Recently, there has been an increased interest in isotopical labeling of peptides. Although there are several techniques allowing for a complete labeling of all carboxyl groups in peptides, regioselective labeling would be beneficial in many situations. Such labeling requires the use of 18O‐labeled Fmoc amino acids. We have designed a method for such labeling that is an improvement on a technique proposed earlier. The new procedure is suitable for microscale synthesis and could be used in peptide and proteomics laboratories. Although for the majority of tested amino acids our method gives good labeling efficiency, it is time consuming. Therefore, we have decided to use microwave‐assisted procedure. The effect of UV-irradiation on four different types of native starch (corn, waxy corn, wheat and potato) have been investigated. Although the changes in the chemical structure of starch specimens were small, indicating good photostability, the samples lost adsorbed water and their crystallinity degree decreased after irradiation. Moreover, a drop in average molecular weight occurred in samples (with the exception of potato starch) as a result of main chain scission. The variations in the properties of investigated specimens of various origin were related to the differences in their structure and macromolecular arrangement. The lowest photostability among the four starches was exhibited by potato starch.  相似文献

Eukaryotic elongation factor 2 kinase (eEF2K) is the best-characterized member of the α-kinase family. Within this group, only eEF2K and myosin heavy chain kinases (MHCKs) have known substrates. Here we have studied the roles of specific residues, selected on the basis of structural data for MHCK A and TRPM7, in the function of eEF2K. Our data provide the first information regarding the basis of the substrate specificity of α-kinases, in particular the roles of residues in the so-called N/D loop, which appears to occupy a position in the structure of α-kinases similar to that of the activation loop in other kinases. Several mutations in the EEF2K gene occur in tumors, one of which (Arg303Cys) is at a highly conserved residue in the N/D loop. This mutation greatly enhances eEF2K activity and may be cytoprotective. Our data support the concept that the major autophosphorylation site (Thr348 in eEF2K) docks into a binding pocket to help create the kinase-competent conformation. This is similar to the situation for MHCK A and is consistent with this being a common feature of α-kinases.  相似文献

The synthesis of diethyl (pyridin-2-, -3-, -4-ylmethyl)phosphate (2-pmOpe, 3-pmOpe, 4-pmOpe) ligands and their palladium (II) complexes of general formula trans-[PdCl2L2] (L = 2-pmOpe, 3-pmOpe,4-pmOpe) has been described. Pyridine phosphate derivatives were synthesized via the condensation of phosphorochloridic acid diethyl ester with an appropriate pyridinylmethanol in the presence of triethylamine. The compounds have been identified and characterized by IR, far-IR, 1H NMR, 31P NMR, 31P CP-MAS NMR and elemental analyses. The crystal and molecular structures of palladium (II) complexes, i.e., [PdCl2(2-pmOpe)2] and [PdCl2(4-pmOpe)2] determined by the X-ray diffraction method, are presented. In both structures, Pd(II) ions are four-coordinated by two chlorine atoms and two pyridine nitrogen atoms. The geometry of complexes is square-planar and adopt a trans configuration, which is consistent with preparation method.  相似文献

Two digestive enzymes from Antarctic krill: an endo-(1----3)-beta-glucanase and a serine proteinase which specifically cleaves native collagen were characterized with regard to their specificity and accommodation to acting at low temperatures. Their presence in the crustacean digestive apparatus proves that krill is an omnivorous organism, and this fact should be considered in estimations of its biomass stock.  相似文献
