Structures of the complexes (η3-C3H5)Pd(μ-η6:1-CH2PhCr(CO)3 and (η3-C3H5)Pd[μ-η6:1-CH(Ph)Ph]Cr(CO)3 in solution were evaluated by NMR (1H and 13C) and IR spectroscopy. The dynamic behaviour of the complexes was investigated. Quick rotation (on the NMR time scale) of the tricarbonylchromium groups around the axis passing through the centre of the η6-coordinated phenyl ring and the chromium atom takes place at room temperature and becomes slow on cooling. The η3-allylic ligand was proved to undergo no dynamic changes in solution. Unlike the solid state, the semi-bridging carbonyl groups between chromium and palladium atoms are absent or very weak in solution. Cross-coupling reactions of the complexes with organohalides are described.  相似文献

The aim of this work is to develop methods for determining the anaerobic threshold according to the rate of ventilation and cardio interval variability during the test with stepwise increases load on the cycle ergometer and treadmill. In the first phase developed the method for determining the anaerobic threshold for lung ventilation. 49 highly skilled skiers took part in the experiment. They performed a treadmill ski-walking test with sticks with gradually increasing slope from 0 to 25 degrees, the slope increased by one degree every minute. In the second phase we developed a method for determining the anaerobic threshold according dynamics ofcardio interval variability during the test. The study included 86 athletes of different sports specialties who performed pedaling on the cycle ergometer "Monarch" in advance. Initial output was 25 W, power increased by 25 W every 2 min. The pace was steady--75 rev/min. Measurement of pulmonary ventilation and oxygen and carbon dioxide content was performed using gas analyzer COSMED K4. Sampling of arterial blood was carried from the ear lobe or finger, blood lactate concentration was determined using an "Akusport" instrument. RR-intervals registration was performed using heart rate monitor Polar s810i. As a result, it was shown that the graphical method for determining the onset of anaerobic threshold ventilation (VAnP) coincides with the accumulation of blood lactate 3.8 +/- 0.1 mmol/l when testing on a treadmill and 4.1 +/- 0.6 mmol/1 on the cycle ergometer. The connection between the measure of oxygen consumption at VAnP and the dispersion of cardio intervals (SD1), derived regression equation: VO2AnT = 0.35 + 0.01SD1W + 0.0016SD1HR + + 0.106SD1(ms), l/min; (R = 0.98, error evaluation function 0.26 L/min, p < 0.001), where W (W)--Power, HR--heart rate (beats/min), SD1--cardio intervals dispersion (ms) at the moment of registration of cardio interval threshold.  相似文献

We identified a family in which five siblings were diagnosed with multiple sclerosis (MS) or clinically isolated syndrome. Several women in the maternal lineage have comorbidities typically associated with Peutz Jeghers Syndrome, a rare autosomal-dominant disease caused by mutations in the serine-threonine-kinase 11 (STK11) gene, which encodes liver kinase B1. Sequence analysis of DNA from one sibling identified a single-nucleotide polymorphism (SNP) within STK11 intron 5. This SNP (dbSNP ID: rs9282860) was identified by TaqMan polymerase chain reaction (PCR) assays in DNA samples available from two other siblings. Further screening was carried out in samples from 654 relapsing-remitting MS patients, 100 primary progressive MS patients, and 661 controls. The STK11-SNP has increased frequency in all female patients versus controls (odds ratio = 1.66, 95% CI = 1.05, 2.64, p = .032). The STK11-SNP was not associated with disease duration or onset; however, it was significantly associated with reduced severity (assessed by MS severity scores), with the lowest scores in patients who also harbored the HLA-DRB1*1501 allele. In vitro studies showed that peripheral blood mononuclear cells from members of the family were more sensitive to the mitochondrial inhibitor metformin than cells from MS patients with the major STK11 allele. The increased association of SNP rs9282860 in women with MS defines this variant as a genetic risk factor. The lower disease severity observed in the context of HLA-DRB1*1501 combined with limited in vitro studies raises the provocative possibility that cells harboring the STK11-SNP could be targeted by drugs which increase metabolic stress.  相似文献

