1H-3-Hydroxy-4-oxoquinaldine 2,4-dioxygenase (Hod) is a cofactor-less dioxygenase belonging to the alpha/beta hydrolase fold family, catalyzing the cleavage of 1H-3-hydroxy-4-oxoquinaldine (I) and 1H-3-hydroxy-4-oxoquinoline (II) to N-acetyl- and N-formylanthranilate, respectively, and carbon monoxide. Bisubstrate steady-state kinetics and product inhibition patterns of HodC, the C69A protein variant of Hod, suggested a compulsory-order ternary-complex mechanism, in which binding of the organic substrate precedes dioxygen binding, and carbon monoxide is released first. The specificity constants, k(cat)/K(m,A) and k(cat)/K(m,O)()2, were 1.4 x 10(8) and 3.0 x 10(5) M(-1) s(-1) with I and 1.2 x 10(5) and 0.41 x 10(5) M(-1) s(-1) with II, respectively. Whereas HodC catalyzes formation of the dianion of its organic substrate prior to dioxygen binding, HodC-H251A does not, suggesting that H251, which aligns with the histidine of the catalytic triad of the alpha/beta hydrolases, acts as general base in catalysis. Investigation of base-catalyzed dioxygenolysis of I by electron paramagnetic resonance (EPR) spectroscopy revealed formation of a resonance-stabilized radical upon exposure to dioxygen. Since in D(2)O spectral properties are not affected, exchangeable protons are not involved, confirming that the dianion is the reactive intermediate that undergoes single-electron oxidation. We suggest that in the ternary complex of the enzyme, direct single-electron transfer from the substrate dianion to dioxygen may occur, resulting in a radical pair. Based on the estimated spin distribution within the radical anion (observed in the model reaction of I), radical recombination may produce a C4- or C2-hydroperoxy(di)anion. Subsequent intramolecular attack would result in the 2,4-endoperoxy (di)anion that may collapse to the reaction products. 相似文献

The response of barley (Hordeum vulgare L. cv. Akcent) to various photosynthetic photon flux densities (PPFDs) and elevated [CO2] [700 μmol (CO2) mol−1; EC] was studied by gas exchange, chlorophyll (Chl) a fluorescence, and pigment analysis. In comparison with barley grown under ambient [CO2] [350 μmol (CO2) mol−1; AC] the EC acclimation resulted in a decrease in photosynthetic capacity, reduced stomatal conductance, and decreased total
Chl content. The extent of acclimation depression of photosynthesis, the most pronounced for the plants grown at 730 μmol
m−2 s−1 (PPFD730), may be related to the degree of sink-limitation. The increased non-radiative dissipation of absorbed photon energy for
all EC plants corresponded to the higher de-epoxidation state of xanthophylls only for PPFD730 barley. Further, a pronounced decrease in photosystem 2 (PS2) photochemical efficiency (given as FV/FM) for EC plants grown at 730 and 1 200 μmol m−2 s−1 in comparison with AC barley was related to the reduced epoxidation of antheraxanthin and zeaxanthin back to violaxanthin
in darkness. Thus the EC conditions sensitise the photosynthetic apparatus of high-irradiance acclimated barley plants (particularly
PPFD730) to the photoinactivation of PS2.

A new class of compounds for the plant family Lamiaceae, benzoxazinoids, was found in Lamium galeobdolon. From the aerial parts of the species were isolated the new 2-O-beta-D-glucopyranosyl-6-hydroxy-2H-1,4-benzoxazin-3(4H)-one (6-hydroxy blepharin) together with four known benzoxazinoids, DHBOA-Glc, blepharin, DIBOA, DIBOA-Glc, as well as harpagide, 8-O-acetyl-harpagide and salidroside. Eight known iridoid glucosides, 24-epi-pterosterone and verbascoside were isolated from Lamium amplexicaule, L. purpureum and L. garganicum. The iridoids, 5-deoxylamiol and sesamoside, as well as the phytoecdysone, 24-epi-pterosterone, were found for the first time for the genus Lamium. Using 77 K chlorophyll a (Chl a) fluorescence spectra in vivo, the development was studied of Photosystems II (PS II) and I (PS I) during greening of barley under intermittent light followed
by continuous light at low (LI, 50 μmol m−2 s−1) and high (HI, 1000 μmol m−2 s−1) irradiances. The greening at HI intermittent light was accompanied with significantly reduced fluorescence intensity from
Chl b excitation for both PS II (F685) and PS I (F743), in comparison with LI plants, indicating that assembly of light-harvesting
complexes (LHC) of both photosystems was affected to a similar degree. During greening at continuous HI, a slower increase
of emission from Chl b excitation in PS II as compared with PS I was observed, indicating a preferred reduction in the accumulation of LHC II. The
following characteristics of 77 K Chl a fluorescence spectra documented the photoprotective function of an elevated content of carotenoids in HI leaves: (1) a pronounced
suppression of Soret region of excitation spectra (410–450 nm) in comparison with the red region (670–690 nm) during the early
stage of greening indicated a strongly reduced excitation energy transfer from carotenoids to the Chl a fluorescing forms within PS I and PS II; (2) changes in the shape of the excitation band of Chl b and carotenoids (460–490 nm) during greening under continuous light confirmed that the energy transfer from carotenoids to
Chl a within PS II remained lower as compared with the LI plants.

