Marek's disease virus (MDV) causes a general malaise in chickens that is mostly characterized by the development of lymphoblastoid tumors in multiple organs. The use of bacterial artificial chromosomes (BACs) for cloning and manipulation of the MDV genome has facilitated characterization of specific genes and genomic regions. The development of most MDV BACs, including pRB-1B-5, derived from a very virulent MDV strain, involved replacement of the US2 gene with mini-F vector sequences. However, when reconstituted viruses based on pRB-1B were used in pathogenicity studies, it was discovered that contact chickens housed together with experimentally infected chickens did not contract Marek's disease (MD), indicating a lack of horizontal transmission. Staining of feather follicle epithelial cells in the skins of infected chickens showed that virus was present but was unable to be released and/or infect susceptible chickens. Restoration of US2 and removal of mini-F sequences within viral RB-1B did not alter this characteristic, although in vivo viremia levels were increased significantly. Sequence analyses of pRB-1B revealed that the UL13, UL44, and US6 genes encoding the UL13 serine/threonine protein kinase, glycoprotein C (gC), and gD, respectively, harbored frameshift mutations. These mutations were repaired individually, or in combination, using two-step Red mutagenesis. Reconstituted viruses were tested for replication, MD incidence, and their abilities to horizontally spread to contact chickens. The experiments clearly showed that US2, UL13, and gC in combination are essential for horizontal transmission of MDV and that none of the genes alone is able to restore this phenotype.  相似文献

Potato late blight, caused by the oomycete pathogen Phytophthora infestans, significantly hampers potato production. Recently, a new Resistance to Phytophthora infestans (Rpi) gene, Rpi-amr1, was cloned from a wild Solanum species, Solanum americanum. Identification of the corresponding recognized effector (Avirulence or Avr) genes from P. infestans is key to elucidating their naturally occurring sequence variation, which in turn informs the potential durability of the cognate late blight resistance. To identify the P. infestans effector recognized by Rpi-amr1, we screened available RXLR effector libraries and used long read and cDNA pathogen-enrichment sequencing (PenSeq) on four P. infestans isolates to explore the untested effectors. Using single-molecule real-time sequencing (SMRT) and cDNA PenSeq, we identified 47 highly expressed effectors from P. infestans, including PITG_07569, which triggers a highly specific cell death response when transiently coexpressed with Rpi-amr1 in Nicotiana benthamiana, suggesting that PITG_07569 is Avramr1. Here we demonstrate that long read and cDNA PenSeq enables the identification of full-length RXLR effector families and their expression profile. This study has revealed key insights into the evolution and polymorphism of a complex RXLR effector family that is associated with the recognition by Rpi-amr1.  相似文献

Patterns in the natural abundance of nitrogen (N) isotopes (1?N and 1?N) can help in the understanding of ecosystem processes along environmental gradients, because some processes fractionate against the heavier isotope. We measured δ1?N in many components of the Franz Josef soil chronosequence in New Zealand to see how each component varied along the sequence and within sites, and to see what this variation can tell us about how ecosystem processes such as N losses change with soil age. We analyzed δ1?N in foliage from 18 woody species, abscised leaves from seven woody species, three soil horizons, bryophytes, lichens, bulk deposition, and nodules from the N-fixing tree Coriaria arborea (Coriariaceae). Foliar δ1?N varied significantly across plant species. Foliage and bulk litter became 1?N-depleted as soil age increased. Soil N from organic and mineral horizons was significantly more 1?N-enriched than bulk litter N at each site. Increasing precipitation also decreased foliar and soil δ1?N. Comparing input and whole ecosystem δ1?N revealed limited evidence for net fractionation during N losses. These trends are consistent with some combination of increasing fractionation during plant N uptake, mycorrhizal transfer, within-plant processing, and soil decomposition as soils age.  相似文献

(1) The ATPase inhibitor protein has been isolated from rat liver mitochondria in purified form. The molecular weight determined by sodium dodecyl sulfate gel electrophoresis is approximately 9500, and the isoelectric point is 8.9.

