CD44 is an adhesion molecule that serves as a cell surface receptor for several extracellular matrix components, including hyaluronan (HA). The proteolytic cleavage of CD44 from the cell surface plays a critical role in the migration of tumor cells. Although this cleavage can be induced by certain stimuli such as phorbol ester and anti-CD44 antibodies in vitro, the physiological inducer of CD44 cleavage in vivo is unknown. Here, we demonstrate that HA oligosaccharides of a specific size range induce CD44 cleavage from tumor cells. Fragmented HA containing 6-mers to 14-mers enhanced CD44 cleavage dose-dependently by interacting with CD44, whereas a large polymer HA failed to enhance CD44 cleavage, although it bound to CD44. Examination using uniformly sized HA oligosaccharides revealed that HAs smaller than 36 kDa significantly enhanced CD44 cleavage. In particular, the 6.9-kDa HA (36-mers) not only enhanced CD44 cleavage but also promoted tumor cell motility, which was completely inhibited by an anti-CD44 monoclonal antibody. These results raise the possibility that small HA oligosaccharides, which are known to occur in various tumor tissues, promote tumor invasion by enhancing the tumor cell motility that may be driven by CD44 cleavage. 相似文献

We investigated the occurrence of patchy stomatal behavior in leaves of saplings and a forest canopy tree of Quercus crispula Blume. Through a combination of leaf gas-exchange measurements and numerical simulation, we detected patterns of stomatal
closure (either uniform or patchy bimodal) coupled with depression of net assimilation rate (A). There was a clear inhibition of A associated with stomatal closure in leaves of Q. crispula during the day, but the magnitude of inhibition varied among days and growing conditions. Aboveground forest structure, biomass, and primary productivity in a tropical heath forest in Central Kalimantan (Indonesian Borneo) were examined using data from 1-ha plots and stand-level allometric equations developed from harvested tree samples. The study site experienced a severe drought in 1997–1998 associated with the El Niño Southern Oscillation event. The drought effect on heath forest productivity was also assessed by evaluating changes in wood mass increment rates. Allometric relationships suggested that heath forest trees had leaves with smaller specific leaf area (SLA), and large heath forest trees allocate more to leaf mass compared to mixed dipterocarp forest trees. Aboveground biomass (for trees ≥ 4.8 cm DBH) in two 1-ha plots, P1 and P4, totaled 244.8 and 232.0 Mg/ha. Aboveground wood mass increment rate was –0.1 and 4.7 Mg/ha/yr in P1 and P4 during the drought period (from February to August 1998), while it quickly recovered to 8.1 and 8.5 Mg/ha/yr during the post-drought period (from August 1998 to August 1999 for P1 and from August 1998 to November 1999 for P4). This suggests a severe impact of the drought on heath forest productivity. Leaf characteristics of heath forest such as small SLA and long-lived leaves probably play a significant role in effective assimilation and maintenance of heath forest productivity under stressful conditions. 相似文献

The bioorganic synthesis of an end-capped anti-HIV peptide from a recombinant protein was investigated. Cyanogen bromide-mediated cleavage of two Met-Gln sites across the target anti-HIV sequence generated an HIV-1 fusion inhibitor (SC35EK) analog bearing an N-terminal pyroglutamate (pGlu) residue and a C-terminal homoserine lactone (Hsl) residue. The end-capped peptide, pGlu-SC35EK-Hsl, had similar bioactivity and biophysical properties to the parent peptide, and an improved resistance to peptidase-mediated degradation was observed compared with the non-end-capped peptide obtained using standard recombinant technology. 相似文献