Analysis of the precursors of bacterial exported proteins revealed that those having bulky hydrophobic residues at position -5 have a high incidence of Pro residues at positions -6 and -4, Val at position -3, and Ser at positions -4 and -2. This led to a hypothesis that the previously observed inhibition of processing by bulky residues at position -5 can be suppressed by introduction of Pro, Ser, or Val in the corresponding nearby positions. Subsequent mutational analysis of Escherichia coli alkaline phosphatase showed that, as it was predicted, Pro on either side of bulky hydrophobic -5 Leu, Ile, or Tyr completely restores efficiency of the maturation. Introduction of Val at position -3 also partially suppresses the inhibition imposed by -5 Leu, while a Ser residue at position -4 or -2 does not restore processing. In addition, effective maturation of a mutant with Pro residues at positions from -6 throughout -4 proved that polyproline conformation of this region is permissive for processing. To understand the effects of the mutations, we modeled a peptide substrate into the active site of the signal peptidase using the known position of the beta-lactam inhibitor. The inhibitory effect of the -5 residue and its suppression by either Pro -6 or Pro -4 can be explained if we assume that Pro-containing -6 to -4 regions adopt a polyproline conformation whereas the region without Pro residues has a beta-conformation. These results permit us to specify sequence requirements at -6, -5, and -4 positions for efficient processing and to improve the prediction of yet unknown cleavage sites.  相似文献

Plasmid pCspA::Km carrying a cloned mutant allele of the cspA gene for the major Escherichia coli cold-shock protein CspA with an insertion of the kanamycin resistance gene cassette from transposon Tn903 into the core region of the coding sequence causes a 2.3-fold increase in radioresistance of wild-type E. coli cells (cspA +). The radiation protective effect of this plasmid is abolished or drastically reduced in mutants recA13and rpoH15defective in RecA protein and in induction of the heat-shock protein regulon, respectively. Plasmid pCspA::Km causes a 1.3-fold elevation in the resistance to -irradiation of E. coli mutants with an intermediate level of radiation resistance (Gamr445 and KS0160) but slightly diminishes resistance of a highly radiation-resistant Gamr444 mutant. In the chromosome of E. coli strain with normal DNA repair systems, the cspA::Km mutation in the homozygous state enhances resistance to the lethal effect of -rays and UV light 2.9 and 1.4 times, respectively. These data suggest that the system of cold-shock proteins can modulate resistance of E. colicells to the lethal effect of -rays and UV light.  相似文献

The African catfish, Clarias gariepinus, possesses a pair of suprabranchial chambers located in the dorsal-posterior part of the branchial cavity having extensions from the upper parts of the second and fourth gill arches, forming the arborescent organs. This structure is an air-breathing organ (ABO) and allows aerial breathing (AB). We evaluated its cardiorespiratory responses to aquatic hypoxia. To determine the mode of air-breathing (obligate or accessory), fish had the respiratory frequency (f R) monitored and were subjected to normoxic water (PwO2 = 140 mmHg) without becoming hyperactive for 30 h. During this period, all fish survived without displaying evidences of hyperactivity and maintained unchanged f R, confirming that this species is a facultative air-breather. Its aquatic O2 uptake ( [(V)\dot]\textO2 \dot{V}{\text{O}}_{2} ) was maintained constant down to a critical PO2 (PcO2) of 60 mmHg, below which [(V)\dot]\textO2 \dot{V}{\text{O}}_{2} declined linearly with further reductions of inspired O2 tension (PiO2). Just above the PcO2 the ventilatory tidal volume (V T) increased significantly along with gill ventilation ( [(V)\dot]\textG \dot{V}_{\text{G}} ), while f R changed little. Consequently, the water convection requirement ( [(V)\dot]\textG /[(V)\dot]\textO2 ) \left( {\dot{V}_{\text{G}} /\dot{V}{\text{O}}_{2} } \right) increased steeply. This threshold applied to a cardiac response that included reflex bradycardia. AB was initiated at PiO2 = 140 mmHg (normoxia) and air-breathing episodes increased linearly with more severe hypoxia, being significantly higher at PiO2 tensions below the PcO2. Air-breathing episodes were accompanied by bradycardia pre air-breath, to tachycardia post air-breath.  相似文献

Predominantly, Hoplias malabaricus inhabits stagnant O2 poor environments, whereas Hoplias lacerdae occurs in well-aerated streams. The present study evaluates the influence of mode of life on O2 uptake and gill ventilation in equally-sized (300 g) specimens of this genus at 25° C. Comparing the species, H. lacerdae was characterized by the highest O2 uptake and gill ventilation combined with a relatively higher cost of breathing and a lower O2 extraction. Both species substantially increased ventilation in response to hypoxia with the difference that H. malabaricus exclusively augmented tidal volume, whereas H. lacerdae also increased breathing frequency.  相似文献