The ability of spring barley (Hordeum vulgare cv. Akcent) to adjust the composition and function of the photosynthetic apparatus to growth irradiances of 25–1200 μmol
m−2 s−1 was studied by gas exchange and chlorophyll a fluorescence measurements and high-performance liquid chromatography. The increased growth irradiance stimulated light- and
CO2-saturated rates of CO2 assimilation expressed on a leaf area basis up to 730 μmol m−2 s−1 (HL730), whereas at an irradiance of 1200 μmol m−2 s−1 (EHL1200) both rates decreased significantly. Further, the acclimation to EHL1200 was associated with an extremely high chlorophyll
a/b ratio (3.97), a more than doubled xanthophyll cycle pool (VAZ) and a six-fold higher de-epoxidation state of the xanthophyll
cycle pigments as compared to barley grown under 25 μmol m−2 s−1 (LL25). EHL1200 plants also exhibited a long-term inhibition of Photosystem II (PS II) photochemical efficiency (F
v/F
m). Photosynthetic capacity, chlorophyll a/b and VAZ revealed a linear trend of dependence on PS II excitation pressure in a certain range of growth irradiances (100–730
μmol m−2 s−1). The deviation from linearity of these relationships for EHL1200 barley is discussed. In addition, the role of increased
VAZ and/or accumulation of zeaxanthin and antheraxanthin in acclimation of barley to high irradiance is studied with respect
to regulation of non-radiative dissipation and/or photochemical efficiency within PS II.

An excess of the proinflammatory substance IL-18 is present in joints of patients with rheumatoid arthritis (RA), and expression of IL-18 receptor (IL-18R) regulates IL-18 bioactivity in various cell types. We examined the expression of IL-18R alpha-chain and beta-chain and the biologic effects of IL-18 in fibroblast-like synoviocytes (FLS) after long-term culture. The presence of both IL-18R chains was a prerequisite for IL-18 signal transduction in FLS. However, all FLS cultures studied were either resistant or barely responsive to IL-18 stimulation as regards cell proliferation, expression of adhesion molecules ICAM-1 and vascular cell adhesion molecule (VCAM)-1, and the release of interstitial collagenase and stromelysin, IL-6 and IL-8, prostaglandin E2, or nitric oxide. We conclude that the presence of macrophages or IL-18R+ T cells that can respond directly to IL-18 is essential for the proinflammatory effects of IL-18 in synovitis in RA. 相似文献

Cytokinin (CK) levels in cotyledons of Cucurbita pepo L. (zucchini) were investigated through the processes of post-germination, greening, natural senescence and subsequent rejuvenation. The concentrations of the physiologically active CK bases, ribosides and nucleotides, as well as the cis -isomers of zeatin derivatives, decreased between the first and fifth weeks of cultivation under controlled light conditions. At the same time, the levels of storage CK O -glucosides and physiologically inactive CK 7- and 9-glucosides increased with senescence. With plant decapitation and subsequent cotyledon rejuvenation, not only the chlorophyll content but also the levels of physiologically active CKs, nucleotides and cis -zeatin derivatives increased. The levels of O -glucosides, however, decreased. When 1-week-old seedlings were transferred to the dark, there was a progressive reduction in cotyledon chlorophyll content, deterioration of chloroplast ultrastructure and a decrease in physiologically active CKs and their nucleotides. In contrast with natural senescence, the storage CK O -glucosides decreased under dark conditions, suggesting different metabolic regulation of endogenous CK levels during natural and dark-induced senescence of zucchini cotyledons. The chlorophyll loss of dark-treated cotyledons could be partially reversed, even after 5 days, with return to light conditions. During this recovery, physiologically active CKs and their nucleotides again increased, whereas the storage CK O -glucosides and cis -zeatins decreased. The present results suggest that dark-induced destruction and subsequent restoration of chloroplasts during light shifts are controlled by changes in the levels of physiologically active CKs and their nucleotides. 相似文献

Cyprus is an island in the eastern Mediter-ranean basin inhabited by people of Caucasian extraction, mostly Greek-Cypriots. The most common inherited disease among Caucasians is cystic fibrosis (CF). Although no careful scientific study had ever been done the impression was that CF was extremely rare among the Greek-Cypriots, with an incidence estimated at around 130,000. About 2 years ago, we introduced molecular diagnostic methodology in an effort to assist clinicians in safer diagnosis of patients presenting with atypical CF symptomatology, and also for testing the hypothesis that mutations that cause milder phenotypes might be responsible for misdiagnosis or for missing entirely some cases of CF. Initial screening for F508 revealed that it is indeed rare in the general population. Further screening of suspected CF patients revealed a novel mutation that converted leucine at position 346 to proline (L346P) in two unrelated families. The second CF mutation was F508 and 1677delTA in the two families respectively, both reportedly associated with severe phenotypes. Yet our patients did not present with typical CF pictures possibly because of the dominant nature of this novel mild mutation in exon 7. Symptoms included failure to thrive, chest infections and electrolyte disturbances. These findings raise the possibility that Cyprus might have been spared very severe CF phenotypes but not cystic fibrosis transmembrane conductance regulator (CFTR) mutations. 相似文献
