Abstract. The biodiversity of species‐rich semi‐natural meadows is declining across Europe due to ceased management. In this study we aimed to find out how successfully the local species richness of an overgrown semi‐natural mesic meadow could be restored by sheep grazing after a long period of abandonment. The cover of vascular plant species in grazed plots and ungrazed exclosures was studied for five years and the responses of different functional plant groups were followed (herbs vs grasses, tall vs short species, species differing in flowering time, species representing different Grime's CSR strategies and species indicative of rich vs poor soil). Grazing increased species number by nearly 30%. On grazed plots the litter cover practically disappeared, favouring small herbs such as Rhinanthus minor, Ranunculus acris, Trifolium pratense and the grass Agrostis capillaris. Grazing decreased the cover of the late flowering tall herb Epilobium angustifolium but had no effect on the abundance of the early flowering tall herbs Anthriscus sylvestris or Geranium sylvaticum. We suggest that to succeed in restoration it is useful to determine the responses of different functional plant groups to grazing. Grassland managers need this information to optimize the methods and timing of management used in restoration. Additional management practices, such as mowing, may be needed in mesic meadows to decrease the dominance of tall species. Swa2p is an auxilin-like yeast protein that is involved in vesicular transport and required for uncoating of clathrin-coated vesicles. Swa2p contains a ubiquitin-associated (UBA) domain, which is present in a variety of proteins involved in ubiquitin (Ub)-mediated processes. We have determined a structural model of the Swa2p UBA domain in complex with Ub using NMR spectroscopy and molecular docking. Ub recognition occurs predominantly through an atypical interaction in which UBA helix α1 and the N-terminal part of helix α2 bind to Ub. Mutation of Ala148, a key residue in helix α1, to polar residues greatly reduced the affinity of the UBA domain for Ub and revealed a second low-affinity Ub-binding site located on the surface formed by helices α1 and α3. Surface plasmon resonance showed that the Swa2p UBA domain binds K48- and K63-linked di-Ub in a non-linkage-specific manner. These results reveal convergent evolution of a Ub-binding site on helix α1 of UBA domains involved in membrane protein trafficking. 相似文献

Little attention has been devoted to the investigation of the formation and functioning of ecotones at the interfaces between the upland and riparian zones. The margins of riparian mires act as redox geochemical barriers. Such biogeochemical hot spots retain elements that have leached from upland soils, favouring the growth of mesomorphic plants in hydromorphic soils. This paper aims to clarify the correspondence of vegetation borders to horizontal redox barriers of a Northern European moraine plain in Estonia.24 random transects were surveyed on the moraine plain, sampled across sharp soil moisture boundaries as proxies of redox barriers. The shifts in vegetation perpendicular to the boundaries were investigated. Curves of Euclidean distance were computed by moving a 20 m split-window along transects. The curves showed peaks of high dissimilarity a few metres below the redox barriers in the sites of natural soil moisture. In artificially drained sites, the relict redox barriers poorly corresponded to the vegetation borders. We conclude that in the studied moraine plain, the vegetation boundaries between riparian mires and uplands correspond to the redox barriers. The same can be set as a hypothesis for Northern European moraine plains. Related ecotones can be observed on the footslopes, revealing redox barrier in the landscape and related hot spots in cultural landscapes. In field margins, the redox barriers and corresponding ecotones occur below cultivation terraces. The ecotones may represent functional boundaries for delineating wetlands. Man-made drainage removes soil redox barriers and corresponding vegetation boundaries, making the landscape uniform and therefore vulnerable to disturbances. As a management implication, footslope ecotones should be preserved in their natural condition. 相似文献

We investigate the evolution of public goods cooperation in a metapopulation model with small local populations, where altruistic
cooperation can evolve due to assortment and kin selection, and the evolutionary emergence of cooperators and defectors via
evolutionary branching is possible. Although evolutionary branching of cooperation has recently been demonstrated in the continuous
snowdrift game and in another model of public goods cooperation, the required conditions on the cost and benefit functions
are rather restrictive, e.g., altruistic cooperation cannot evolve in a defector population. We also observe selection for
too low cooperation, such that the whole metapopulation goes extinct and evolutionary suicide occurs. We observed intuitive
effects of various parameters on the numerical value of the monomorphic singular strategy. Their effect on the final coexisting
cooperator–defector pair is more complex: changes expected to increase cooperation decrease the strategy value of the cooperator.
However, at the same time the population size of the cooperator increases enough such that the average strategy does increase.
We also extend the theory of structured metapopulation models by presenting a method to calculate the fitness gradient in
a general class of metapopulation models, and try to make a connection with the kin selection approach. 相似文献

