hybridization was performed on samples collected from the SBR, and Candidatus 'Accumulibacter phosphatis' cells were detected with PAOmix probes through the operational periods. Thus, Candidatus 'Accumulibacter phosphatis' seemed to perform P removal in the SBR as shown in previous studies on P removal under strictly aerobic conditions.  相似文献