(2) The protein inhibits both the soluble ATPase and the particle-bound ATPase from rat liver mitochondria. It also inhibits ATPase activities of soluble F1, and inhibitor-depleted submitochondrial particles derived from bovine heart mitochondria.

(3) On particle-bound ATPase the inhibitor has its maximal effect if incubated in the presence of Mg2+. ATP at slightly acidic pH.

(4) The inhibitor has a minimal effect on Pi-ATP exchange activity in sonicated submitochondrial particles. However, unexpectedly the inhibitor greatly stimulates Pi-ATP exchange activity in whole mitochondria while the low ATPase activity of the mitochondria is not affected. The possible mechanism of action of the inhibitor on intact mitochondria is offered.  相似文献

Causative agents of Lyme disease and relapsing fever, including Borrelia burgdorferi and Borrelia hermsii, respectively, are unusual among bacteria in that they possess a segmented genome with linear DNA molecules terminated by hairpin ends, known as telomeres. During replication, these telomeres are processed by the essential telomere resolvase, ResT, in a unique biochemical reaction known as telomere resolution. In this study, we report the identification of the B. hermsii resT gene through cross-species hybridization. Sequence comparison of the B. hermsii protein with the B. burgdorferi orthologue revealed 67% identity, including all the regions currently known to be crucial for telomere resolution. In vitro studies, however, indicated that B. hermsii ResT was unable to process a replicated B. burgdorferi type 2 telomere substrate. In contrast, in vivo cross-species complementation in which the native resT gene of B. burgdorferi was replaced with B. hermsii resT had no discernible effect, even though B. burgdorferi strain B31 carries at least two type 2 telomere ends. The B. burgdorferi ResT protein was also able to process two telomere spacing mutants in vivo that were unresolvable in vitro. The unexpected differential telomere processing in vivo versus in vitro by the two telomere resolvases suggests the presence of one or more accessory factors in vivo that are normally involved in the reaction. Our current results are also expected to facilitate further studies into ResT structure and function, including possible interaction with other Borrelia proteins.  相似文献

In the present study we examined the effect of seminal plasma (SP) on angiogenesis in the porcine oviduct. Gene expressions of vascular endothelial growth factor (VEGF) and its two receptors (Flt-1: fms-like tyrosine kinase and Flk-1/KDR: fetal liver kinase-1/kinase insert domain-containing receptor) as well as fibroblast growth factors (FGF-1 and 2) and von Willenbrand factor (VWF) were determined in the oviduct of SP-treated and control (PBS-treated) gilts. Moreover, vascular density (VD) indicated by endothelial cell area immunolocalized by VWF staining, was assessed in the oviducts. Real-time PCR revealed significantly higher expression of FGF-2 and VWF on day 1 (p < 0.05) after SP administration in comparison to control animals. In contrast, Flt-1 mRNA level on day 1 was lower in SP-treated gilts compared to controls (p < 0.05). In the examined oviductal sections, VD did not differ between control and SP-treated animals. However, in SP-treated animals VD was higher on day 5 than on day 1 (p < 0.05) or 3 (p < 0.01). SP had no significant effect on VEGF, Flk-1/KDR and FGF-1 mRNA expression. In conclusion, our results suggest that SP affects the vascular network by changing the expression of factors contributing to angiogenesis.  相似文献

Isolated peritoneal membrane, freshly harvested from pigs, was studied in vitro to determine the permeability of the membrane to saccharide nutrients. Studies were performed with glucose and sucrose at various transmembrane gradients ranging from 150 to 600 mosm/liter (mmol/liter) and with polysaccharide at a 300 mosm/liter gradient. Findings indicate that flux of these nutrients across the isolated peritoneum is related to molecular weight and that sufficient kilocalories will traverse the membrane to provide for adequate carbohydrate nutrition (calculated to deliver more than 3000 kcal/day at gradients of 150 mosm/liter). Polysaccharide flux studies suggest that carbohydrate calories may be provided in the form of complex molecules and not only in the form of glucose. Studies of 1:1 serum and polysaccharide mixtures showed the conversion of polysaccharide to glucose by serum in vitro. When rats were given intraperitoneal polysaccharide their blood glucose levels rose promptly although they remained fasting, suggesting an in vivo conversion also occurs. These findings provide support for use of polysaccharides intraperitoneally as means to reduce the osmolarity of fluid administered for nutritional support; this would increase tolerance while still providing ample calories. The in vitro apparatus used in these studies provides a mechanism to study individual nutrients and yields highly reproducible results. These studies add further evidence that intraperitoneal nutrition is feasible.  相似文献

