Cellular entry of pathogenic hantaviruses had been shown to be mediated by beta3 integrins. However, no direct evidence exists that hantavirus binds to beta3 integrins, and integrin beta3 subunit is not expressed on some cells permissive to hantavirus infection. In this report, utilizing beta3-integrin-transfected CHO cells, we demonstrated that integrin beta3 subunit renders CHO cells susceptible to Chinese Hantaan virus (HTN) strain A9 (isolated in China), and the viral infection was correspondingly inhibited by antibodies to alphavbeta3, alphaIIbbeta3, beta3, and alphav integrins. Furthermore, virus overlay protein-binding assay and 'quarternary Western' analysis indicate that HTN A9 directly interacts with beta3 integrins and an unidentified 70kDa protein. These findings indicate that beta3 integrins play a crucial role in cellular entry of HTN A9 via specific interactions with the virus. In addition, a novel 70kDa protein may serves as a candidate receptor or alternative cellular component for interaction with HTN.  相似文献

The circular dichroism (CD) spectra of single-stranded DNAs (ssDNAs) are significantly perturbed by the binding of single-stranded DNA binding proteins such as the Ff bacteriophage gene 5 protein (g5p) and the A domain of the 70 kDa subunit of human replication protein A (RPA70-A). These two proteins have similar OB-fold secondary structures, although their CD spectra at wavelengths below 250 nm differ greatly. The spectrum of g5p is dominated by a tyrosyl L(a) band at 229 nm, while that of RPA70-A is dominated by its beta secondary structure. Despite differences in their inherent spectral properties, these two proteins similarly perturb the spectra of bound nucleic acid oligomers. CD spectra of free, non-protein-bound ssDNAs are dependent on interactions of the nearest-neighboring nucleotides in the sequence. The CD spectra (per mol of nucleotide) of simple repetitive sequences 48 nucleotides in length and containing simple combinations of A and C are related by nearest-neighbor equations. For example, 3 x Deltaepsilon[d(AAC)(16)] = 3 x Deltaepsilon[d(ACC)(16)] + Deltaepsilon[d(A)(48)] - Deltaepsilon[d(C)(48)]. Moreover, nearest-neighbor equations relate the spectra of ssDNAs when they are bound by g5p, indicating that each type of perturbed nearest neighbor has a similar average structure within the binding site of the protein.  相似文献

As an essential repressor, the homeobox gene Hesx1/HESX1 is required within the anterior neural plate for normal forebrain development. Mutations within the Hesx1 gene have been associated with GH deficiency or combined pituitary hormone deficiency. We detected the polymorphism of Hesx1 gene by PCR-SSCP and DNA sequencing methods in 702 individuals from four Chinese cattle breeds. A novel single nucleotide polymorphism (SNP) (IVS1 + 382T > C) was detected. The frequencies of genotype TC in four breeds were 0.000–0.222. Polymorphism of the Hesx1 gene was shown to be associated with growth in the Nanyang breed. Individuals with genotype TC was significantly lower average daily gain than TT at 18 months (P < 0.05).  相似文献

The toxic interaction of melamine with herring sperm DNA (hs‐DNA) was investigated by using fluorescence and UV–vis absorption spectra techniques. The experimental results showed that the toxic interaction between melamine and hs‐DNA occurred. Fluorescence quenching experiments indicated the existence of electrostatic binding between melamine and hs‐DNA. The binding constants KA and the binding site numbers were calculated by means of the Stern–Volmer equation and were 9.8 × 104 L mol?1 and 1.3, respectively. Both the results of fluorescence spectra and UV–vis absorption spectra verified that there are electrostatic binding between melamine and hs‐DNA. The possibility in the presence of a classical intercalation binding mode could be ruled out by using DNA unwinding experiments. © 2010 Wiley Periodicals, Inc. Release from enemies can lead to rapid evolution in invasive plants, including reduced metabolic investment in defence. Conversely, reassociation with enemies leads to renewed evolution of defence, but the potential costs of this evolution are poorly documented. We report increased resistance of the invader Ambrosia artemisiifolia after reassociation with a coevolved specialist herbivore, and that this increase corresponds with reduced abiotic stress tolerance. Herbivore resistance was higher, but drought tolerance was lower in plants from populations with a longer reassociation history, and this corresponded with changes in phenylpropanoids involved in insect resistance and abiotic stress tolerance. These changes were corroborated by shifts in the expression of underlying biosynthetic genes and plant anti-oxidants. Together, our findings suggest rapid evolution of plant traits after reassociation with coevolved enemies, resulting in genetically based shifts in investment between abiotic and biotic stress responses, providing insights into co-evolution, plant invasion and biological control.  相似文献

The role of sucrose as a signal molecule in plants was in debate for a long time, until recently, it gradually becomes more prominently accepted. Sucrose plays roles in a vast array of developmental processes in plants, however, its function in fruit ripening has not been well elucidated. In this study, the influence of exogenous sucrose treatment (500 mM) on postharvest tomato fruit ripening was investigated. It was found that, in comparison with mannitol treatment (500 mM, set as control), sucrose accelerated the ripening process with higher levels of respiration rate and ethylene production during the storage. Sucrose treatment up-regulated its biosynthetic genes, whilst stimulated expressions of genes encoding degradation related enzymes in the fruits. However, higher sucrose content was observed in sucrose-treated fruits only in the first few days. In addition, sucrose application had minor effect on the contents of its degrading products, glucose and fructose. Moreover, exogenous sucrose treatment up-regulated expressions of ethylene biosynthetic genes, and promoted ethylene signal transduction via influencing critical genes of the signaling pathway in different patterns. These results indicate that sucrose stimulates tomato fruit ripening may through mediating its own metabolism, which facilitates nutrients fluxes and metabolic signaling molecules activation, and also by enhancing ethylene biosynthesis and signal transduction.  相似文献

对中国13条主要河流淡水鱼类分布进行的系统聚类分析结果表明,分异最小的是闽江与九龙江。瓯江与钱塘江、淮河与黄河、东北的水系分别较早聚为一类。相异系数在0.60~0.69时分为三类: 东北的水系、黄河到珠江间的水系和云南的元江。淡水鱼种类数与流域特征的相关和回归分析揭示了:鱼种数分别与河流流域面积、年平均流量呈显著或极显著的正相关,单位干流长和单位流域面积的鱼种类数分别与河流径流深、比降呈显著或极显著的正相关, 而与河流平均纬度呈显著负相关。主成分分析得到的第一和第二主成分分别综合反映了河流鱼种类数和相对种类数与河流特征值的关系。  相似文献