Linear-amplification mediated PCR (LAM-PCR) has been developed to study hematopoiesis in gene corrected cells of patients treated by gene therapy with integrating vector systems. Due to the stable integration of retroviral vectors, integration sites can be used to study the clonal fate of individual cells and their progeny. LAM- PCR for the first time provided evidence that leukemia in gene therapy treated patients originated from provirus induced overexpression of a neighboring proto-oncogene. The high sensitivity and specificity of LAM-PCR compared to existing methods like inverse PCR and ligation mediated (LM)-PCR is achieved by an initial preamplification step (linear PCR of 100 cycles) using biotinylated vector specific primers which allow subsequent reaction steps to be carried out on solid phase (magnetic beads). LAM-PCR is currently the most sensitive method available to identify unknown DNA which is located in the proximity of known DNA. Recently, a variant of LAM-PCR has been developed that circumvents restriction digest thus abrogating retrieval bias of integration sites and enables a comprehensive analysis of provirus locations in host genomes. PINK1 and parkin constitute a mitochondrial quality control system mutated in Parkinson’s disease. PINK1, a kinase, phosphorylates ubiquitin to recruit parkin, an E3 ubiquitin ligase, to mitochondria. PINK1 controls both parkin localization and activity through phosphorylation of both ubiquitin and the ubiquitin‐like (Ubl) domain of parkin. Here, we observed that phospho‐ubiquitin can bind to two distinct sites on parkin, a high‐affinity site on RING1 that controls parkin localization and a low‐affinity site on RING0 that releases parkin autoinhibition. Surprisingly, ubiquitin vinyl sulfone assays, ITC, and NMR titrations showed that the RING0 site has higher affinity for phospho‐ubiquitin than phosphorylated Ubl in trans. We observed parkin activation by micromolar concentrations of tetra‐phospho‐ubiquitin chains that mimic mitochondria bearing multiple phosphorylated ubiquitins. A chimeric form of parkin with the Ubl domain replaced by ubiquitin was readily activated by PINK1 phosphorylation. In all cases, mutation of the binding site on RING0 abolished parkin activation. The feedforward mechanism of parkin activation confers robustness and rapidity to the PINK1‐parkin pathway and likely represents an intermediate step in its evolutionary development. 相似文献

Phytoplankton, mesozooplankton, mysids and fish larvae were studied during 15–29 annual cycles measured weekly to monthly in Pärnu Bay, the Gulf of Riga. The monthly variability of the biological data was related to temperature, ice conditions, salinity, influx of nutrients, the North Atlantic Oscillation (NAO) index, cloudiness and solar activity. Phytoplankton development was mainly a function of the NAO index. For the whole study period the abundance of zooplankton increased with increasing water temperature and solar activity. Significant correlations between phytoplankton and zooplankton densities were found until 1990. After the invasion of the predatory cladoceran Cercopagis pengoi in 1991, the zooplankton community was likely to be regulated by the introduced species rather than phytoplankton dynamics. The increased abundances of rotifers and copepods triggered the increase in mysid densities. The development of herring larvae was positively affected by the high density of copepods and rotifers but also by increased eutrophication. Until 1990 there was no significant relationship between the density of zooplankton and herring larvae. A negative relationship between the density of zooplankton and herring larvae in the 1990s suggests that the major shift in zooplankton community resulted in food limitation for herring larvae. The results indicated that (1) atmospheric processes in the northern Atlantic explain a large part of the interannual variation of the local phytoplankton stock, (2) trophic interactions control the development of pelagic communities at higher trophic levels, and (3) the introduction of an effective intermediate predator has repercussions for the whole pelagic food web in Pärnu Bay. 相似文献

Vertical migration of two dinoflagellate species (Peridiniellacatenata and Scrippsiella hangoei) and a phototrophic ciliate(Mesodinium
rubrum) were studied during the peak and decline of avernal bloom at the SW coast of Finland. During the diel cycle, part
of thepopulations of P. catenata and M. rubrum wereobserved in the deeper layers with elevated nutrient concentrations, whileS.
hangoei remained in the upper nutrient depleted mixed layer.Using a correspondence analysis the vertical distribution patterns
of thespecies and chlorophyll a were examined over a temporal scale of hoursand weeks. The vertical migration was reflected
in much higher variabilityin the depth distribution of P. catenata and M. rubrum over a diel scale, compared to S. hangoei.
The analysisrevealed also significant differences in species specific depth distributionpatterns over both time scales. It
is discussed that the co-existence of thetwo dominant dinoflagellate species during the vernal bloom is due to nicheseparation
through behavioural adaptations.
