We examined host evaluation behaviour in three species of aphid parasitoids, Ephedrus californicus Baker, Monoctonus paulensis (Ashmead), and Praon pequodorum Viereck (Hymenoptera: Aphidiidae). Mated females were provided with pairwise choices among three kinds of hosts in the laboratory: (green) pea aphid, Acyrthosiphon pisum (Harris), and a green and a pink colour morph of alfalfa aphid, Macrosiphum creelii Davis. Patterns of attack and host acceptance were species-specific. Females of E. californicus did not respond to the presence of aphids prior to making antennal contact. Variations in rates of parasitization (pea aphid>green alfalfa aphid>pink alfalfa aphid) were consistent with differences in aphid defensive behaviours; no ‘preference’ for any host type was evident when aphids were anaesthetized with carbon dioxide. In M. paulensis, the order of preference (pea aphid>green alfalfa aphid>pink alfalfa aphid) did not vary when aphids were immobilized, or presented in the dark, or both. Host movement did not influence the rate of attack by M. paulensis. In contrast, the ranked order of preference in P. pequodorum varied with circumstance. In the light, females attacked pea aphid and green alfalfa aphid with equal frequency, but parasitized significantly more of the former; both kinds of aphids were attacked and parasitized at higher rates than pink alfalfa aphid. In the dark, P. pequodorum females parasitized green and pink alfalfa aphids equally and at higher rates than pea aphids. Whereas E. californicus was more successful ovipositing in immobilized hosts, P. pequodorum females attacked and laid more eggs in normal than anaesthetized aphids. Patterns of host recognition and evaluation are compared across six species representing four genera in the family Aphidiidae.  相似文献

We discovered the missense mutation, A226V, in the ornithine-delta-aminotransferase (OAT) genes of two unrelated patients with gyrate atrophy of the choroid and retina (GA). One patient, who was a compound for A226V and for the premature termination allele R398ter, showed a significant (P < .01) decrease in mean plasma ornithine levels, following pyridoxine supplementation with a constant protein intake: 826 +/- 128 microM (n = 5; no pyridoxine supplementation) versus 504 +/- 112 microM (n = 6; 500 mg pyridoxine/d) and 546 +/- 19 microM (n = 6; 1,000 mg pyridoxine/d). In extracts of fibroblasts from a second GA patient homozygous for A226V and from Chinese hamster ovary cells expressing an OAT-cDNA-containing A226V, we found that OAT activity increased from undetectable levels to approximately 10% of normal when the concentration of pyridoxal phosphate was increased from 50 to 600 microM. A226V is the fourth disease-causing pyridoxine-responsive human mutation to be reported.  相似文献

A method for the extraction of ctDNA from isolated chloroplast was developed. This method is simple and adapted particularly to broad-leaved trees, including sclerophyllous species with high phenolic and polysaccharide contents. This method includes two major steps: first, chloroplasts are isolated in non-aqueous solutions to avoid oxidation and phenolic problems; second, ctDNA is extracted from the chloroplasts using aqueous solutions and specific methods to provide highly purified ctDNA.  相似文献

An anaerobic liquid enrichment method followed by plating on a selective medium revealed that the soft rot coliform bacterium Erwinia carotovora subsp. carotovora was generally present in water from drains, ditches, streams, rivers and lakes (including reservoirs) in southern Scotland and in Colorado, United States, in mountainous, upland and arable areas through the year. Many sites were remote from susceptible or diseased crops. Erwinia carotovora subsp. atroseptica was isolated much less frequently and no Erwinia bacteria were isolated from underground waters. Erwinia bacteria were also found in rain-water in Scotland, in winter snow from mountain passes in Colorado, and in sea water from the west and east coasts of Scotland and from the coasts of Oregon, California, Texas, Louisiana and Florida. The significance of the occurrence of these bacteria in water is discussed in relation to the control of blackleg and soft rot diseases of potato by production of Erwinia -free stocks.  相似文献

