Fusicoccin-like ligands in higher plants were sought by combining high performance liquid chromatography with radioreceptor analysis and radioimmunoassay. Such substances were found in genetically transformed cultured roots of horseradish, alteus and lupine as well as in native horseradish, cucumber, horse chestnut, and maize plants. In root crops such as carrot and sugar beet or in potato tubers, only traces of fusicoccin-like ligands were detected. A total of 44 accessions of Brachiaria decumbens were analysed for chromosome count and meiotic behaviour in order to identify potential progenitors for crosses. Among them, 15 accessions presented 2n = 18; 27 accessions, 2n = 36; and 2 accessions, 2n = 45 chromosomes. Among the diploid accessions, the rate of meiotic abnormalities was low, ranging from 0.82% to 7.93%. In the 27 tetraploid accessions, the rate of meiotic abnormalities ranged from 18.41% to 65.83%. The most common meiotic abnormalities were related to irregular chromosome segregation, but chromosome stickiness and abnormal cytokinesis were observed in low frequency. All abnormalities can compromise pollen viability by generating unbalanced gametes. Based on the chromosome number and meiotic stability, the present study indicates the apomictic tetraploid accessions that can act as male genitor to produce interspecific hybrids with B. ruziziensis or intraspecific hybrids with recently artificially tetraploidized accessions.  相似文献

The incorporation of rifampicin into multilayer phospholipid vesicles depending on the concentration of antibiotic and phospholipid content was studied. The extent of incorporation of rifampicin into monolayer vesicles (liposomes), obtained by the homogenization of multilamellar vesicles, was determined by the method of gel filtration. It was found that rifampicin better penetrates and is retained in membranes consisting of a mixture of phosphatidylcholine and cardiolipin, the maximum incorporation of rifampicin into liposomes being 17%. It was shown by 31P NMR spectroscopy that, during the interaction of rifampicin with the phospholipid membrane, the bilayer packing of phospholipids is destroyed.  相似文献

Cse4p is a structural component of the core centromere of Saccharomyces cerevisiae and is a member of the conserved CENP-A family of specialized histone H3 variants. The histone H4 allele hhf1-20 confers defects in core centromere chromatin structure and mitotic chromosome transmission. We have proposed that Cse4p and histone H4 interact through their respective histone fold domains to assemble a nucleosome-like structure at centromeric DNA. To test this model, we targeted random mutations to the Cse4p histone fold domain and isolated three temperature-sensitive cse4 alleles in an unbiased genetic screen. Two of the cse4 alleles contain mutations at the Cse4p-H4 interface. One of these requires two widely separated mutations demonstrating long-range cooperative interactions in the structure. The third cse4 allele is mutated at its helix 2-helix 3 interface, a region required for homotypic H3 fold dimerization. Overexpression of wild-type Cse4p and histone H4 confer reciprocal allele-specific suppression of cse4 and hhf1 mutations, providing strong evidence for Cse4p-H4 protein interaction. Overexpression of histone H3 is dosage lethal in cse4 mutants, suggesting that histone H3 competes with Cse4p for histone H4 binding. However, the relative resistance of the Cse4p-H4 pathway to H3 interference argues that centromere chromatin assembly must be highly regulated.  相似文献

Chronic endobronchial inflammation and bacterial infection are the main causes of morbidity and mortality in cystic fibrosis (CF), an autosomal recessive genetic disorder associated with improper function of chloride channels. Inflammation in CF lung is greatly amplified after Pseudomonas aeruginosa infection. In this study the relationship between P. aeruginosa status and inflammatory markers has been investigated. Seventeen CF children in acute lung exacerbation were examined. CF patients without P. aeruginosa infection were characterized by elevated activity of sputum elastase, reduced response of peripheral blood lymphocytes to PHA and significant resistance to the antiproliferative action of glucocorticoids. These parameters were normalized after antibiotic treatment. The patients with prolonged P. aeruginosa infection demonstrated extremely high levels of elastase activity and elevated amounts of sputum IL-8 and TNF-alpha. Although antibiotic treatment resulted in clinical improvement, it failed to suppress excessive immune response in the lung. The data indicate that CF patients with prolonged P. aeruginosa need the modified treatment, which should include immunomodulating drugs and protease inhibitors as well as antibacterial therapy.  相似文献

本文报道了我国4个地理品系卤虫的染色体倍性组成及卵径分布,分析了卤虫染色体的非整倍性,讨论了卤虫种的分布。 天津卤虫有2倍体(2n=42),占33.3%和5倍体,占16.2%;染色体数的波动为21—108;卵径为262±17μm。 海南卤虫有2倍体(2n=42),占8%;4倍体,占17.7%;5倍体,占18.3%;染色体数的波动为16—111;卵径为285±15μm。 新疆卤虫有2倍体(2n=42),占39.2%;染色体数的波动为17—106;卵径为269±15μm。 山西卤虫有2倍体(2n=42),占45.1%,染色体数的波动为17—84;卵径为234±13μm。  相似文献

The effects were investigated of LTC4, a synthetic leukotriene, and BW 755C, a blocker of LTC4 biosynthesis, on the operation of Ca channels at the cell membrane and on contraction of muscle fibers using intracellular dialysis and voltage clamping at the membrane of isolated nerve cells and by recording spontaneous contraction of the uterus in white rats at advanced stages of pregnancy. It was found that 1·10–7 M LTC4 stimulates the contraction of the uterus without altering its response to oxytocin application. The same concentration of LTC4 was found to increase calcium conductance by 60±27%. At the same time, a 25±6 mV shift in peak current-voltage relationship along the voltage axis toward negative values was recorded for calcium current. BW 755C, a blocker of the key enzyme in the lipoxygenase metabolic pathway of arachidonic acid, exerts an action similar to leukotriene on calcium conductance, although brief contraction of the uterus is rapidly replaced by complete inhibition of this activity.Institute of Bioorganic Chemistry, Academy of Sciences of the Ukrainian SSR, Kiev.