Glutaredoxin1 (GRX1) is a glutathione (GSH)-dependent thiol oxidoreductase. The GRX1/GSH system is important for the protection of proteins from oxidative damage and in the regulation of protein function. Previously we demonstrated that GRX1/GSH regulates the activity of the essential copper-transporting P1B-Type ATPases (ATP7A, ATP7B) in a copper-responsive manner. It has also been established that GRX1 binds copper with high affinity and regulates the redox chemistry of the metallochaperone ATOX1, which delivers copper to the copper-ATPases. In this study, to further define the role of GRX1 in copper homeostasis, we examined the effects of manipulating GRX1 expression on copper homeostasis and cell survival in mouse embryonic fibroblasts and in human neuroblastoma cells (SH-SY5Y). GRX1 knockout led to cellular copper retention (especially when cultured with elevated copper) and reduced copper tolerance, while in GRX1-overexpressing cells challenged with elevated copper, there was a reduction in both intracellular copper levels and copper-induced reactive oxygen species, coupled with enhanced cell proliferation. These effects are consistent with a role for GRX1 in regulating ATP7A-mediated copper export, and further support a new function for GRX1 in neuronal copper homeostasis and in protection from copper-mediated oxidative injury.  相似文献

In this work, the binding mechanism of new Polyketide Synthase 13 (Pks13) inhibitors has been studied through molecular dynamics simulation and free energy calculations. The drug Tam1 and its analogs, belonging to the benzofuran class, were submitted to 100 ns simulations, and according to the results obtained for root mean square deviation, all the simulations converged from approximately 30 ns. For the analysis of backbone flotation, the root mean square fluctuations were plotted for the Cα atoms; analysis revealed that the greatest fluctuation occurred in the residues that are part of the protein lid domain. The binding free energy value (ΔGbind) obtained for the Tam16 lead molecule was of ?51.43 kcal/mol. When comparing this result with the ΔGbind values for the remaining analogs, the drug Tam16 was found to be the highest ranked: this result is in agreement with the experimental results obtained by Aggarwal and collaborators, where it was verified that the IC50 for Tam16 is the smallest necessary to inhibit the Pks13 (IC50 = 0.19 μM). The energy decomposition analysis suggested that the residues which most interact with inhibitors are: Ser1636, Tyr1637, Asn1640, Ala1667, Phe1670, and Tyr1674, from which the greatest energy contribution to Phe1670 was particularly notable. For the lead molecule Tam16, a hydrogen bond with the hydroxyl of the phenol not observed in the other analogs induced a more stable molecular structure. Aggarwal and colleagues reported this hydrogen bonding as being responsible for the stability of the molecule, optimizing its physic-chemical, toxicological, and pharmacokinetic properties.  相似文献

1. Collaboration means actively working together to achieve things which could not be done alone. This article attempts to provide an overall, unified, guiding framework for collaboration in freshwater ecology by discussing aspects of collaboration at individual and organizational levels, and addressing international linkages.
2. The essential elements of collaboration are communication and trust, and effective project management. Barriers to effective collaboration include competition, organizational cultures and organizational instabilities.
3. The success of collaboration can be measured by tangible benefits such as increased numbers of peer-reviewed publications, the production of working models and a number of intangible benefits.
4. Interactions between individuals lie at the heart of an effective collaboration; organizational arrangements should facilitate this interaction. Some governments are encouraging collaboration to increase cost efficiency and allocate accountability. This trend should continue on an international level.