Significant differences were observed among 11 alfalfa cultivars for the percentage of plants resitant to Leptospbaerulina briosiana after one inoculation. Screening for resistance was done by inoculating 18 day old plants (cv. Europe), held in a moist chamber, with L. briosiana. The self- and cross-progenies of plants resistant to L. briosiana after two inoculations showed slightly higher resistance to Leptosphaerulina leaf spot than the unselected original population but the difference was significant for the cross-progeny only. Larger variations were observed among self-progenies than cross-progenies. No correlation was found between self - and cross-progenies. At least three inoculations may be necessary to eliminate most susceptible plants. Mas selection was as effective as the polycross progeny test to improve the level of resistance of the selected populations as compared with the unselected one but the difference was agronomically unimportant. Under field conditions, the plants selected as resistant after two inoculations in the laboratory showed better resistance to L. briosiana than the susceptible ones but only on the first of three notations. The population derived from plants selected from cv. Europe as resistant to L. briosiana in the laboratory was not significantly more resitant than cv. Europe in the filed but generally showed less foliar infection.  相似文献

Twelve day old winter wheat seedlings (cvs Kharkov, frost hardy and Champlein, less hardy) accumulated linolenic acid at the expense of linoleic acid during controlled hardening. The change was most pronounced in the roots, where it was not specific to the phospholipid fraction. It was less marked in the leaves, but occurred there mainly in the phospholipids. The lack of differences between fatty acid profiles of the two cultivars rules out the explanation of varietal differences in frost hardiness in winter wheat on the basis of major changes in fatty acid unsaturation.  相似文献

Protein import to the nucleus is a signal-mediated process that exhibits saturation kinetics. We investigated whether signal bearing proteins compete with U2 and U6 snRNPs during import. When injected into Xenopus oocytes, saturating concentrations of P(Lys)-BSA, a protein bearing multiple nuclear localization signals from SV40 large T-antigen, reduce the rate of [125I]P(Lys)-BSA and of [125I]nucleoplasmin import, consistent with their competing for and sharing the same limiting component of the import apparatus. In contrast, saturating concentrations of P(Lys)-BSA do not reduce the rate of HeLa [32P]U2 snRNP assembly or import. The import of U6 snRNP is also competed by P(Lys)-BSA. We conclude that U2 snRNP is imported into oocyte nuclei by a kinetic pathway that is distinct from the one followed by P(Lys)-BSA, nucleoplasmin, and U6 snRNP.  相似文献

The inhibitory effects of wheat germ agglutinin and mAb 414 on the nuclear import of all types of U snRNAs indicate that they cross the nuclear envelope through the nuclear pore complex. However, the import of different U snRNAs occurs by kinetically distinct targeting pathways that can be distinguished from one another by the competitive effects of free trimethylguanosine cap dinucleotide (m3GpppG) and P(Lys)-BSA, an efficient synthetic karyophile based on the nuclear localization signal of SV40 large T antigen. The import of U snRNAs that contain 5' m3GpppN caps and are complexed by Sm proteins (U1, U2, U4, and U5) is competed by coinjection with free m3GpppG, indicating a shared transport factor, but not by P(Lys)-BSA. The import of U6 snRNA, which lacks a m3GpppN cap and is not complexed by the Sm proteins, is competed by P(Lys)-BSA but not by free m3GpppG. Thus, by the criterion of kinetic competition, U6 snRNA import is identical to that of the karyophilic proteins P(Lys)-BSA and nucleoplasmin. Uniquely, the import of U3 snRNA, which contains a m3GpppN cap but does not bind Sm proteins is not competed by either free m3GpppG or P(Lys)-BSA. Thus, U3 snRNA appears to be imported by a novel third kinetic pathway.  相似文献

Five polymorphic enzymatic loci were studied in 30 populations of Quercus ilex distributed from Crete to North Africa. A high within-population genetic diversity was found for each locus studied. Most of the alleles were common to all the populations, indicating that the holm oak corresponds to a single genetic entity. Observation of breeding system characteristics showed that the holm oak is monoecious, wind-pollinated, and likely to possess genetic autoincompatibility. Furthermore, studies of flowering phenology in a single population showed that the period of flowering varied notably among trees. An average of 29% of the trees showed inter-annual variation in flowering time. The male, female or vegetative investment also varied widely among trees from one year to another. This results in a diversification of pollen source received by a given tree each year, and also from one year to another.  相似文献

Phosphorus (P) concentrations in water and sediment of a highly eutrophic lake were monitored before and after application of diquat to control the macrophyte Potamogeton crispus. Only a relatively small and short-term increase in P concentration in water occurred shortly after plant die-off resulting from herbicide application. Phosphorus concentrations in shallow water sediments decreased during the summer, and those in deep water sediments increased. Although a large increase in P concentration in the water occurred in late summer, it was not attributed to diquat. No major secondary effects of herbicide application were found during this study.  相似文献
